Genetic variation of the amplified VNTR polymorphism COL2A1 in Chinese and German populations.

The amplifiable VNTR polymorphic system COL2A1 has been investigated in Chinese and German population samples. In order to accurately identify COL2A1 alleles, a number of human allele ladders were prepared. A total of 14 different alleles in 23 genotypes was observed in the Chinese population, while in the German population there were 11 alleles in 24 genotypes. Four new alleles were observed. All of them were found in the Chinese and one was also observed in the German population. There was no statistical difference in the allele distribution between two German populations from Münster and Köln, while the allele frequency distribution in the Chinese population differed significantly from the German.

[Possibilities of DNA sex determination in hair roots].

Species- and sex-determination on hair roots were simultaneously performed using extracted DNA and a human Y chromosomespecific probe. (pH Y 2.1) After Southern hybridization, Hae III-digested DNA fragments were detected by non-radioactive digoxigenin detection system. DNA was extracted from one to five freshly plucked hair roots. The specific 2.12 kb fragment was successfully detected in male DNA samples from a single hair root. A positive identification of female DNA was more difficult. The hair root DNA was revealed to be stable at room temperature for at least 2 weeks (examination time) and produced the same specific band pattern as the DNA of fresh hair roots. In the blind tests with DNA samples from randomly plucked one to four hair roots, the rate of successful sex-determination was 95.8% on male samples (23 out of 24 samples) and 25% on female samples (4 out of 16 samples).

[Corpse dismemberment with sequential identification and classification of body parts].

In the reported case of criminal dismembering of a corpse the preservation in vinyl-bags allowed seven years after murder, to find out if the body parts fitted to those which were detected years ago. Furthermore, it was possible to find out the presumed cause of death, the morphologic characteristics of the injuries caused by stabbing with a single edged tool and successful DNA typing by PCR amplification for identification of the body parts and for interpretation of the biological stains from the crime scene.
