Immune monitoring after renal transplantation. II. Post-transplant disturbances in the CD3 complex expression may be caused by immunosuppression.

The in vitro effect of major immunosuppressive agents on the CD3-TCR complex expression on T cells was studied. Cyclosporine and azathioprine caused a marked diminution of alpha/beta, but not gamma/delta and CD3 chains detectability. Prednisolone effect was less manifested. Our data suggest that the reported decrease in alpha/beta chains expression on T cells from renal allograft recipients may be caused by post-transplant immunosuppression.

Rejection-associated upregulation of the alpha/beta-T cell receptor expression on T cells of renal allograft recipients.

Peripheral T lymphocytes of renal allograft recipients were phenotyped for their expression of the CD3-T cell receptor (TCR) complex. The majority of T cells from patients with stable graft function had normal CD3 but diminished TCR alpha/beta heterodimer expression, while TCR gamma/delta + T cells were increased in some cases. Acute rejection was associated with an increase in TCR alpha/beta + T cells to normal levels.

Immunosuppressive drugs inhibit class II antigen (HLA-D) synthesis by B and T lymphocytes.

The influence of immunosuppressive drugs on class II (HLA-D) antigen synthesis and expression by monocytes and B and T cells was studied. The constitutive HLA-D expression on monocytes and malignant B cells was resistant, while the acquisition of those markers by stimulated T cells very sensitive to immunosuppression. Active derivative of cyclophosphamide (4HCy) and cyclosporine proved to be most efficacious inhibitors of class II antigens.

Large granular lymphocyte lymphocytosis with neutropenia: a case report.

A case of large granular lymphocyte lymphocytosis with neutropenia was studied as evidenced by peripheral lymphocytosis of cells having typical morphology and profound neutropenia. Surface markers analysis revealed that almost all cells were CD8+ and their subpopulation DR+. The cells had strong spontaneous and inducible suppressor functions in vitro. Moreover, their ADCC activity was strong but NK activity very weak.

An optimal assay of human B cell proliferation in vitro.

The purpose of this work was to optimize an in vitro assay for measuring B cell proliferation in response to S. aureus Cowan I (SAC). It has been demonstrated that the best proliferative activity is obtained when 10(5) cells are cultured in flat-bottomed microtiter plates in the presence of 1:10,000 SAC dilution. Standard RPMI 1640 culture medium should be enriched with mercaptoethanol and 10% FCS or 0.5% human albumin. Human serum, found to be an optimal supplement for T lymphocyte responses, blocks B cell proliferation.

Morphometric ultrastructural evaluation of human peripheral blood T lymphocyte subpopulations isolated on the basis of their affinity for sheep red blood cells.

The present investigation were undertaken to compare on the basis of ultrastructural morphometric analysis human T lymphocyte subfractions forming rosettes with sheep red blood cells (SRBC). The following T lymphocyte subfractions were obtained: ARFC (active rosettes), T1RFC (rosettes after 1 h at 4 degrees C) and T2RFC (rosettes after 2 h at 4 degrees C). Analysis of the mean cell and nuclei volumes of lymphocyte subfractions led to the separation of two cell groups differing in volume in each donor: a cell group consisting of large cells (T1RFC) and group of small cells (ARFC and T2RFC).

Graft-induced polyclonal activation of B cells. II. Humoral response of renal allograft recipients (RAR).

Ig production by peripheral blood lymphocytes of renal allograft recipients was studied. It was demonstrated that lymphocytes of patients with acute graft rejection produce elevated numbers of PFC in culture in response to PWM (a T-cell dependent stimulus) and Salmonella paratyphi B (a T-cell independent stimulus) as well as spontaneously. PFC responses were significantly higher in patients with acute rejection than in a group of patients with stable graft function. The B cells of the recipients with stable graft function showed a normal humoral response in vitro. During chronic rejection no significant changes were found in the production of antibodies by B cells.

Immunomodulatory action of ampicillin.

Ampicillin was found to have an immunoenhancing effect on the expression of FcM human lymphocyte receptors, on the angiogenic potential of human lymphocytes and on the production of anti-SRBC antibody after in vivo administration to low-responder strains of mice. The effect of ampicillin on the graft-versus-host disease in irradiated F1 recipients of spleen cells from presensitized donors was found to be stimulatory when the drug was given to the recipients and suppressive when the drug was administered to the donors which had been preimmunized with a lower dose of antigen.

The effect of azathioprine on terminal differentiation of human B lymphocytes.

The effect of azathioprine on the terminal differentiation of human B cells triggered in vitro by pokeweed mitogen and S. aureus Cowan I bacteria was studied. AZ caused an enhancement of Ig synthesis in its lower concentrations, a phenomenon dependent on the inactivation of suppressor T cells. Higher drug concentrations impaired T-helper function as well as B cell differentiation of peripheral blood, lymph node, and spleen lymphocytes.
