Correlation between urinary oestrogen levels determined by haemagglutination inhibition reaction and serum oestradiol levels determined by radioimmunoassay.

Measurement of urinary or blood oestrogen is essential for a precise monitoring of follicular development in gonadotrophin therapy. Blood oestradiol levels are considered most accurately to reflect follicular maturation. Although radioimmunoassays (RIA) for oestradiol yielding rapid results are now available, they require qualified technicians and equipment for RIA. Recently, a simple, rapid method for determining urinary total oestrogen has been developed by Mochida Pharmaceutical Co. Ltd., Japan. To determine whether this kit may be applicable to monitoring gonadotrophin therapy, urinary total oestrogen concentrations measured by the kit were compared with serum oestradiol levels determined by RIA in patients who were treated with gonadotrophin. Urinary total oestrogen concentrations in samples obtained either at 07.00 h (first morning urine specimen) or at 09.00 h significantly correlated with serum oestradiol levels. Thus, urinary total oestrogen concentrations determined by the kit appear to reflect follicular development accurately, and are considered applicable to monitoring gonadotrophin therapy. Correction of the urinary total oestrogen concentrations by the creatinine level significantly improved the correlation between urinary total oestrogen values and serum oestradiol levels. Therefore, the primary total oestrogen values should be corrected by the creatinine level.

[Fundamental studies on aspoxicillin in the field of gynecology. Penetration of the drug into the genital organs and the dead space fluid. Antibacterial activity of the drug against various strains of clinical isolates].

A new semisynthetic penicillin aspoxicillin (ASPC, TA-058) has been evaluated in regard to distribution into the genital organs and the dead space fluid and also in regard to antibiotic activity against various strains clinically isolated in the field of gynecology. The pharmacokinetic studies were made by using two-compartment model on the data of concentration in ovary, oviduct, endometrium, myometrium, cervix uteri and portio vaginalis of the genital organs and in dead space fluid. The following results were obtained. After one shot intravenous administration of 2.0 g ASPC, no marked difference was observed in concentration pattern between peripheral vein and uterine artery. Half-lives were 1.8 hours and 3.5 hours, respectively. The concentrations in various tissues of genital organs reached to the maximum at 36.2-73.6 micrograms/g during 0.07-0.44 hour with half-life varied from 0.53 to 1.3 hours. Area under the curves (AUC) of various tissues were reached to 30-65% of that uterine artery. After 2.0 g administration of ASPC drip intravenously (d.i.v.; for 30 or 60 minutes), there are no remarkable difference on simulation curve of concentration between peripheral vein and uterine artery. The peak level of various tissues attained to 22.5-40.3 micrograms/g during 0.53-0.56 hours and AUC of their concentration achieved about 20-40% of that of uterine artery after intravenous drip infusion for 30 minutes. After intravenous drip infusion for 60 minutes, tissue level reached to the maximum during 1.01-1.82 hours with half-life of 0.65-1.4 hours and AUC of tissue concentration achieved about 20-57% of that of uterine artery. The concentration of the drug in dead space fluid was reached to the maximum at 3.1 hours after the administration and its half-life was 5.3 hours. The in vitro activities of ASPC against clinical isolates were examined. The values of MIC70 and MIC80 were ranged 0.35-5.1 micrograms/ml against S. aureus, S. epidermidis, E. faecalis and P. mirabilis, respectively. Against E. coli, the value of MIC70 was attained 2.9 micrograms/ml, however, the value of MIC80 was more than 100 micrograms/ml due to the two-peaked activity of this drug against the bacteria. AUC and duration time of various tissue concentration were scrutinized regarding to the value of MIC80 of ASPC against S. aureus, S. epidermidis, E. faecalis and P. mirabilis.(ABSTRACT TRUNCATED AT 400 WORDS)

[5-FU concentration in tissues of gastric cancer patients with preoperative administration of UFT--especially in comparison with FT-207].

FT-207 or UFT was administered preoperatively to 74 patients with gastric cancer. The 5-FU and FT-207 concentrations in the serum and various tissues were investigated. The 5-FU concentration in various tissues was higher than in the serum after the administration of FT-207 or UFT. Especially it showed that 5-FU concentration in tumor was highest in other normal tissues. The 5-FU concentration in the tumor for the UFT group was higher than for the FT-207 group. There was a significant difference between the UFT and FT-207 groups (P less than 0.05). Levamisole had no influence on the concentration of 5-FU.

Anterior thalamic afferents from the mamillary body and the limbic cortex in the rat.

Anterior thalamic afferents from the mamillary body and the limbic cortex were studied by using single and double retrograde transport methods in the rat. The medial mamillary nucleus was divided on the basis of the cytoarchitecture into four subnuclei: the pars medialis centralis, pars medialis dorsalis, pars lateralis, and pars basalis. Extensive connections were seen between each of these subdivisions of the mamillary body and the anterior thalamic nuclei, topographically organized so that the anteromedial thalamic nucleus receives projections exclusively from the pars medialis centralis, while the anteroventral thalamic nucleus receives projections from the pars medialis dorsalis and pars lateralis. Nuclei in the dorsal half of these two mamillary subdivisions project predominantly to the medial half of the anteroventral thalamic nucleus, and those in the ventral half to the lateral half of the nucleus. The pars basalis was found to have numerous projections to the magnocellular part of the anteroventral nucleus. All limbic cortical areas send projections bilaterally to all regions of the anteromedial nucleus as well as to the parvicellular parts of the anteroventral thalamic nucleus, while the anterodorsal nucleus receives ipsilateral projections originating exclusively from the preagranular, anterior limbic, and cingular regions. The magnocellular part of the anteroventral nucleus, however, receives only ipsilateral projections from all of the limbic cortex. Some neurons in the infralimbic region also project bilaterally to all of the anterior thalamic nuclei except the anterodorsal nucleus. All of these cortical projections to the anterior thalamus originate in layers V and VI of the limbic cortex.

Effect of the strandedness of cofactor DNA on the activities of DNA-dependent ATPases B and C3 from FM3A cells.

The requirements of cofactor DNA for DNA-dependent ATPases B and C3 were analyzed in detail. ATPase B and C3 required the presence of a polynucleotide for their activities. Among the DNAs tested, ATPase B showed a preference for poly(dT) as its cofactor. The other deoxyhomopolymers, except poly(dG) and heat-denatured DNA also were effective. The alternating polydeoxyribonucleotide, poly[d(A-T)] had an efficiency 23% that of heat-denatured DNA. Unlike ATPase B, ATPase C3 showed almost no activity with deoxyhomopolymers. The most effective cofactor for ATPase C3 so far tested is poly[d(A-T)]. Relatively high activity was obtained with heat-denatured DNA. The high activity of ATPase B with poly(dT) was reduced by the addition of poly(dA). The addition of noncomplementary homopolymers did not affect enzyme activity. ATPase C3 activity in the presence of 10 microM poly(dT) increased gradually with concentrations of poly(dA) up to 20 microM, after which it decreased. Almost no increase in activity was observed when noncomplementary homopolymers were added. The relatively high activity of ATPase C3 with heat-denatured DNA was suggested by its high sensitivity to ethidium bromide to be due to the double-stranded region in the heat-denatured DNA formed by self-annealing.

Morphological characteristics of the intestinal mucosa in the Afghan pika (Ochotona rufescens rufescens).

The intestinal mucosa of the pika was examined with the naked eyes, a light microscope and a scanning electron microscope and was compared with that of the rabbit. The duodenal mucosa of the rabbit showed wavy folds different from so-called villi, while that of the pika exhibited leaf-like or columnar villi. In addition to the specific ileocecal lymphoid apparatuses, the pika had the peculiar region between the cecum and the proximal colon. That region called "the constricted portion" possessed characteristic net-arranged folds and well-developed muscular layers. At the lateral surface of these folds, small villus-like protrusions projected into the lumen in large numbers. The spiral fold ran around the mucosal surface of the rabbit cecum, whereas numerous slender protrusions, cecal digitations, projected into the lumen of the pika cecum. Although the colon of the pika showed a similar external figure to that of the rabbit, some differences were obviously found in histological structures. The first segment of the pika proximal colon with three teniae possessed several protrusions and well-developed mucous glands, while that of the rabbit had neither protrusions nor mucous glands. The second segment of the pika proximal colon with one tenia was covered with numerous villus-like protrusions, while that of the rabbit was composed of wart-like protrusions. The tubular mucous glands were observed in the lamina propria of the pika as well as the rabbit. The distal colon of the pika showed a flat mucosal surface and possessed tubular mucous glands as observed in the rabbit.

Toxicological studies on Amfenac sodium (AHR-5850) (I) Acute toxicities in mice and rats.

Acute toxicities of Amfenac sodium (AHR-5850), new nonsteroidal anti-inflammatory agent, were studied in mice and rats. Each value of LD50 by oral, sc, im, iv and ip administration with this compound was 1190, 580, 540, 550 and 790 mg/kg for male mice and 1450, 625, 610, 630 and 710 mg/kg for female mice, respectively. Rats showed higher lethality than mice. There was no significant difference of sex in the values of LD50 for mice and rats. Movement and respiration rate followed by gastrointestinal ulcer, secondary peritonitis and systemic emaciation. These results suggest that the death is caused by secondary peritonitis and systemic emaciation due to gastrointestinal ulcer.

[Kinin forming system and ovulation].

The aim of the study is to clarify the role of the kinin forming system on ovulation. The results were as follows, Plasma prekallikrein of superovulated prepubertal rats was decreased to 50-60% of the normal level after PMS injection and 80-90% of the normal level after hCG injection. The lowest level was 33.8% 39 hours after PMS injection. Aprotinin suppressed the superovulation of prepubertal rats at doses of 16.7 X 10(4) KIE and 37.8 X 10(4) KIE (p less than 0.01). These results showed that the kinin production system has an effect on ovulation. As the sex steroid hormone, estradil benzoate, progesterone and testosterone propinate were used in oophorectomized mice. They had no effect on the plasma prekallikrein level. The plasma prekallikrein level during the estrus cycle of mature female mice was 85.5% of the control level in preestrus, 80.3% in estrus, 83.7% in metestrus and 89.8% in diestrus. The mean was 83.9%, which was significantly lower than that for oophorectomized mice (94.1%). It is proved that plasma prekallikrein was consumed and the kinin forming system was accelerated in the ovary with ovulation.

[Toxicological studies on a new cephamycin, MT-141, IV. Its acute toxicity in beagle dogs].

The acute toxicity of MT-141 was studied in adult Beagle dogs with intravenous (i.v.) or intramuscular (i.m.) administration to obtain following results. MT-141 at the doses ranging from 2,500 to 7,500 mg/kg i.v. caused no effect on life, bodyweight, food intake, eyeground and ECG in male and female Beagle dogs. MT-141 produced an increase in water intake, urine volume, WBC and LAP and a decrease in Lymph., U-Na, U-K and OP, but any histopathological change was not caused in the organs and tissues. It is suggested that these changes in blood, serum and urine are due to mechanical and transient effects induced by infusing a large volume of hypertonic solution of MT-141 into cephalic vein. When 1,000 or 2,000 mg/kg of MT-141 was injected into the muscles of hind legs, the hind legs had difficulty in walking. It is very probable that this change was due to mechanical effects induced by injecting a hypertonic solution of MT-141 at a rate of 70--130 ml/dog. An injection of 1,000 or 2,000 mg/kg i.m. of MT-141 changed activity of GPT, GOT and CPK in the serum within the limit of physiological variations but did not caused any effect on the other toxicological parameters such as bodyweight, food intake, water intake, urine volume, eyeground examination, ECG and histopathological examination. It is concluded from the above-mentioned results that MT-141 at the dose of 2,500--7,500 mg/kg i.v. or 1,000--2,000 mg/kg i.m. has no significant toxicity in Beagle dogs.
