The influence of Hymenolepis nana infection on antibody responses to sheep red blood cells in mice.

The influence of Hymenolepis nana infection on the primary antibody responses to both sheep red blood cells (SRBC) and dinitrophenylated Ficoll-coated SRBC (DNP-coated SRBC) was examined in BALB/c mice. The in vitro cultures of spleen cells (Sp C) obtained from mice inoculated with either 100 or 1,000 eggs developed fewer plaque forming cells (PFC) than those of Sp C from non-infected mice, when the cells were exposed to SRBC. Thereafter, the number of PFC was gradually increased and almost reached the initial levels by day 21. On the other hand, the ability of Sp C obtained from mice infected with 10,000 eggs to produce PFC gradually dropped as the course of infection proceeded. However, H. nana infection did not reduce the capacity of Sp C to produce PFC to DNP-coated SRBC. Addition of T-cell enriched population from infected mice, which was obtained by passing through a nylon wool column, to anti-Thy-1.2 antibody treated non-infected Sp C significantly inhibited the production of PFC to SRBC. These results strongly suggest that H. nana infection impairs T-cell function in mice.

An unusual case of bronchial carcinoid predominantly consisting of large cells.

A 58-year-old woman presented an unusual variant of bronchial carcinoid. A tumor measuring 20 x 25 mm was recognized upon gross examination in the upper lobe of the right lung. Microscopically, the tumor consisted of large polyhedral cells with a pseudoglandular arrangement similar to pheochromocytoma cells. Immunohistochemically, the tumor cells contained serotonin, S-100 protein and neuron-specific enolase. Thus, we consider this neoplasm to be a large cell variant of bronchial carcinoid which, to our knowledge, has not been described in the literature.

[Clinical study of estramustine phosphate (Estracyt) on prostatic cancer].

Clinical effect of Estracyt was investigated in prostatic cancer patients. Twenty seven patients had been previously treated and 20 had not received prior treatment. Improvement rate of subjective symptoms was 85% in the previously untreated patients and that of objective findings was 85%, while those rates were 44% and 50% in the previously treated patients, respectively. Most of the adverse reactions were changes in mamma and mammary papilla which were considered to be due to the estrogenic activity.

20 beta-hydroxysteroid dehydrogenase of neonatal pig testis: purification and some properties.

20 beta-Hydroxysteroid dehydrogenase was purified from a cytosol fraction of neonatal pig testes to homogeneity as demonstrated by polyacrylamide gel electrophoresis (PAGE) and by isoelectric focusing. The molecular weight was estimated to be 30,500 using PAGE with sodium dodecyl sulfate and the gel filtration method. Molecular estimations showed that the purified enzyme consisted of a single polypeptide chain. It catalyzed the reduction of 17 alpha-hydroxyprogesterone to 17 alpha,20 beta-dihydroxypregn-4-en-3-one with NADPH. Furthermore, the C21-steroids, such as progesterone, pregnenolone, 17 alpha-hydroxypregnenolone, deoxycorticosterone, and deoxycortisol were also reduced by the purified enzyme. Apparent Km values for 17 alpha-hydroxyprogesterone, progesterone, pregnenolone, and deoxycorticosterone were 9.4, 1.5, 4.0, and 8.6 microM, respectively. The enzyme did not show 20 alpha-hydroxysteroid dehydrogenase activity. The maximum rate of enzyme activity was observed at 45 degrees C and optimum pH was at pH 5.5. The enzyme activity was strongly inhibited by heavy metal ions such as Hg2+ and Cu2+.

[Differences of behavioral rhythms observed by flat cage and running-wheel cage in female rat].

The ambulatory, wheel-running, and drinking activities were measured in Wistar-Imamichi strain female rats under 12 L:12 D condition (6:00-18:00), using Gundai type ambulo-drinkometer (for simultaneous measurements of ambulation and drinking) and wheel-drinkometer (for simultaneous measurements of wheel-running and drinking) to compare the rhythmicities of each behavioral activity. These apparatuses are able to measure the behavioral activities over a long period, successively and automatically. The circadian patterns of ambulatory activity had two large peaks at 21 or 24:00 and at 6:00 (acrophase). Contrary to the above results, the wheel-running activity exhibited clear mono-peak at 21:00 (acrophase). Thus, apparent differences of the pattern were observed between the two activities. However, ambulatory and wheel-running activities fluctuated showing 4-days rhythmicity, and both activities increased in estrus and proestrus stages, respectively. The circadian rhythms of drinking activities measured by both apparatuses showed almost same patterns with acrophases at 6:00, and 4-days rhythmicities were also observed and were characterized by remarkable decrease of activity in every proestrus stages. From these results, it is concluded that circadian pattern of ambulatory activity is different from that of wheel-running activity, but circadian patterns of drinking activities are stable regardless of different methods of the measurement. The ambulatory, wheel-running and drinking activities reflect the behavioral changes in sexual cycles.

[Treatment of oligospermic patients with varicocele].

The management of oligospermic patients with varicocele was reviewed. Varicocelectomy was performed in 46 oligospermic patients with varicocele (group 1) and kallikrein treatment was followed for those who failed to improve the semen quality after varicocelectomy (group 2). Twenty oligospermic patients with varicocele were given kallikrein for over 6 months as an initial treatment (group 3). A significant increase in total motile sperm count was noted in 34.8% of the patients in group 1, 22.7% in group 2, and 30.0% in group 3 respectively. Conception rate which was calculated after excluding couples, in which the partner received either artificial insemination by husband or any gynecological treatment, was 20.8% in group 1, 15.4% in group 2, 25.0% in group 3 respectively. No significant changes in hormonal data was observed after either treatment, but semen quality was not improved in patients who showed high serum level of luteinizing hormone (LH) or follicle stimulating hormone (FSH). Kallikrein is useful for oligospermic patients with varicocele as an initial treatment and for those in whom semen quality was not improved after varicocelectomy. Varicocelectomy and/or kallikrein treatment can be recommended mainly for those who show normal serum level of LH and FSH.

Peripheral neuroepithelioma in childhood. Immunohistochemical demonstration of epithelial differentiation.

Peripheral neuroepithelioma arising from the chest wall of a 4-year-old girl is described. She died of local recurrence 15 months after surgery. Light- and electron-microscopic as well as immunohistochemical findings confirmed the neuroectodermal nature of the tumor. A distinctive histologic feature was the presence of clustered epitheliallike cells, which immunohistochemically stained positive for both keratin and carcinoembryonic antigen. The epithelial nature of these peculiar cells is presented.

[Postoperative therapy of recurrent esophageal squamous cell carcinoma].

Some 218 resected cases and 43 autopsied cases after resection during the last 13 years were analyzed with regard to the mode of recurrence and survival. Several factors were investigated with the following results. 1) Extensive clearance of upper mediastinal lymph nodes during radical surgery may reduce the incidence of tumor recurrence. It is preferable to remove the cervical lymph node, however, after positive metastasis to cervical lymph node. 2) Beginning in 1981, interstitial brachytherapy of 192Ir seed assemblies using afterloading techniques was administered to two patients with esophageal carcinoma infiltrating the aorta and four patients infiltrating the trachea and main bronchus which were not completely resected in either case due to invasion of adjacent organs. No significant difference in life-prolongation was observed, but it may have been effective for prevention of localized recurrence in cases with aortic infiltration and with infiltration of the trachea and bronchus. 3) From 1984, eleven patients with recurrent squamous cell esophageal carcinoma were treated with combination chemotherapy sandwiched with irradiation (CDDP/5FU-RT-CDDP/5FU). Eight patients (73%) had complete or partial response, and six patients survived over one year after combination chemotherapy sandwiched with irradiation. Combination chemotherapy sandwiched with irradiation appears to be effective in the treatment of patients with recurrent squamous cell carcinoma of the esophagus. Toxic effects are moderate and manageable.

Immunohistochemical identification of somatostatin-containing neurons projecting to the median eminence of the rat.

Immunohistochemical staining of somatostatin (SRIF) and a retrograde transport method with horseradish peroxidase (HRP) were simultaneously applied to the same section of the rat brain to identify the specific SRIF-containing neurons sending their fibers to the median eminence. After HRP injection into the median eminence, SRIF-positive neurons in the rostral parts of the periventricular nucleus were shown to be labeled with HRP, SRIF neurons in the other brain areas, such as the amygdala, the ventromedial nucleus, dorsomedial nucleus and arcuate nucleus, had no HRP-positive material. The present findings appear to demonstrate that SRIF neurons in the periventricular nucleus project directly to the median eminence.

[A case of bilateral testicular tumors--a review of 115 cases in Japanese literature].

A case of bilateral testicular tumours of germ cell origin is reported. A 35-year-old man with bilateral testicular swelling visited our clinic. Bilateral high orchiectomy was performed. Histological examination of both resected testis revealed typical seminoma. Postoperative irradiation was performed. The patient is doing well after one year of postoperative follow-up without any sign of metastasis. Besides this case, we found 115 cases of bilateral testicular germ cell tumours in the Japanese literature. There were 39 cases with different and 76 cases with similar histology between the left and right testis. The age distribution, location of testis, time of onset and histological classification of these cases is discussed.
