Activity and isoenzymes of acid phosphatase in human B-cell lymphomas of low-grade malignancy: a novel aid in the classification of malignant lymphoma.

Activity and isoelectric focusing (IEF) pattern of lysosomal acid phosphatase (E.C.3.1.3.2.) were investigated in 55 cases of low-grade malignant B-cell lymphoma, classified as chronic B-lymphocytic leukemia (B-CLL), centroblastic/centrocytic follicular lymphoma (CB/CC), lymphoplasmacytic/lymphoplasmacytoid lymphoma (Immunocytoma, IC), and plasmacytoma (PC), applying the criteria of the Kiel classification. The results show (1) that the four lymphoma types present a characteristic range of enzyme activity in an increasing order: B-CLL, BC/CC, IC, and PC. B lymphocytes, germinal center cells, and plasmacytes are the main constituents of these lymphomas. This sequence might reflect one possible mode of B-cell transformation into plasmacytes traversing an amplification stage in germinal centers under normal conditions. (2) All cases showed the basic IEF pattern of normal B lymphocytes with 12 bands localized in three regions between pH 6.1 and 3.9. This finding supports the B-cell origin and the close phenotypical relationship among the investigated lymphomas. (3) The IEF patterns of B-CLL and CB/CC did not differ from that of normal B lymphocytes, whereas two additional isoenzymes were encountered in cases of IC and seven in PC; this suggests that the higher enzyme activity of IC and PC is at least partly due to the appearance of "new" isoenzymes. The results support the validity of the underlying classification and indicate the individually, B-cell origin, and close relationship among the four lymphoma entities investigated.

Lysosomal acid phosphatase: activity and isoenzymes in separated normal human blood cells.

The present study was devised to investigate the activity and isoenzymes of lysosomal acid phosphatase in individual normal human blood cells, including the T- and B-population of lymphocytes, with the aim to contribute to the classification of haematopoietic neoplasias on the basis of cell specific isoenzyme patterns. Platelets, erythrocytes, granulocytes, monocytes and T-lymphocytes were isolated from blood by gradient centrifugation or immune adsorption. B-lymphocytes were obtained from human tonsils. After purity control and isolation of lysosomes the concentration of acid phosphatase was assayed using the conventional spectrophotometric method. Isoenzymes were separated by isoelectric focusing on polyacrylamide thin layer slabs. Monocytes revealed the highest activity with 14 mU/10(7) cells, about three times more than granulocytes. T-lymphocytes showed an activity of 2.85 mU/10(7) cells and B-lymphocytes of 1.83 mU/10(7) CELLS. The lowest activity was found in platelets with 0.08 mU/10(7) cells. Granulocytes showed 12 isoenzyme bands, whilst the number for monocyte, B-lymphocytes, T-lymphocytes and platelets were respectively 11, 12, 1 and 4 isoenzyme bands. Thus it became evident that the different blood cell populations can be distinguished on the basis of their acid phosphatase isoenzyme pattern.

An unusual labeling index pattern of neutrophilic granulocytes after a 3H-thymidine flash labeling injection.

After flash labeling of neutrophilic granulocyte precursors with 3H-Thymidine in 3 hematologically normal subjects the postinjection labeling indices (LI) and the labeling intensities of venous blood neutrophils were determined. The LI pattern during 18 days after injection showed a reproducible triphasic course of the LI curves: (a) a low "plateau", (b) a steep rise, (c) an undulating decrease until the end of the experiments. On the basis of these findings a simultaneous "at random" and "first in--first out" release of neutrophils from the postmitotic bone marrow compartment to the blood was suggested. Moreover, the intravascular half-life time of the neutrophils was determined as 9.3 h, the corresponding mean circulation time was 14 h. From the undulating course of the LI curves and the labeling intensity curves the duration of the mean neutrophil precursor cell cycle time in the mitotic bone marrow compartment was estimated to range between 19 and 24 h.

Serum profile of non-esterified fatty acids (NEFA) in patients with hyperlipoproteinemia under xantinol--nicotinate (XN) medication.

The effect of 50 mg/kg body weight Xantinol-nicotinate (XN) on serum lipids and on non-esterified fatty acids (NEFA) was tested in 94 out-patients with hyperlipoproteinemia Types IIa, IIb, IV and V. The lipids were measured before treatment, during a 3-week period of drug administration and 10 days after rejection. In each group of hyperlipoproteinemic patients the lipid lowering effect of XN was accompanied by a constant fall in serum NEFA levels. The time dependent course of NEFA showed that the higher the initial level, the longer the time span required to achieve the lowest values. In the context of the partially conflicting data on the hypolipidemic action of nicotinic acid and its derivatives, it is suggested that the constant decrease in NEFA induced by XN treatment might contribute significantly to the lipid lowering effect of this drug.

[Selective staining of T-lymphoblastic leukemia cells by glycyl-prolyl-4-methoxy-beta-naphthylamiden-peptidase (DAP IV) (author's transl)].

In search for new methods to maintain a selective and specific visualization of blast cells in cases of acute lymphoblastic leukemia the glycyl-prolyl-4-methoxy-beta-nephthylamide-peptidase reaction (DAP IV) was performed in cytochemical tests. Blood and marrow slides from 15 cases of acute lymphoblastic leukemia of childhood were stained. The results were compared with findings in Pappenheim and acid phosphatase stained slides. In addition slides from concentrated blood lymphocytes of 50 normal subjects served as control. The following results were achieved. 1. In normal blood only about 50% of T-lymphocytes reveal a positive DAP IV reaction. 2. Lymphocytes positive to DAP IV show a typical dot-like paranuclear pattern of activity for acid phosphatase. 3. Only 5 cases out of 15 acid phosphatase positive acute lymphoblastic leukemias presented a positive reaction for the DAP IV. DAP IV reaction is specially useful in visualization and quantification of blast cells in hypercellular marrow slides. In positive cases it further allows a reliable evaluation of the mark infiltration and duration of remission.

Isoelectric focusing pattern of acid phosphatase and acid esterase in human blood cells, including thymocytes, T lymphocytes, and B lymphocytes.

Lysosomal acid phosphatase and acid esterase from highly purified, viable human thymocytes, T lymphocytes, B lymphocytes, granulocytes, monocytes, platelets, and erythrocytes were subjected to direct measurement of activity and to isoelectric focusing in polyacrylamide thin-layer slabs. The typical patterns for each cell line were found to be distinctly different from the results recently presented in this journal. The study described here opens a new possibility for the biochemical identification of cells and their functional derivatives and neoplastic variants.

Spontaneous rupture of the Achilles tendon - a sign of hyperlipoproteinaemia (HLP) type II.

In biopsies from spontaneously ruptured Achilles tendons a serum lipid and lipoprotein analysis was performed in fifty nine patients. In nine males considerable histological signs of lipid deposition were detectable. The tendon fibers were found to be partly desintegrated. Only in these cases LDL-cholesterol values were found to exceed the normal range indicative of type II hyperlipoproteinaemia (HLP). None of the fifty remaining patients presented biochemical or clinical signs of disorders of lipid metabolism. The observations demonstrate that a spontaneous rupture of Achilles tendom may indicate a primary type II HLP.

Estimation of effective eosinopoiesis and bone marrow eosinophil reserve capacity in normal man.

The eosinophil reserve capacity of the post-mitotic granulocyte compartment in the bone marrow and the effective eosinopoiesis in three haematologically normal men have been quantified by means of kinetic parameters of [3H]thymidine flash-labelled peripheral blood eosinophils. From (a) the time of the appearance in the blood of labelled eosinophils after the tracer injection, (b) the inflow characteristics of the labelled eosinophils in the blood and (c) the magnitude of the eosinophil granulocyte pool in the venous blood, the effective eosinopoiesis (i.e. the eosinophil turnover) was calculated to range between 0.014 and 0.031 x 10(9) cells/kg body weight per day (mean 0.22 x 10(9) cell/kg per day). The post-mitotic eosinophil reserve capacity of the bone marrow ranged from 0.09 to 0.20 x 10(9) cells/kg body weight (mean 0.14 x 10(9) cells/kg). The large reserve pool and the high turnover rate may contribute to sudden rises of the peripheral blood oesinophil counts in some cases of eosinophilia.

Efficacy of hypolipidemic treatment in inhibition of experimental atherosclerosis: the effect of nicotinic acid and related compounds.

The hypolipidemic and antiatherogenic effects of different nicotinic acid derivatives were studied. Five rabbit groups maintained on an atherogenic diet were given simultaneously various nicotinic acid derivatives (50 mg/kg body weight/day): nicotinic acid, Xantinol-nicotinate, beta-pyridylcarbinol or Pirozadil (bis-3,4,5-trimethoxybenzoate, 2,6-pyridindiyldimethylene). All 4 compounds showed a clear hypocholesterolemic and antiatherogenic effect, as measured by serum cholesterol, and by planimetric evaluation of the aortic lesions in terms of percent surface area affected in the aortas and coronary lumen. The simultaneously observed elevation of the HDL/LDL cholesterol ratio of the aortic tissue possibly indicates an antiatherogenic effect of these changes.
