[Comparative characteristics of several properties of the estradiol-binding system of the human uterus and fallopian tubes].

Kinetic and thermodynamic parameters of estradiol-receptor interactions were studied in cytosol of human uterus endometrium and of uterus tubes. Specificity of these reactions was investigated. The receptor systems studied were distinctly dissimilar in the values of Ka equilibria, rate constants of association and dissociation with estradiol as well as in the affinity to a series of natural and synthetic estrogens.

[Possible heterogeneity of cytoplasmic estrogen receptors].

In order to elucidate whether specific estrogen-binding sites were heterogeneous, these sites were studied in uterine cytosols of guinea pig and green monkey Cercopithecus griseus. Concentrations of the estrogen-binding sites for labelled estrogens (estrone, estradiol, estriol and diethylstilbestrol) were estimated in the cytosols by means of the exchange method and Scatchard plot analysis. High linear correlation was shown between the Scatchard plots and the curves of estrogen-binding site complexes dissociation plotted in semilogarithmic coordinates (r-0.98 and 0.99, respectively; p less than 0.05). The linearity of the curves was confirmed by means of F-criterion. The data obtained suggest that only one type of specific estrogen-binding sites occurs in the cytosols at 30 degrees and at concentration of estrogens which did not exceed 50 nM. These estrogen-binding sites exhibited the same limited binding capacity for all the estrogens studied.

[Characteristics of estradiol-receptor interaction in the cytosol of the human Fallopian tubes].

Physico-chemical parameters (the rate constants of association and dissociation, half life of the estradiol-receptor complex, equilibrium constant, alteration of free energy of the bond) of estradiol-receptor interaction were studied in cytosole in human Fallopian tubes. Amounts of the cytoplasmic binding sites per cell were estimated. Specific interaction of the tube cytosole receptor system with 13 substances was examined. Steroid-receptor reaction has mainly been determined by presence of hydroxyl groups in the 3-d phenol and 17beta-positions. Nearest surroundings at the hydroxyls was also important for manifestation of the affinity of the steroids to the system studied.

[Estimation of 25-hydroxyvitamin D in blood serum using competitive binding with protein].

A modified procedure is described for estimation of 25-hydroxyvitamin D (25OHD) in blood serum, which involves a principle of the saturating analysis. Blood serum from rats, maintained on a ration free of vitamin D, was used as a binding system. Evaluation of the procedure was carried out by conventional criteria (accuracy, reproducibility, sensitivity). Content of 25OHD was estimated in blood of rats, maintained on rations containing various amount of vitamin D, as well as in blood of women in childbirth and in newborns from different climatogeographical zones.

[Multiple correlation analysis of the ratio between the sex hormone content of the peripheral blood and follicular fluid and the size of the follicle].

A multiple correlative analysis of the relationship between the follicle size, steroid hormone concentration in the follicular fluid and their peripheral blood content during different stages of the follicle growth was performed. It was suggested that both the growth of dominant follicle and the change in steroid hormone concentration are non-linear processes. It was demonstrated that steroid hormone content in the peripheral blood reflects steroidogenic activity of the dominant follicle thecal tissue.

[Effect of norethisterone microdoses on the functional state of the hypophyseal-ovarian system].

Effect of noretisterone microdoses (300 microgram) on a functional condition of the hypophysio-ovarian system was investigated. Hormonal studies were conducted in the time course of the control menstrual cycle and during 6 months of continuous noretisterone administration. Ovulation persisted in half of the women under study. Gestagen microdoses proved to produce a less pronounced effect on the hypophysio-ovarian system than estrogen-gestagen drugs.

[Effect of norethisterone acetate on ovarian functional activity].

Nine women were examined for a period of the 13th to 15th day of the menstrual cycle before and during norethisterone acetate use (10 mg within 10 days). Two-phase character of hormone secretion persisted in 6 of 9 cycles during norethisterone acetate administration. The changed hormone concentration in the 2nd phase and the shortened period of luteinizing hormone functional activity were seen in each cycle. Large individual differences were observed in the blood plasma norethisterone content. No correlation between norethisterone level and the ovarian response pattern was found.

[Specificity characteristics of estrogen interaction with the cytoplasmic receptors of the uterus and oviducts in green monkeys (Cercopithecus griseus)].

The specificity of interaction in Cercopithecus griseus uterus and oviduct cytosol receptor systems with 14 estrogenic substances has been characterized. It was shown that the activity of steroid-receptor interaction was mainly due to presence of hydroxyl groups in C3- and C17b-positions. The nearest environment of these functional groups in molecules affected also the affinity of steroids to the systems investigated. Analysis of the cytosol estrogen-receptor system specificity from Cercopithecus griseus, guinea pig, human uterus and oviduct showed the general pattern of the estrogen binding to the above-mentioned systems. Similarity was also noted between these Cercopithecus griseus and human estrogen-receptor systems.

[Role of the uterus in human cyclic ovarian function].

The ovarian function of 20 women with congenital uterine and vaginal aplasia was examined. Cyclic character of estradiol and progesterone secretion persisted in 19 patients. The concentration values of these hormones were within physiological norm in the majority of the women. Estradiol-estrone concentration ratio was lower than in the healthy subjects, showing a possible activation of estradiol metabolism in the uterus absence. The duration of the luteal hormone steroidogenic activity was the same as in the healthy women. Possible realization of the human reproductive system cyclic activity without the uterus is discussed.

Hormonal and immunologic methods in the diagnosis and treatment of patients with benign ovarian tumors and endometriotic cysts.

OBJECTIVE: To compare efficacy of various methods for characterizing endometriosis and benign ovarian tumors. DESIGN: Two parallel groups of patients, 54 with endometriotic ovarian cysts and 48 with benign tumors. SETTING: Central clinic. PATIENTS: Patients referred from all regions over a 5-year period. OUTCOME MEASURES: Estrogen Estrogen and progesterone receptor levels of endometrium and endometriotic foci, CA-125 concentration, and estradiol, progesterone, LH, and testosterone blood levels. RESULTS: LH levels elevated during follicular phase in patients; estradiol levels reduced; receptor levels abnormal in endometriosis, but not in benign ovarian cysts. CONCLUSIONS: Hormone therapy is adequate treatment for endometriotic cysts. CA-125 is not a reliable indicator for differential diagnosis.

[An intensive regimen of supersonic-frequency-current therapy in chronic inflammation of the adnexa uteri].

As shown by clinical, electrophysiological and hormonal examinations of 34 patients with chronic inflammation of the uterine appendages and control subjects, the disease treatment with intensive low-frequency current has no negative effects on cardiovascular and endocrine systems, improves function of the reproductive system. Intensive low-frequency currents are believed to be a useful modality in management of chronic genital inflammation.

[The clinical use of andriol in the hormonal therapy of male sterility].

The paper presents the experience in administration of testosterone andekanoate (andriol) in males suffering from infertility due to idiopathic oligozoospermia. Andriol produced a positive effect on ejaculate. As a result, 25% of the wives got pregnant. The response to andriol arises from posttesticular effects on maturation of spermatozoa and stimulation of the accessory sexual glands.
