[Therapy of hyperlipoproteinemia type II with Xantinol-nicotinate (author's transl)].

In 61 out-patients with hyperlipoproteinemia type II the action of 50 mg Xantinol-nicotinate per kg body weight on lipids of plasma and lipoproteins was investigated in a three week trial. The results of six patients had been discharged: two patients failed to attend the follow-up examinations, in four patients a lipid lowering effect could not be demonstrated. 40 Type IIa patients and 15 Type IIb patients were examined. In this two groups cholesterol, phospholipids and triglycerides of plasma were significantly lowered. In Type IIa hyperlipoproteinemia mainly the LDL lipids and in Type IIb additionally the VLDL lipids decreased. In Type IIb patients the decrease of lipids were more rapid than in Type IIa patients. This effect is interpreted by the short half life of VLDL. Withdrawel of the drug resulted in an increase of the lipids to the mean values of the untreated patients within ten days.

[Crystalline corneal dystrophy (Schnyder) in the presence of familial type IIa hyperlipoproteinaemia (author's transl)].

A family with dominant autosomal hyperlipoproteinaemia typ IIa is reported. One sibling, a 19-year-old man, showed in addition to the hyperlipoproteinaemia typical corneal signs of crystalline corneal dystrophy of Schnyder. From 1964 to 1976 these corneal deposits increased. The problems of hyperlipoproteinaemia combined with corneal crystalline dystrophy are discussed.

[Diagnosis of primary hyperlipoproteinemia in umbilical cord blood (author's transl)].

The aim of the present investigation was to assay the frequency of primary dyslipoproteinemia in a random sample of one hundred newborns and to describe the minimal methodical requirements for sound diagnosis. After comparison of different methods total lipids were determined by gravimetry, cholesterol and triglycerides by enzymatic methods, nonesterified fatty acids by direct colorimetry; phospholipids were estimated indirectly. All measurements were applied to umbilical cord sera and to lipoprotein fractions separated by selective precipitation. The diagnosis of hyperlipoproteinemia type IV, which is the most frequent one in adults, is highly afflicted with pitfalls in the postnatal period. A primary hyper-alpha-liproteinemia occured in one case and type II-hyperlipoproteinemia in two cases, one of the parents being involved in each case. For mass screening triglycerides should be assayed in serum and cholesterol in precipitated and resolubilized LDL-fraction, for which the minimal requirements are described.

[Xantinol-nicotinate in primary type-V hyperlipoproteinaemia (author's transl)].

The effect of xantinol-nicotinate (50 mg/kg body-weight) on serum lipids and lipoproteins was tested in 16 out-patients with primary type V hyperlipoproteinaemia. The lipids and lipoproteins were measured before treatment, during a three-week period of drug administration and ten days after it had been stopped. There were no side effects such as flushing or gastritis, and no notable reduction of weight. Each serum-lipid fraction (triglycerides, nonesterified fatty acids, phospholipids, ester-cholesterol and free cholesterol) decreased significantly, regaining the initial values ten days after the drug had been stopped. While chylomicrons and very low-density lipoproteins (VLDL) decreased, there was a significant increase in low-density lipoproteins (LDL). High-density lipoproteins were not significantly changed. The decrease in chylomicrons and the significant decrease in VLDL with xantinol-nicotinate was opposite to that seen with dietary treatment. In none of the patients did LDL increase to abnormally high levels.

The peripheral kinetics of human radiolabelled eosinophils.

Peripheral kinetics of eosinophils were studied in three subjects with normal hematological conditions after a puls labelling with 0.1 mu Ci 3HTdR per g bodyweight. The computer recorded labelling index curves (LI) presented an undulating course in all three subjects. Eosinophils proved to disappear randomly. The emergence time of 13 h was followed by the first LI peak of 17%. 80 h p.i. a massive immigration of highly labelled cells with a LI value of 58% was encountered. 50% of eosinophils remained labelled up to 11th day p.i. Cells with different labelling intensities arrayed to grain groups presented a clear synchronism of their LI-curves which become less clear with time. The mean t/2-value for all labelling intensities amounted to 8 h. Cells with low labelling intensities had a longer and the highly labelled eosinophils a much shorter t/2-values. These effects suggest the existence of two proliferating eosinophil precursor pools: a rapid and a slow one. The early influx of low labelled as well as the late emergence of great number of highly labelled eosinophils are in good accord with this assumption. The time span for S-phase indirectly calculated revealed 13 h. The myelocyte generation time was estimated with 34 h. Both values were found to well agree with the corresponding data for other granulocytes as reported on, in the literature.

[Corneal arcus and hyperlipoproteinemia (author's transl)].

In 60 patients with corneal arcus the incidence of hyperlipoproteinemia was examined by serum lipid and lipoprotein analysis. An elevation of the serum lipid, or lipoprotein contents was found in every patient. 52 patients showed a primary type II hyperlipoproteinemia, three had a type IV, and one a type V pattern. In three cases hyperlipoproteinemia was found to be secondary to a severe cholestasis. The results show that the occurrence of corneal arcus in nearly every subject under 50 years of age and in most subjects over 50 years of age is highly suggestive of the presence of type II hyperlipoproteinemia. Until atherosclerotic complications appear, the course of the type II hyperlipoproteinemia is lingering and shows poor symptoms. Therefore, corneal arcus is of special importance for the early detection of this type of hyperlipoproteinemia.

Leukemic anaplasias reflecting physiologic cytogenesis of myeloid system.

Naphthol AS-D chloroacetatesterase activity in peripheral blood granulocytes is confined to neutrophils which are all positive and to a minor part of monocytes. Its occurance in eosinophils and basophils indicate a myeloproliferative disease. This chemical property can reliably be applied to separate neoplastic from reactive forms of quantitative and qualitative leukocyte alterations. The developmental line of this specific myeloid cellular attribut has been presented to elucidate its diagnostic significance and its validity as proof for existence of a common promyelocyte from which neutrophils, monocytes, basophils and eosinophils originate.

[Lipid constellation in low density lipoproteins of primary type II hyperlipoproteinemia (author's transl)].

The lipid composition of purified low density lipoproteins (LDL) was investigated in normal subjects and in type II a and b hyperlipoproteinemia. A significant increase of ester cholesterol in LDL of type II a patients and a decrease of phospholipids in LDL of type II b patients as compared with normal controls, were encountered. The results are compared with other reports from the literature and discussed with regard to the disturbance of LDL metabolism in type II hyperlipoproteinemia.

[Quantitation of basophil cell-line in human bone marrow (author's transl)].

Appropriately fixed basophil cell-line can selectively be visualized by anionic dyes as toluidine blue. Typical activities for Leder's esterase distinguish individual maturity stages. Both methods were successively applied to bone marrow smears from 22 healthy subjects in order to assess the basophil population among the total marrow leukocytes (lymphocytes, monocytes and granulopoietic cell-lines) and the portion of each basophil maturity stages. Total basophils represent 0.45+/-0.13% of marrow leukocytes. Basophil precursors capable of division (promyelocytes and myelocytes) comprise 46.7%, mitoses 3.7+/-1.76% of the total marrow basophil population. The frequency distribution of these subpopulations approximated a normal distribution pattern. No clear central tendencies could be encountered in the dispersion of other stages. The low peripheral basophil counts, the high incidence of divisible precursors and the considerable mitotic activity suggest a short half-life time for blood basophils.

[5-Aminoacridine hydrochloride for fixation of human blood basophils (author's transl)].

5-aminoacridine hydrochloride (0.4 percent in 50 percent aqueous ethanol) proved to be a useful fixative for the preservation of human blood basophils pretreated with methanol-formaldehyde (9:1 V/V). Leaching of the water-soluble basophil granules or other artificial cell alterations common to fixatives in current use were not observed. The organic compound used did not induce any chemographic artifacts in nuclear tracers, in contrast to most of the other fixatives hitherto applied. 5-aminoacridine hydrochloride is therefore particularly suitable for autoradiography of the human blood basophils.

An autoradiographic study of human blood basophils.

The aim of this paper is the selective visualization of human blood basophils in autoradiographs. [(3)H]Thymidine-labelled basophils in buffy coat smears were fixed in methanol-formaldehyde follwed by 5-aminoacridine hydrochloride and stained with basic Aldehyde Fuchsin prior to autoradiographic processing. The technique described represents a simple method for quantitative autoradiographic studies on basophil kinetics and the interaction of these cells with IgE-mediated atopias.

[Autoradiographic demonstration of human basophils].

A new technique is described which enables a selective tinctorial visualization of human blood basophils in autoradiographic preparations. Consecutive administration of 5-aminoacridine hydrochloride and aldehyde fuchsin rendered a sufficient preservation of basophils preventing leaching of their water soluble granules. Extensive control tests provided sufficient evidence to exclude chemographic artifacts produced by other histochemical precedures. The technique eliminates the obstacles to study the kinetics of human blood basophils and their realtionshp to atopic hypersensitivity reactions applying tritiated thymidine and radioiodine labeled IgE or corresponding antisera.
