[New microbiological Techniques in diagnosis of tuberculosis].

Microbial spectrum in patients with disorders of the upper respiratory tract consisted mainly of streptococci, staphylococci and pneumococci in 1991-1993, 1994 and later, respectively. Most specific for detection of nontuberculous flora was brushing with protecting agar plugs in combination with transport medium and anaerobic culturing. Direct immunofluorescence allows detection of antigens M. chlamydia in 29.4%, M. pneumoniae in 18.5% of cases. In diagnosis of tuberculosis the following new technologies were introduced: bacterioscopy with previous culturing of the material on liquid culture media followed by biological test and polymerase-chain reaction; determination of drug resistance based on nitrate reductase activity of M. tuberculosis indicating resistance of M. tuberculosis on liquid media in 4-7 days, dense egg media in 8-12 days, usage of Popesku medium enables correction of tuberculosis chemotherapy one month after the test initiation; M. tuberculosis identification using Western blot with monoclonal antibodies in some cases for 7 days.

[The use of immunoenzyme analysis and immunoblotting for the serological characterization of mycobacterial antigens].

The enzyme immunoassay and immunoblotting were used for the study of the serological activity of different mycobacterial antigens and the spectrum of antibodies to them in patients with different forms of tuberculosis and healthy persons. Antibodies in patients' sera were shown to bind antigens with different molecular weight. The level and spectrum of antibodies to purified protein fraction I made it possible to differentiate between patients with various forms of tuberculosis and healthy persons.

[Use of the method of integral rheography of the body for measuring the extracellular fluid volume in children].

To measure the volume of extracellular fluid, integral rheography is suggested. Investigations were carried out in 190 practically normal children aged 3 to 14 years. A well-defined reverse relationship was established between basic resistance and the volume of extracellular fluid. The given conclusion was confirmed by examination of children with the nephrotic syndrome, made during disappearance of edema. In edema, basic resistance was found to be reduced whereas in edema disappearance, it increased.

[The use of the microdot immunoenzyme method for determining antibodies to Mycobacterium tuberculosis antigens].

The microdot enzyme immunoassay (EIA) has been used for the determination of antibodies to M. tuberculosis protein fractions, crude antigenic preparations, PPD and old tuberculin in tuberculosis patients and healthy persons. Purified protein fractions have been found to possess the highest sensitivity and specificity in microdot EIA. The determination of antibodies to these fractions has permitted the differentiation of persons infected with M. tuberculosis from healthy ones. The use of M. tuberculosis protein fractions permits the determination of IgA and IgC in the sera of tuberculosis patients.

[Effect of dioxidine on extracellular proteins and enzymes in Staphylococcus aureus].

Dioxidine++-induced changes were shown to occur in the protein composition of the cells of Staphylococcus aureus. The most significant damages were observed in the composition of exoproteins. Dioxidine++ had a specific inhibitory effect on intracellular nuclease, which was accompanied by a decrease in virulence and disorders in the toxin formation.

[Transamination processes in mycobacteria with different biological properties].

The activity of aspartate and alanine aminotransferases in mycobacteria of different specific affinity and the degree of resistance to antituberculous drugs were studied. Differences in alanine and aspartic acid transamination processes taking place in mycobacteria with different biological properties were demonstrated.

[A comparative study of delayed hypersensitivity due to mycobacteria and their isolated protein fractions].

The role of intracellular protein fractions, isolated from M. tuberculosis virulent strains in the process of the electrophoretic separation, in the development of delayed hypersensitivity reactions has been studied. Low reactogenicity of protein fractions has been established on the basis of the development of faint immunopathological reactions in sensitized animals. High-molecular fraction 1 may be used as a sensitive preparation for the differentiation of individual mycobacterial species.

[Problems of the molecular mechanism of the effects of synthetic antiviral drugs].

Results of experimental study on the effect of bonafton, tebrofen, florenal and riodoxol on synthesis of virus specific proteins and RNA are presented. Selective inhibition of RNA-transcripts and respective proteins under the effect of bonafton was revealed. Florenal had a selective inhibitory effect on synthesis of virus specific proteins. Riodoxol did not inhibit synthesis of viral RNA and proteins of the influenza virus.

[The use of enzymes for searching for biologically active compounds].

Possible use of extracellular staphylococcal DNAase in screening substances with potential antibacterial activity was studied on quinoxalin as an example. For screening substances with antiviral activity the possible use of the influenza virus neuraminidase was studied on fluoren as an example. Close correlation between the biological activity of quinoxalin derivatives and the ability to inhibit DNAase was revealed. The most active inhibitors of the enzyme were dioxidin and other biologically active analogs of quinoxalin 1,4-di-N-oxide. The use of the extracellular nuclease as a biochemical model permitted to establish the structure/function dependence with respect to the quinoxalin derivatives. The effect of the fluoren derivatives on activity of the influenza virus neuramididase was studied. It was shown that florenal, an antiviral drug inhibited the virus specific enzyme by 80 to 90 per cent and had no effect on catalytic activity of bacterial neuraminidase. Biologically inactive and slightly active derivatives of the compounds did not inhibit the influenza virus enzyme. At the same time some of them lowered the activity of the bacterial enzyme.

[Characteristics of the protein complexes of clinical strains of mycobacteria].

The protein composition of Mycobacterium tuberculosis and the molecular weight of proteins contained in these organisms have been determined by the method of electrophoresis in the porosity gradient of polyacrylamide gel. Close similarity between the electrophoregrams of M. tuberculosis clinical and laboratory strains has been revealed. The study of 18 strains of different groups of mycobacteria has shown that M. tuberculosis are essentially different from opportunistic mycobacteria and acid-resistant saprophytes. These data may be important for the identification and taxonomy of mycobacteria.
