[The influence of antisecretory agents on the antral gastric mucosa in patients with duodenal ulcer].

Changes in the antral gastric mucosa of patients with doudenal ulcer after a long-term administration of antisecretory agents were studied. The subjects were five patients with a different duration of the disease. The methods applied included light and electron microscopy of tissue samplings taken from the antral part of the stomach during the periods of exacerbation and remission. In addition to well-known morphological changes, cellular principalis not typical of antral gastric mucosa were found in one patient during exacerbation and remission periods.

[Morphological characteristics and immunophenotype of dendritic cells generated from monocytes of human peripheral blood].

Morphological features and immunophenotype of mature dendritic cells (DC) generated from the monocytes of healthy donor blood after culture with GM-CSF and IL-4 with the addition of TNF, were studied using light and electron microscopy, as well as flow-cytometry. It was shown that DC were characterized by a number of morphological features such as: large size, eccentrically located nucleus, highly developed system of extensions, large vacuolated cytoplasm, prominent and activated Golgi complex and ribosomal apparatus. Mature DC are characterized by active surface expression of MHC and co-stimulatory molecules (MHC I, MHC II, CD40, CD80, CD86).

[Ultrastructural changes in cells of the antral gastric mucosa in patients with duodenal ulcers treated with melatonin].

AIM: To study ultrastructural condition of the cels in antral gastric mucosa in patients with duodenal ulcer on melatonin treatment. MATERIAL AND METHODS: Electron microscopy was made of biopsy specimens of antral stomach mucosa obtained from 14 patients with duodenal ulcer (DU) in exacerbation and in clinicoendoscopic remission after therapy with proton pump blockers (group 1) and after this therapy combined mth melatonin (group 2). RESULTS: In patients of group 2 there was an increased number of differentiated cover and principal cells, moderately differentiated parietal cells predominated. CONCLUSION: Melatonin is one of the drugs potentiating trophic processes in gastric mucosa.

[Ultrastructural criteria of clinical prognosis in hormonally non-active adrenocortical tumors].

The investigation was concerned with histological and ultrastructural features of adrenocortical tumors of the adrenals, which have a differential-diagnostic and clinico-prognostic relevance. Histological and electron-microscopical examination of 60 tumors (adrenocortical adenoma--12; adrenocortical cancer--48) was carried out and the findings were compared with clinical data. No significant correlation between histological pattern of adrenocortical tumors and survival was established. Ultrastructural evidence showed electron-microscopical examination to be a reliable procedure for making prognosis of adrenocortical cancers. There was a direct correlation between the level of differentiated cells and degree of their differentiation, on the one hand, and the quality of prognosis and survival, on the other. The significant predominance of differentiated cells was characteristic of adrenocortical adenomas.

[Cell differentiation in human melanoma cells caused by dicarbamine (ultrastructural investigation)].

The effects of dicarbamine (r) and polytransretinoic acid (PTRA) on human melanoma MEL-6 transplanted into Balb/c nude female mice have been compared by histological and electron microscopic procedures. It was discovered that long-term administration of 1.5 mg/kg dicarbamine caused melanoma MEL-6 cells to undergo terminal differentiation. As a result, the number of melanosome-containing cells was 3-4 times and the number of melanosomes in them--5 times those in intact controls. Dicarbamine effect was double that of PTRA.

[Immunohistochemical identification of hormonally-inactive tumors of the adrenal gland].

An immunohistochemical study was carried out to differentiate between such neoplasms of the cortical and cerebral layers of the adrenal as adrenocortical tumor and pheochromocytoma. It used antibodies to chromogranin A, synaptophysin, vimentin, cytokeratin 8, pancytokeratin, carcinoembryonic antigen, epithelial membrane antigen, neurofilaments, alpha-fetoprotein, alpha-antitripsin and neuron-specific enolase. Chromogranin A and synaptophysin appeared to be immunohistochemical markers for pheochromocytoma since it responded negatively to vimentin and cytokeratin. Adenocortical tumors were associated with positive reaction to vimentin and a negative one to cytokeratin. These characteristics were used to differentiate adrenocortical tumors from adenomas which responded to cytokeratin and hardly to vimentin. A possibility of adrenal tumors consisting of endocrine and neurogenic cells is considered.

[New models of experimental chemotherapy of tumors].

New neoplastic models such as orthotopic solitary hepatic tumor (cholangiocellular cancer PC-1) as well as different strains of malignant pleuritis (hemoblastosis and ascitic tumors) have been evolved by transplanting tumor cell suspension to rat liver or murine pleural cavity. Intrahepatic cancer PC-1 has a solid mucosa-excreting structure and is characterized by low activity of detoxication enzymes of such xenobiotics as glutathione-S-transferase, NAD(p)N-chinonoxyreductase and aldehyde dehydrogenase. Orthotopic hepatic tumor PC-1 may be used for evaluating systemic and regional therapy. Models of tumorous pleuritis described with respect to their response to chemotherapy, may have an application in screening and preclinical examination of newly-developed antitumor drugs.

[Histochemical and biochemical study of radiation injury of the kidneys in gas hypoxia].

A radioprotective effect of hypoxia was studied in kidney radiation. The use of hypoxic respiratory mixture containing 8% oxygen (the normal content is 21%) enhances kidney resistance to local single and fractionated irradiation. Determination of renal activity of succinate dehydrogenase, lactate dehydrogenase, acid and alkaline phosphatase and urinary N-acetyl-beta-D-glucosaminidase, gamma-glutamyl transferase, alkaline phosphatase, lactate dehydrogenase is recommended as tests for kidney radiation damage.

[The expression of cytokeratin No. 17 in squamous cell cancer of the cervix uteri: an immunohistochemical study of 19 cases].

Monoclonal antibodies E3 against cytokeratin No. 17 were used for immunohistochemical examination of samples from 19 squamous-cell carcinomas of the uterine cervix and 10 cases of squamous-cell metaplasia of the endocervix. The above antibodies selectively labelled reserve cells of the cervical canal mucosa, basal layer of mature metaplastic epithelium and parabasal and basal cells of immature metaplastic squamous epithelium. In large-cell keratinizing, non-keratinizing and small-cell carcinomas, the expression of cytokeratin No. 17 revealed tendency to basal orientation and was in many respects similar to that in metaplastic squamous epithelium. It was suggested that all the squamous-cell carcinomas studied originated from reserve (metaplastic) cells.

[A method for the primary biological assessment of the photodynamic activity of photosensitizers in a nontumor model in vivo].

In comparison with traditional laboratory methods of photosensitizers screening in vitro and in vivo on transplanted tumors, the proposed method of PDA evaluation has the advantages. Criterion of PDA is the quantitative characteristic--the area of photodynamic necrosis on the surface of laboratory mice liver 24 hours after combined action. According to the screening results, PDA of photosensitizers is distributed: AIPcS2-3 > AIPcS1 > Photofrin-2 > AIPcS3 > or = HpDChina > Fotosan-3 > Fotosan-2. Morphological research allows to characterize PDA of photosensitizers qualitatively. Thus, AIPcSx derivatives have more expressed cytolytic and nucleotropic effect on parenchyma cells (hepatocytes) in comparison with HpD derivatives.

[The role of monoclonal antibodies to the epithelial membrane antigen in the diagnosis of human tumors].

The paper discusses the possibility of the use of the Soviet-made ICO 25 monoclonal antibodies to membrane antigen of lipid globules of the human milk for differential diagnosis of human tumors. ICO 25 monoclonal antibodies reliably detected the above antigens in normal epithelial and breast cancer cells. However, these antigens cannot be considered strictly specific for breast tissue. They were found in various human epithelial tissues, in the majority of epithelial tumors and lymph node metastases. Staining for ICO 25 monoclonal antibodies was negative in non-epithelial tumors. The above antibodies proved a useful marker for the identification of epithelial origin of primary tumors and their metastases showing unclear histology. They can be used to differentiate between epithelial and non-epithelial malignancies as well as to detect micrometastases and areas of microinvasion. Paraffin-embedded samples can be used for immunohistochemical examination.
