Heparin and platelet interactions influence alloantigen-induced proliferative responses of human T lymphocytes and immunoglobulin synthesis in vivo.

In contrast to defibrinated blood, cells isolated from heparinized blood give lower T-lymphoproliferative responses. Such responses can be also suppressed by addition of heparin to lymphocyte cultures. Platelets inhibit T cell proliferation and B cell differentiation, but their interactions with heparin may produce contrasting effects on different immune functions.

Immunomodulating activity of heparin.

Aside from its well-known anticoagulant action, heparin has many other biologic effects. Recent data emphasize the immunomodulatory effects of low dose heparin. The agent alters the traffic of lymphocytes blocking their expression of heparanase, an enzyme that digests the extracellular matrix allowing cell penetration to target tissues. It also has a weak direct immunosuppressive action in vitro and in vivo. Oral administration of heparin may cause immunosuppression, although the effects are weaker than after subcutaneous administration.

Nitrogranulogen-dependent inhibition of antibody synthesis. I. In vitro studies in man.

Nitrogranulogen causes a comparable suppression of immunoglobulin synthesis by human lymphocytes in vitro as a standard alkylating agent, 4-hydro-peroxycyclophosphamide. Mixed lymphocyte culture reaction-elicited T and B cell responses are especially sensitive to the agent. The immunosuppressive activity of NTG is enhanced by a thymic hormone (TFX).

Effect of FK506 and cyclosporine on primary and secondary skin allograft survival in mice.

The ability of FK506 and cyclosporine to prolong mouse skin allograft survival when given alone and in combination was studied during primary and secondary responses. The drugs were efficient immunosuppressants of both types of anti-graft responses but--as reported earlier--much lower FK506 doses were required to produce those effects. Both agents synergized when administered together during primary responses but not secondary responses. This suggests that the combination treatment may be less successful in a presensitized host, a finding with potential clinical application.

The effect of cyclophosphamide derivatives on cellular cytotoxicity (NK, ADCC) in vitro.

We have studied the effect of alkylating agents (AA): cyclophosphamide (Cy), 4-hydroperoxycyclophosphamide (4HCy), mafosfamide (MF), and nitrogranulogen (NTG) on cytotoxicity evaluated in ADCC and NK assays. While NK cells were sensitive to AA, higher concentrations of 4HCy and MF were necessary to affect killer cells active in ADCC assay. NTG which was without any effect on ADCC was found to be the most efficacious inhibitor on NK activity.

Immunological cellular response in denture stomatitis mycotic.

Antibody-dependent cellular cytotoxicity (ADCC) and lymphocyte-induced angiogenesis (LIA) of mononuclear leucocytes (MNC) isolated from the peripheral blood of patients with denture stomatitis mycotic were examined. The obtained results have shown a decrease in ADCC in patients with mycotic infection of oral cavity. Similarly, the ability to induce angiogenesis by MNC isolated from these patients was decreased in comparison with angiogenesis induced by control MNC. After the anti-mycotic treatment, an increase in ADCC and LIA was noted.

Effects of systemic etretinate treatment on natural cytotoxicity, immune angiogenesis and neutrophil adherence in patients with various forms of psoriasis.

The effects of etretinate treatment on natural killer cell activity, on angiogenic capability of peripheral blood mononuclears and on neutrophil adherence were studied in 20 patients with various forms of psoriasis. In all forms of active psoriasis, etretinate was found to affect first the angiogenic reaction and PMN adherence, whereas NK cell activity usually normalized only after a long-term therapy. The earliest normalization of all three parameters was noticed in pustular psoriasis, and it was correlated with the clinical improvement. In two cases of palmo-plantar pustular psoriasis, in 5 cases of arthropathic variety, and in 5 cases of common psoriasis, the normalization preceded clearing of the skin lesions.

Effects of psoriatic patients' neutrophils and sera on angiogenic capability and natural cytotoxicity of normal human peripheral blood mononuclear cells.

We studied the effects of products released from adhering neutrophils and serum factors from patients with psoriasis on angiogenic capability and natural cytotoxicity of normal human mononuclear cells. In active disseminated and guttate psoriasis, in which polymorphonuclear granulocytes were most adherent to the human vascular endothelium, supernatants from adhering neutrophils only slightly affected natural killer cell activity and angiogenic capability of normal human mononuclears. In peripherally spreading, plaque-like psoriasis, characterized by lower than in guttate psoriasis increase in granulocyte adherence, both neutrophil factors and patients sera markedly decreased NK cell activity and enhanced angiogenic capability of normal human peripheral blood mononuclear cells. The results suggest that neutrophil factors may affect some immune responses in patients with psoriasis.

Nitrogranulogen: a strong inhibitor of human lymphocyte proliferation.

The immunosuppressive activity of an alkylating agent Nitrogranulogen (NTG) was studied and compared with a major active metabolite of cyclophosphamide (4HCy). Both agents inhibit the human T and B cell proliferation in vitro induced by pure T cell (PHA) and B cell (SAC) mitogens. NTG is slightly less active than 4HCy and it is much less toxic to human lymphocytes. Furthermore, NTG appears to have a more selective inhibitory action against B cells.

Large granular lymphocyte lymphocytosis with neutropenia: a case report.

A case of large granular lymphocyte lymphocytosis with neutropenia was studied as evidenced by peripheral lymphocytosis of cells having typical morphology and profound neutropenia. Surface markers analysis revealed that almost all cells were CD8+ and their subpopulation DR+. The cells had strong spontaneous and inducible suppressor functions in vitro. Moreover, their ADCC activity was strong but NK activity very weak.

An optimal assay of human B cell proliferation in vitro.

The purpose of this work was to optimize an in vitro assay for measuring B cell proliferation in response to S. aureus Cowan I (SAC). It has been demonstrated that the best proliferative activity is obtained when 10(5) cells are cultured in flat-bottomed microtiter plates in the presence of 1:10,000 SAC dilution. Standard RPMI 1640 culture medium should be enriched with mercaptoethanol and 10% FCS or 0.5% human albumin. Human serum, found to be an optimal supplement for T lymphocyte responses, blocks B cell proliferation.
