[Toxicological studies on a new cephamycin, MT-141. VI. Its chronic toxicity in beagle dogs].

MT-141 was dosed to male and female Beagle dogs through intravenous (i.v.) route at 100, 200, 400 and 800 mg/kg/day for 182 days. The toxic effects of MT-141 were as follows. The male and female Beagle dogs exhibited no particular behavior and symptom except vomiting and soft stool after treatments with 800 mg/kg/day i.v. of MT-141. MT-141 even at the high doses of 400 to 800 mg/kg/day i.v. did not significantly change toxicological parameters such as consumption of food and water, volume of excreted urine, electrocardiogram, eyeground and analysis of blood, serum and urine. Only one of 4 male Beagle dogs died of severe intestinal invagination, which is not related to the drug effect, at 120th day after the start of treatments with the highest dose of 800 mg/kg/day i.v. of MT-141. The fatality of Beagle dogs for 800 mg/kg/day of MT-141 was 25% (1/4) in males and 0% (0/4) in females. MT-141 caused the hemorrhage accompanied with fibrosis and round cells at the site of injections but the change induced by MT-141 was not so much different from that by saline. MT-141 at the highest dose induced histopathological changes in only 2 females, such as atrophy and degeneration of cardiac muscle, necrosis and fibrosis of hepatic cells and hyperplasia of spleen and bone marrow cells. Electron-microscopic examinations revealed no ultrastructural change related to the toxicity of MT-141 in the livers and kidneys. It is considered from the above-mentioned results that the maximal "no effective" dose is 400 mg/kg/day i.v. in male and female Beagle dogs.

Hemoglobin Kariya [alpha 40 (C5) Lys leads to Glu]: a new hemoglobin variant with an increased oxygen affinity.

A new abnormal hemoglobin, Hb Kariya [alpha 40 (C5) Lys leads to Glu], with an amino acid substitution at the alpha 1 beta 2 contact was discovered in a young Japanese man. This variant migrated to the anode faster than Hb A, being nearly the same as Hb I in electrophoretic mobility. It amounted to about 6% of the total hemoglobin of the hemolysate. This hemoglobin showed an increased oxygen affinity, decreased heme-heme interaction and a lowered 2,3-DPG effect.

Emergency blood pool scintigraphy in fresh-blood-aspirated focal liver disease.

Emergency blood pool scintigraphy was performed in a patient clinically diagnosed as having a liver abscess, in whom percutaneous drainage revealed that the lesion containing fresh blood. 99mTc-RBC images 18 h after the injection showed only patchy activity in the lesion, therefore, we considered there was no communication between the lesion and the circulating blood. Drainage of the lesion was performed again and the abscess with a blood clot was drained.

[Basic and clinical studies on ceftazidime in the field of obstetrics and gynecology].

Ceftazidime (CAZ), a new cephalosporin antibiotic, was studied in the field of obstetrics and gynecology, and the following results were obtained. The absorption and tissue penetration of CAZ into intrapelvic genital organs were good after a single drip infusion of 1.0 g for 30--60 minutes. The maximum level of 76.4 micrograms/ml was obtained in uterine artery serum at 8 minutes after administration. The high concentrations were also obtained in genital tissues; the maximum concentrations ranged from 46.8--62.1 micrograms/g at 20 minutes after administration and the levels were as high as 2.1--7.7 micrograms/g at 5 hours and 40 minutes after administration. The concentration curves in tissues were consistent with those of serum levels. The concentrations of CAZ in retroperitoneal dead space exudate were determined after intravenous drip infusion of 1 g. The peak levels ranged from 26 to 32 micrograms/ml after 30 minutes of administration and the level of 8.53 micrograms/ml was sustained even 6 hours later. Good response was obtained in cases of gyneco-obstetric infections such as intrauterine infection, intrapelvic infection and external genital infection with daily dose of 2--4 g. CAZ was effective in 13 out of 14 cases (the efficacy ratio; 92.9%). As to side effects, gastric discomfort and vomiting were observed in 1 case.

Chromosomal aberrations observed in 52 mouse myeloid leukemias.

Chromosomes of 52 cases of mouse myeloid leukemia were examined. There were 5 myeloblastic leukemias, 22 granulocytic leukemias, 17 myelomonocytic leukemias, and 8 monocytic leukemias. Fifty cases were radiation induced and the other 2 were nonirradiated. Each case had leukemic cells with 1 to 10 marker chromosomes. Partially deleted No. 2 chromosomes appeared in 49 cases, including 2 nonirradiated cases. These deleted No. 2 chromosomes were varied in size, and they were classified into 7 types according to morphological features. There was no type-dependent difference in histological or cytological features among the 7 types. It was found that the chromosomal segment lying between Regions 2C and 2D was commonly missing from all of the deleted No. 2 chromosomes. In addition to such No. 2 chromosomes, an anomaly in chromosome 6 was observed in 16 cases, of which 12 cases were granulocytic leukemia. The abnormalities of chromosomes 3 and 9 were next most frequent, appearing in 14 cases each. Besides such structural anomalies, numerical changes involving the Y chromosome (33 cases), chromosome 6(6 cases), and chromosome 15 (4 cases) were also found. Characteristics of the karyotypes of the mouse myeloid leukemia in comparison with other leukemias were noted. The significance of the specific segments of the chromosomes which were commonly missing or trisomic in the karyotypes of neoplasias in mice, rats and humans was discussed. It was suggested that the genesis of myeloid leukemia was greatly influenced by the genetic information on chromosome 2 in mice.

Human ovum recovery via operative laparoscopy and in vitro fertilization.

Between January 1979 and December 1980, 142 laparoscopies were performed on 114 patients with long-standing tubal factor infertility. Twenty-eight were performed during spontaneous menstrual cycles, 20 following continuous human menopausal gonadotropin/human chorionic gonadotropin (hMG/hCG) therapy, and 94 following intermittent hMG/hCG stimulation. Follicular ripeness was judged by multiple criteria, which were also used to time the laparoscopy. Operative procedures were performed in all cases and follicular puncture was attempted in all but 16 subjects, where adhesions prevented access to the ovaries. In 28 spontaneous cycles, 28 follicles were punctured and 17 ova recovered; whereas in the 98 patients where ovulation was stimulated, 217 follicles were punctured and 43 ova collected. Following in vitro fertilization with the husband's spermatozoa and embryo culture, pronucleus formation, 2-cell, 4-cell, 8-cell, and 16-cell stages were observed in 18 oocytes obtained from 17 patients. Embryo transfer has, however, not yet been performed.

Comparison of fluid hysteroscopy and CO2 hysteroscopy.

A total of 247 women were subjected to liquid and CO2-hysteroscopy used as adjuncts in the screening of bleeding abnormalities in reproductive and post-menopausal groups. A point system of scoring proved quite useful. Using CO2-hysteroscopy the percentage of the detected malignancies corresponded to the percentages obtained by curettage in the age group of these patients. In liquid hysteroscopy, a group already suspicious for malignancy yielded a higher percentage of malignancies detected than normally expected. In post-menopausal women 41.7% of the cases examined wer malignant. The nine point scoring system used in liquid hysteroscopy correlated well with the histological findings. Both hysteroscopic techniques have their application and both are complementary, since they can be used for different indications in the visualization of the uterine cavity.

Influence of spermatozoal antibodies in the reproduction of mice.

Under in vitro and in vivo conditions, the influence of spermatozoal antibodies on isolated gametes and early embryonic stages up to implantation and into postimplantation fetal development was tested. In the in vitro experiment, a clear-cut correlation between the amount of antisera added and the reduction of fertilization rates was observed. In the in vivo experiments, female mice were immunized with homologous washed sperms. A correlation between the amount of immunization and the reduction of fertilization rate occurred. In the sperm-immunized female group, the percentage of large blastocysts showed statistically significant reduction in correlation to the amount of immunization. Spermatozoal antibodies had an effect on isolated gametes, on fertilization, and on the early development of the embryo as well as on implantation. No influence of sperm antibodies either in low or high titer sperm antisera on developmental stages occurred. No spermatozoal antibody-induced abortions occurred. However, an influence of high titer antisera on implantation was evident. (Am J Reprod Immunol. 1982; 2:225-232.)
