Chemical identity of tryptensin with angiotensin.

Tryptensin, a vasopressor substance generated from human plasma protein fraction IV-4 by trypsin, has been isolated and the amino acid composition analysed. The procedures used for the isolation were: (a) adsorption of the formed tryptensin on Dowex 50W (X2; NH4+ form); (b) gel filtration through Sephadex G-25; (c) cation-exchange chromatography on CM-cellulose; (d) anion-exchange chromatography on DEAE-cellulose; (e) re-chromatography on CM-cellulose; (f) gel filtration on Bio-Gel P-2; (g) partition chromatography on high-pressure liquid chromatography. The homogeneity of the isolated tryptensin was confirmed by thin-layer chromatography and thin-layer electrophoresis. The amino acid analysis of the hydrolysate suggested the following proportional composition: Asp, 1; Val, 1; Ile, 1; Tyr, 1; Phe, 1; His, 1; Arg, 1; Pro, 1. This composition is identical with that of human angiotensin.

Structure, expression and mutational analysis of the hBRAG gene on 10q in the frequently deleted region in human endometrial cancer.

We previously reported that chromosome arm 10q is one of the target regions of allelic loss in human endometrial cancer. To identify the gene in this region responsible for endometrial cancer, we further characterized this region and localized the hBRAG gene. The function of hBRAG has not yet been fully studied, and there is the possibility that this gene works as a tumor suppressor. This gene consist of 7 exons and 6 introns encoding 503 amino acids; all the introns start with GT and end with AG in agreement with the GT-AG rule. Expression of this gene was studied by Northern hybridization and suppressed expression was observed in one (SK-UT-1B) of the six endometrial cancer cell lines. Mutation analysis in 38 primary EC tissues and six EC cell lines disclosed no genetic alterations. The genomic structure of hBRAG elucidated in this study should contribute to the future analysis of the hBRAG gene.

Multifocal primary lymphoma of bone: scintigraphy and MR findings before and after treatment.

A case of multifocal primary lymphoma of bone is reviewed. Ga-67 citrate, Tc-99m HMDP, and Tl-201 scintigraphs all showed multifocal radiotracer accumulation. CT showed subtle osteolytic and osteosclerotic changes with surrounding soft tissue density mass. MRI clearly showed abnormal signal intensity in the bone marrow. After systemic chemotherapy and infield radiotherapy, the patient showed clinically complete remission. MRI showed reduction of the extraosseous components, but there was little signal change in the bone marrow despite the clinical response to the therapy. Scintigrams were more useful than MRI and CT for both staging and assessing the early response to therapy. The soluble interleukin-2 receptor level was found to be related to tumor cell activity.
