The axonal localization of large Drosophila ankyrin2 protein isoforms is essential for neuronal functionality.

In polarized cells, such as neurons, ankyrin-type proteins are the major molecules that link the actin-spectrin-based membrane cytoskeleton to the plasma membrane. In Drosophila the second ankyrin gene, Dank2, is exclusively expressed in neuronal cells. Similar to ankyrin genes in other organisms, the Dank2 gene generates several ankyrin protein isoforms by differential splicing. Here we report that in Drosophila, the short Dank2 protein isoform is restricted to neuronal cell bodies and is excluded from axons, whereas the long Dank2 isoforms are localized specifically to axons. Thus the long and short Dank2 protein isoforms are localized to complementary neuronal subdomains, demonstrating that in vivo the composition of the neuronal cortical cytoskeleton is highly polarized. We show that once polarization is established, it persists during later stages of Drosophila development. We also present genetic evidence that the absence of axonal Dank2 protein is lethal.

[Screening and identification of egg mimotope of Schistosoma japonicum].

OBJECTIVE: To explore egg mimotope of Schistosoma japonicum that can be used in the development of diagnostic reagents for schistosomiasis. METHODS: By performing three rounds of biopanning in the affinity selection before picking out single clones for identification, target specific phages were effectively enriched from a random 15-peptides phage library with an immobilized mAb 6B12 which is specific to egg antigen of S. japonicum as a bait. By using ELISA, competitive ELISA with natural egg antigen as competitor, and Western blotting, 13 phage clones with high affinity and specificity to 6B12 were obtained and amplified from 400 single clones. RESULTS: DNA sequencing revealed that all the 13 selected single clones were identical in displaying gene sequence. Serum samples were tested by ELISA for the presence of IgG antibodies to the phages with mimic epitope, and showed that there was a significant difference between healthy volunteers and schistosomiasis patients. CONCLUSION: It is possible that the phage display peptide antigen, egg mimotope of S. japonicum may be a replacement of natural egg antigen for the diagnosis of schistosomiasis.

[Cloning and analysis of highly repetitive sequence fragments from takin (Budorcas taxicolor)].

Takin (Budorcas taxicolor) is a large animal living in China and other adjacent countries, which belongs to Bovidae of Artiodactyla. The anatomy, morphology and behavior of takin are between species of the subfamily Bovinae and Caprinae. It is now in a separated genus or in the same genus with muskox (Ovibos moschatus). To gain insight into the evolution of takin, we purified and cloned its highly repetitive BamHI fragments from the genomic DNA. The sequences of three fragments were highly homologous, indicative of units of a large repetitive DNA arrays. Southern hybridization using these fragments as probes showed identical patterns among individuals in both the same and different subspecies, implying conserved sequences and distributions of the BamHI clusters in takin genome. Sequence comparison with 1.714 and 1.715 satellite DNA of other species in Bovidae supports that takin has a closer relationship with the subfamily Caprinae than with Bovinae. It also suggests that these BamHI fragments may represent repeat units of the centromeric satellite DNA of takin.
