The nucleus accumbens shell is critical for normal expression of pup-retrieval in postpartum female rats.

The nucleus accumbens (NA) plays an important modulatory role in the control of normal expression of maternal behavior (termed maternal performance). The present study investigated the relative functions of two subregions of the NA (the shell and core) in maternal performance. Electrolytic lesions of the shell or core were performed either before parturition or immediately after a varying amount of maternal experience (none, 1 or 24h) during the immediate postpartum period. Maternal performance was tested on Day 1 postpartum and re-tested 9 days later using a pup sensitization technique. Results show that lesions of the shell, but not the core, significantly disrupted pup-retrieval: the shell-lesioned rats took significantly longer to finish retrieving all test pups, but their retrieval latency for the first pup was not affected. Neither lesion affected other components of maternal behavior (pup licking, nest building and nursing). These findings suggest that the shell, but not the core, is critical for the normal expression of pup-retrieval behavior possibly through its role in maintaining maternal motivation or attention.

Tandem PDZ repeats in glutamate receptor-interacting proteins have a novel mode of PDZ domain-mediated target binding.

The interaction of the glutamate receptor subunits 2 and 3 (GluR2/3) with multi-PDZ domain glutamate receptor-interacting protein (GRIP) is important for the synaptic trafficking and clustering of the receptors. Binding of GluR2/3 to GRIP requires both the fourth and fifth PDZ domains (PDZ4 and PDZ5) to be covalently linked, although only one PDZ domain is directly involved in binding to the receptor tail. To elucidate the molecular basis of this mode of PDZ domain-mediated target recognition, we solved the solution structures of the PDZ45 tandem and the isolated PDZ4 of GRIP. The two PDZ domains form a compact structure with a fixed interdomain orientation. The interdomain packing and the stable folding of both PDZ domains require a short stretch of amino acids N-terminal to PDZ4 and a conserved linker connecting PDZ4 and PDZ5. PDZ4 contains a deformed aB-bB groove that is unlikely to bind to carboxyl peptides. Instead, the domain stabilizes the structure of PDZ5.

[Ca2+]i regulates trafficking of Cav1.3 (alpha1D Ca2+ channel) in insulin-secreting cells.

Chronic exposure of pancreatic beta-cells to high concentrations of glucose impairs the insulin secretory response to further glucose stimulation. This phenomenon is referred to as glucose desensitization. It has been shown that glucose desensitization is associated with abnormal elevation of beta-cell basal intracellular free Ca2+ concentration ([Ca2+]i). We have investigated the relationship between the basal intracellular free Ca2+ and the L-type (Cav1.3) Ca2+ channel translocation in insulin-secreting cells. Glucose stimulation or membrane depolarization induced a nifedipine-sensitive Ca2+ influx, which was attenuated when the basal [Ca2+]i was elevated. Using voltage-clamp techniques, we found that changing [Ca2+]i could regulate the amplitude of the Ca2+ current. This effect was attenuated by drugs that interfere with the cytoskeleton. Immunofluorescent labeling of Cav1.3 showed an increase in the cytoplasmic distribution of the channels under high [Ca2+]i conditions by deconvolution microscopy. The [Ca2+]i-dependent translocation of Cav1.3 channel was also demonstrated by Western blot analysis of biotinylation/NeutrAvidin-bead-eluted surface proteins in cells preincubated at various [Ca2+]i. These results suggest that Cav1.3 channel trafficking is involved in glucose desensitization of pancreatic beta-cells.

[Reduced resistin levels in patients with type 2 diabetes mellitus].

OBJECTIVE: To measure the serum resistin level of patients with type 2 diabetes mellitus so as to examine whether there exists a relationship between resistin, obesity and diabetes. METHODS: ELISA was used to examine the fasting serum resistin, leptin, and true insulin and those 2-hours after taking 75-g glucose in 51 untreated type 2 diabetic patients, 30 males and 21 females, and 52 sex and age-matched normal control subjects. Blood glucose, blood pressure, height, weight, waist circumstance, hip girth were measured. Body mass index (BMI), waist to hip ratio (WHR), and quantitative insulin sensitivity check index (QUICKI) were calculated. RESULTS: In comparison with the control, the diabetic group had higher waist-to-hip ratio (WHR) and serum insulin levels (P < 0.05), but significantly lower resistin levels both in the fasting status (23 ng/ml +/- 15 ng/ml vs 30 ng/ml +/- 18 ng/ml, P < 0.05) and 2 hours after glucose loading (22 ng/ml +/- 11 ng/ml vs 31 ng/ml +/- 15 ng/ml, P < 0.001). The leptin level was not statistically different between the two groups (P > 0.05). The resistin level 2 hours after glucose loading was not significantly different between these 2 groups. Correlation analysis demonstrated that fasting resistin level was not correlated with sex, BMI, leptin, and blood pressure, but positively correlated with QUICKI (r = 0.30, P < 0.01) and negatively correlated with blood glucose (r = -0.21, P < 0.05). CONCLUSION: The serum resistin level of patients with type 2 diabetes is reduced rather than increased in fasting status and 2 hours after glucose taking. Resistin may not be the major link between obesity and diabetes in human beings. Since human resistin level is positively correlated with insulin sensitivity, the use of term "resistin", originally for its resistance to insulin, may be somewhat premature.

Pkd2 haploinsufficiency alters intracellular calcium regulation in vascular smooth muscle cells.

Autosomal-dominant polycystic kidney disease is a multiorgan disease and its vascular manifestations are common and life-threatening. Despite this, little is known about their pathogenesis. Somatic mutations to the normal PKD allele in cystic epithelia and cyst development associated with the unstable Pkd2(WS25) allele suggest a two-hit model of cystogenesis. However, it is unclear if this model can account for the cardiovascular pathology or if haploinsufficiency alone is disease-associated. In the present study, we found a decreased polycystin-2 (PC2, protein encoded by Pkd2 gene) expression in Pkd2( +/-) vessels, roughly half the wild-type level, and an enhanced level of intracranial vascular abnormalities in Pkd2 (+/-) mice when induced to develop hypertension. Consistent with these observations, freshly dissociated Pkd2 (+/-) vascular smooth muscle cells have significantly altered intracellular Ca(2+) homeostasis. The resting [Ca(2+)](i) is 17.1% lower in Pkd2 (+/-) compared with wild-type cells (P=0.0003) and the total sarcoplasmic reticulum Ca(2+) store (emptied by caffeine plus thapsigargin) is decreased (P<0.0001). The store operated Ca(2+) (SOC) channel activity is also decreased in Pkd2 (+/-) cells (P=0.008). These results indicate that inactivation of just one Pkd2 allele is sufficient to significantly alter intracellular Ca(2+) homeostasis, and that PC2 is necessary to maintain normal SOC activity and the SR Ca(2+) store in VSMCs. Based on these findings, and the fact that [Ca(2+)](i) signaling is essential to the regulation of contraction, production and secretion of extracellular matrix, cellular proliferation and apoptosis, we propose that the abnormal intracellular Ca(2+) regulation associated with Pkd2 haploinsufficiency is directly related to the vascular phenotype.

Genetic diversity of HIV-1 in western Kenya: subtype-specific differences in mother-to-child transmission.

BACKGROUND: Little is known about the impact of HIV-1 group M subtypes on mother-to-child transmission (MTCT) of HIV-1 in African settings where multiple HIV-1 group M subtypes are co-circulating. OBJECTIVE: To assess the role of subtype variation on MTCT. METHODS: HIV-1-infected women attending an antenatal clinic in western Kenya were enrolled for a prospective study (1996-2000) of MTCT. HIV-1 subtype analysis of p24gag and gp41env identified potential recombinants, and their role in MTCT was determined. RESULTS: Among 414 women for whom HIV-1 subtype and HIV transmission status were available, MTCT occurred in 80 (19.3%). MTCT rates were higher among women with subtype D compared with subtype A in either the gp41 region [31.6 versus 16.1%, relative risk (RR) 2.0, P=0.002] or p24 region (29.9 versus 18.0%, RR 1.7, P=0.02). Discordant subtype combinations were identified in 103 of the women (25.9%), and were associated with higher rates of MTCT (28.2 versus 17.0%, RR 1.7, P=0.01). In multivariate analysis, women with subtype combinations D/D, D/A, and A/D had an increased risk of MTCT (adjusted odds ratios 3.5, 2.5, 6.2; P=0.005, 0.05, and 0.0003, respectively) compared with A/A women after adjustment for maternal HIV viral load, placental malaria infection, episiotomy or perineal tear, and low birthweight. CONCLUSION: MTCT appears to be more common among mothers infected with subtype D compared with subtype A. Such differences in MTCT frequency may be caused by altered cellular tropism for placental cell types.

atTic110 functions as a scaffold for coordinating the stromal events of protein import into chloroplasts.

The translocon of the inner envelope membrane of chloroplasts (Tic) mediates the late events in the translocation of nucleus-encoded preproteins into chloroplasts. Tic110 is a major integral membrane component of active Tic complexes and has been proposed to function as a docking site for translocation-associated stromal factors and as a component of the protein-conducting channel. To investigate the various proposed functions of Tic110, we have investigated the structure, topology, and activities of a 97.5-kDa fragment of Arabidopsis Tic110 (atTic110) lacking only the amino-terminal transmembrane segments. The protein was expressed both in Escherichia coli and Arabidopsis as a stable, soluble protein with a high alpha-helical content. Binding studies demonstrate that a region of the atTic110-soluble domain selectively associates with chloroplast preproteins at the late stages of membrane translocation. These data support the hypothesis that the bulk of Tic110 extends into the chloroplast stroma and suggest that the domain forms a docking site for preproteins as they emerge from the Tic translocon.

Cav3.1 (alpha1G) T-type Ca2+ channels mediate vaso-occlusion of sickled erythrocytes in lung microcirculation.

In the present study, we demonstrate that lung microvascular endothelial cells express a Cav3.1 (alpha1G) T-type voltage-gated Ca2+ channel, whereas lung macrovascular endothelial cells do not express voltage-gated Ca2+ channels. Voltage-dependent activation indicates that the Cav3.1 T-type Ca2+ current is shifted to a positive potential, at which maximum current activation is -10 mV; voltage-dependent conductance and inactivation properties suggest a "window current" in the range of -60 to -30 mV. Thrombin-induced transitions in membrane potential activate the Cav3.1 channel, resulting in a physiologically relevant rise in cytosolic Ca2+. Furthermore, activation of the Cav3.1 channel induces a procoagulant endothelial phenotype; eg, channel inhibition attenuates increased retention of sickled erythrocytes in the inflamed pulmonary circulation. We conclude that activation of the Cav3.1 channels selectively induces phenotypic changes in microvascular endothelial cells that mediate vaso-occlusion by sickled erythrocytes in the inflamed lung microcirculation.

Synthesis and characterization of N2S3X-Fe models of iron-containing nitrile hydratase.

A series of iron complexes based on the pentadentate ligand 4,7-bis(2'-methyl-2'-mercaptopropyl)-1-thia-4,7-diazacyclononane), (bmmp-TASN)(2)(-), have been synthesized and characterized as models of iron-containing nitrile hydratase (NHase). The chloro derivative [(bmmp-TASN)Fe(III)Cl].0.5EtOH (1) contains a labile chloride which facilitates synthesis of related complexes via substitution reactions. Complex 1 is high-spin, g = 4.28. Addition of NEt(4)CN with 1 in CH(2)Cl(2) results in the cyanide ligated complex [(bmmp-TASN)Fe(III)CN] x 0.5EtOH (2), which shows a single intense nu(CN) band at 2083 cm(-)(1) in the IR region. Complex 2 is low-spin, g(1) = 2.31, g(2) = 2.16, and g(3) = 1.96. Under basic conditions complex 1 affords a mu-oxo bridged dimeric Fe(III) complex [(bmmp-TASN)Fe(III)](2)O (3), which shows an intense band at 799 cm(-)(1). Complex 3 was recrystallized from CH(2)Cl(2)/hexane solution in the triclinic space group P1, with a = 10.5486(15) A, b = 13.0612(19) A, c = 8.1852(12) A, alpha = 96.923(2) degrees, beta = 112.729(2) degrees, gamma = 81.048(2) degrees, and Z = 1. Density functional theory (DFT) calculations of the previously communicated iron-nitrosyl complex [(bmmp-TASN)Fe(III)(NO)][BPh(4)] (4) (Inorg. Chem. 2002, 41, 1039-1041) reveal that the HOMO region is dominated by Fe-S bonding. Complexes 1-4 display irreversible or quasi-reversible reductions in the cyclic voltammograms. All of the iron complexes and the zinc derivative, (bmmp-TASN)Zn (5), display an irreversible oxidation. Complex 5 was crystallized in the monoclinic space group P2(1)/n with a = 9.5759(6) A, b = 20.9790(13) A, c = 10.7113(7) A, beta = 91.283(1) degrees, and Z = 4.

Glutathione S-transferase pi is upregulated in the stromal compartment of hormone independent prostate cancer.

BACKGROUND: Glutathione S-transferase (GST) pi is a detoxifying enzyme abundant in normal prostate basal cells but only rarely expressed in prostate cancer cells. The current studies are the first to focus on GST pi in the stromal compartment of prostate tumors. METHODS: We employed immunohistochemical, immunofluorescence, and Western blot analysis to measure GST pi expression and subcellular localization in 21 primary and metastatic tumors from patients with hormone independent prostate cancer, as well as seven lymph node metastases and six prostatectomy specimens. RESULTS: GST pi was detectable in stromal cells in 17 of the 21 hormone independent prostate tumors. GST pi tissue distribution in hormone independent tumors coincided with vimentin staining, suggesting that GST pi is expressed by reactive fibroblasts and/or myofibroblasts. CONCLUSIONS: The current results suggest that prostate cancer cells induce an injury response in the stroma during progression to hormone independence, which results in GST pi expression. Stromal GST pi may contribute to chemoresistence of advanced prostate cancer.

Separation and identification of different refolding components.

A dual-gradient ion-exchange chromatography (IEC) refolding process has been successfully applied to renature lysozyme at high concentration. The different refolding components were separated by size-exclusion chromatography with low concentrations of urea and NaCl added into elution buffer. The hydrodynamic characteristics during separation and the hydrophobic properties of these components were also tested.

Reduced polyspermic penetration in porcine oocytes inseminated in a new in vitro fertilization (IVF) system: straw IVF.

High incidence of polyspermy is still a major problem in the in vitro fertilization (IVF) of porcine oocytes matured in vitro. This study was designed to examine whether embryo cryopreservation straws can be used to conduct IVF in porcine oocytes. The efficiency of this system was further compared with traditional microdrop IVF. Immature oocytes were aspirated from antral follicles and matured in vitro. After maturation, oocytes were inseminated either in straws or in microdrops with frozen-thawed boar spermatozoa. For straw IVF, sperm concentration and the presence of air columns between insemination segment and oil column were examined. Sperm-oocyte binding and cortical granules (CGs) before and after sperm penetration were examined by confocal microscopy. When various sperm concentrations were used for IVF in the straws with air columns, it was found that 5 x 106 cells/ml of sperm concentration was the optimal concentration; a high penetration rate (94.0%) and normal fertilization (oocytes with both male and female pronuclei) rate (38.2%) were obtained. Increasing sperm concentration to 10 x 106 cells/ml increased polyspermic penetration (61.9%) without affecting sperm penetration (86.9%). Reducing sperm concentration to 1 x 106 cells/ml reduced polyspermic penetration (25.6%), but sperm penetration rate (69.9%) was also reduced. When IVF was conducted in the straws with or without air columns, and in the microdrops, it was found that sperm penetration in the straws with air columns (96.5%) was significantly (p < 0.05) higher than that in the straws without air columns (81.7%) and in the microdrop (72.9%). However, the incidence of polyspermic penetration in the straws with air columns (34.2%) and without air columns (36.6%) was significantly (p < 0.05) lower than that (52.4%) in the microdrops. The number of spermatozoa bound to the oocytes was increased gradually in the straws but not in the microdrops in which more spermatozoa bound to the oocytes soon after insemination. CG exocytosis was more complete and faster in the oocytes inseminated in the straws than in the microdrops. These findings indicate that IVF of porcine oocytes in the straws provides a better condition in which more oocytes are fertilized normally than that in the microdrop IVF.

[Role of leptin in the pathogenesis of obesity-related hypertension].

OBJECTIVE: To assess the role of leptin in the pathogenesis of obesity-related hypertension and the relationship between blood pressure (BP), and body mass index (BMI), insulin resistance and leptin were examined. METHODS: 560 non-diabetic men, aged 35 - 75, selected from volunteers of health screening test during 2 000 were divided into hypertension group (n = 321, BP >or= 140/90 mm Hg without antihypertensive medication). and normal blood pressure group (n = 239). The body weight, waist hip ratio,BP, plasma glucose, serum lipids, true insulin (TI) and leptin were measured after overnight fast. Insulin sensitivity was assessed by the HOMA insulin resistance index (HOMA-R). RESULTS: Fasting leptin level showed good correlation with BMI, fasting TI, HOMA-R, BP and also triglycerides (all P < 0.01). After adjustment for age, BMI and HOMA-R, serum leptin was still positively correlated to SBP (r = 0.11, P < 0.05), and was significantly higher in hypertensive subjects than in normotensive subjects (geometric mean 6.4 vs 4.7 micro g/L, P < 0.001). Logistic regression analysis demonstrated that leptin remained significantly associated with hypertension after adjustment for potentially confounding factors. CONCLUSION: Leptin may play an important role in the pathogenesis of obesity related hypertension.

Vortex fluctuations in underdoped Bi(2)Sr(2)CaCu(2)O(8+delta) crystals.

Vortex thermal fluctuations in heavily underdoped Bi(2)Sr(2)CaCu(2)O(8+delta) (T(c)=69.4 K) are studied using Josephson plasma resonance. From the zero-field data, we obtain the c-axis penetration depth lambda(L,c)(0)=230+/-10 micrometer and the anisotropy ratio gamma(T). The low plasma frequency allows us to study phase correlations over the whole vortex solid state and to extract a wandering length r(w) of vortex pancakes. The temperature dependence of r(w) as well as its increase with dc magnetic field is explained by the renormalization of the vortex line tension by the fluctuations, suggesting that this softening is responsible for the dissociation of the vortices at the first order transition.

Craniofacial variation of the Chinese macaques explored with Morphologika.

In order to analyze, separately and dynamically, the variation in cranial size and shape of Chinese macaques, a new method, Morphologika, was used to illustrate 3D profiles based on the coordinates of 26 landmarks on the skull. Striking image variation between the two sexes was detected on the facial region: males exhibited a larger and more protrusive facial structure. Males also displayed a bigger cranium than females. The two sexes also showed quite different images in skull shape. However, they expressed the same allometric pattern with regard to the relationship between size and shape, which was significantly positively associated with each other along the first axis. The same relationship was negatively displayed along the second axis when the two sexes were analyzed together. However, only the relationship for females reached a significant level when the two sexes were studied separately. This was considered to be related to their differentiation in growth trajectory. This study also tested the concept of size revealed by the second axis of principal components analysis based on traditional morphometric methods on the same taxa.
