Transformation of human diploid cells by adenovirus type 4 irradiated with ultraviolet light.

Inoculation of ultraviolet (UV)-irradiated adenovirus type 4 (Ad4) led to in vitro transformation of human diploid cells (HDC). Two transformed cell lines could be established: cell line H 1418, from HDC inoculated with the 10(-3) dilution of Ad4 UV-irradiated for 20 min at a distance of 20 cm, co-cultivated with uninfected HDC, and cell line H 1557, from HDC inoculated with the 10(-2) dilution of Ad4 irradiated at the same distance for 12 min. Both transformed cell lines were resistant to superinfection with homologous virus. Virus-specific antigen could be made evident by the indirect immunofluorescence technique in the nuclei of both H 1418 and H 1557 cells.

Rapid detection by immunofluorescence of multiple viral infections in patients with keratitis.

The presence of 12 viral (herpes 1 and 2, influenza A(H1N1), A(H3N2) and B, parainfluenza 1, 2, 3, adenovirus 5) and inframicrobial (Chlamydia, mycoplasma, Rickettsia burneti) antigens was investigated by direct or indirect immunofluorescence (IF) reactions in exfoliated conjunctival cells from 110 patients with nonbacterial keratitis and keratoconjunctivitis. A rapid etiological diagnosis could be obtained in 101 (92%) of the cases, parainfluenza and herpes antigens being the most frequently detected. In most of the cases the simultaneous presence of several antigens was made evident. Encouraging results were obtained by the application of a specific treatment based on the diagnosis provided by the IF reaction.

[Mucosal antrectomy with truncal vagotomy in duodenal ulcer resection to exclude the pylorus)].

The duodenal ulcers with which the surgeon is confronted increasingly take on tumoural postbulbar forms, penetrating in the pancreas or liver pedicle. Such a duodenal stump can neither be anastomized to the residual stomach ulcer inversed safely. Therefore it has become increasingly necessary to find a tight inversion method. One of the authors (D. Gavriliu) updated mucous antrectomy + truncular vagotomy and Reichel-Polya gastrectomy, which leaves an adequate seromuscular cuff for a double closing suture of the duodenum; after ablation of the antral mucosa the patient benefits by a veritable humoral antrectomy. In point of fact the operation is increasingly applied (truncular vagotomy + antrectomy and Reichel-Polya gastrectomy), with the difference however that antrectomy only removes the mucosa, and the advantage of ensuring perfect closure of the duodenal stump.

Experimental investigations in hamsters and rabbits with DNA extracted from human uterine tumors.

Experimental inoculation of a DNA preparation extracted from a fragment of non-irradiated human uterine cervix carcinoma was followed by the appearance of neoplasia in four hamsters and of lymphosarcoma in one rabbit. Similar DNA preparations obtained from three cases of irradiated human uterine cervix carcinoma and from a human uterine fibroma proved to have no biological activity.

Nucleic acid dot blot hybridization with fragmented digoxygenin labelled probes in human papillomavirus DNA screening.

Twenty human genital tissue samples from patients with different premalignant and malignant lesions were investigated by nucleic acid hybridization, using the dot blot technique. Molecular probes of various length--whole or fragmented papillomavirus genome--were digoxygenin-labelled. Human papillomavirus 16 DNA was identified in fifteen cases, well correlated with the clinical diagnosis. Fragments of papillomavirus genome proved to be efficient probes in nucleic acid hybridization as compared with the whole genome used for the same purpose.
