Excretion of urinary volatile metabolites in response to alloxan induced diabetes of short duration in rats.
Alterations in urine volatile metabolites due to the induction of alloxan diabetes in the rat were examined by capillary gas chromatography and gas chromatography--mass spectrometry for the five days immediately following the onset of chronic hyperglycemia. Elevations of a number of metabolites were observed including several short chain ketones, acetophenone, 2-acetylfuran and indole. The value of urine volatile metabolic profiles as characteristic indicators of the diabetic condition is demonstrated through profiles obtained from a diabetic animal which spontaneously reverted to normal.

Studies of hyperlipidemia in drug-induced diabetic rats by high-performance liquid chromatography.
A hyperlipidemic condition is often associated with diabetes. The possibility that specific serum lipids (i.e., individual triglycerides or cholesterol esters) may be altered in the diabetic state was investigated. Serum lipids from both controls and streptozotocin- and alloxan-treated rats were separated into approximately twenty chromtographic fractions by reversed-phase high-performance liquid chromatography; a number of individual triglycerides and cholesterol esters were identified. The methodology described allowed subtle changes in individual lipid components to be detected. Only minor variations in the cholesterol and cholesterol ester fractions were observed between the control and diabetic samples. While not uniform throughout, elevations in the triglyceride fractions occurred in the diabetics. Also, differences in triglyceride content were found to exist between the groups of streptozotocin- and alloxan-treated animals.
Small-volume electrochemical detector for microcolumn liquid chromatography.
A submicroliter electrochemical detector for liquid chromatography has been designed, using pressure-annealed pyrolytic graphite technology. The analytical performance of this detector was studied in connection with a reversed-phase packed microcapillary column at very low flow-rates. Although the miniaturized version of the electrochemical detector is less sensitive, a direct analysis of a number of urinary metabolites in 0.1--1.0 microliters samples is feasible.

Application of pattern recognition and feature extraction techniques to volatile constituent metabolic profiles obtained by capillary gas chromatography.
The applicability of threshold logic units, a form of nonparametric pattern recognition, to the processing of metabolic profile data obtained by high-efficiency glass capillary column gas chromatography has been investigated. The test data included profiles of the volatile constituents of urine from normal individuals and from individuals with diabetes mellitus. A feature extraction algorithm allowed for dimensionality reduction and indicated the constituents most important in the normal versus pathological distinction. With an optimum number of dimensions, a normal versus pathological prediction rate of 93.75% was achieved. Gas chromatography-mass spectrometry was utilized to identify important profile constituents.

Fatty acid synthesis in vitro in the liver and adipose tissue of rats of various ages.
In white male Wistar rats aged 2, 4, 6, 9, 12, 18 and 32 months, receiving a standard laboratory diet (25 cal% proteins, 22 cal% fat and 53 cal% glycides) we assessed the total amount of fatty acids in the carcass and in vitro lipogenesis in liver and adipose tissue by the means of glucose-14C(U). The amount of body fat increased with age; the highest rate of increase in the percentage of body fat occurs 2 to 4 months after birth. Incorporation of 14C of glucose into total liver lipids attains maximum values in the group of rats age 2 to 4 months. By the age of 6 months lipogenesis decreased to a level observed in all the remaining age groups. In adipose tissue, the incorporation of 14C into total lipids decreased between the age of 2 to 4 months, a further decrease was observed around the age of 6 months. No appreciable changes were noted in any of the older age groups.

Analysis of the acidic fraction of marijuana smoke condensate by capillary gas chromatography-mass spectrometry.
A method for the analysis of organic acids and phenols isolated from marijuana smoke condensate has been developed. Comparative analyses of standard tobacoo, Mexican, and Turkish marijuana smoke condensates carried out by means of capillary gas chromatography indicated both qualitative and quantitative changes in the constituents of chromatographic profiles. Samples were converted to volatile derivatives by methylation and trimethylsilylation; whereupon, 49 aliphatic acids, aromatic acids, and phenolic compounds were identified by means of capillary gas chromatography-mass spectrometry.
Automated high-resolution gas chromatographic system for recording and evaluation of metabolic profiles.
A fully automated high-resolution gas chromatographic system is described that incorporates repetitive sampling of volatile biological samples onto a glass capillary column, and a simultaneous flame ionization and nitrogen-sensitive detection prior to data acquisition and computer handling. Reliable headspace sampling procedure and reproducible column characteristics that are essential to automation have been studied. The application of this system to the reproducible chromatographic recording of human urinary volatile constituents is shown.
