[Clinical study on the permeability of FT-207 and 5-FU in primary lung cancer].

Distribution of the FT-207 and 5-FU between plasma and lung tissue as well as tumor tissue was studied in 28 patients with lung cancer after administration of FT-207 preoperatively. Two methods of administration were employed, one is intravenous drip infusion (IV Group) (800 mg/day for three days) and the other is suppository (Supp Group) (750 mg 2/day for three days). The level of FT-207 was higher in plasma than in normal lung tissue or tumor tissue in IV Group. There was no difference in the level of FT-207 between plasma, normal lung tissue and tumor tissue in Supp Group. The level of 5-FU was higher in tumor tissue than in plasma both in IV Group and Supp Group. Furthermore, intravenous drip infusion was more effective method to give FT-207 because of higher level of 5-FU in tumor tissue than in normal tissue. Comparison of the tissue level in IV Group between two histologic types of tumor, i.e. squamous cell carcinoma and adenocarcinoma, disclosed that there was no significant difference in the concentration of FT-207 and 5-FU both in normal lung tissue and in tumor tissue.

Enzyme immunoassay of 11-deoxycortisol.

Enzyme immunoassay of serum 11-deoxycortisol was established by using alkaline phosphatase as a label. The labelled alkaline phosphatase was stable for at least a year. 11-Deoxycortisol in serum was purified by applying to Sephadex LH-20 column prior to present enzyme immunoassay. Separation of bound and free form was carried out by adding anti-rabbit gamma-globulin goat antiserum as a second antibody. The minimal amount of 11-deoxycortisol detected was 40 pg/tube and the measurable range was from 0.04 to 10 micrograms/dl. Intra- and interassay coefficient of variations were 3.8-4.5% and 3.4-5.3%, respectively. 11-Deoxycortisol values determined by the present method correlated well with those determined by radioimmunoassay (r = 0.98, y = 0.93 x + 0.19, n = 28). This enzyme immunoassay satisfied the standard criteria of dilution and accuracy, and is applicable to routine determination of serum 11-deoxycortisol in any clinical laboratory as a non-isotopic immunoassay.

Solid phase fluoroimmunoassay for 11-deoxycortisol in serum using 21-amino-17-hydroxyprogesterone.

Solid phase fluoroimmunoassay of serum 11-deoxycortisol (17,21-dihydroxy-4-pregnene-3,20-dione) was established using fluorescein isothiocyanate-labelled 11-deoxycortisol and anti-11-deoxycortisol antibody-conjugated polyacrylamide beads. 21-Amino-17-hydroxyprogesterone (21-amino-17-hydroxy-4-pregnene-3,20-dione) was synthesized as a useful derivative for preparing the fluorescent dye conjugate. Serum 11-deoxycortisol was measured with this assay system after extraction and purification by Sephadex LH-20 column chromatography. The minimal amount of 11-deoxycortisol detected was 40 pg/tube and the measurable range was from 0.04 to 5.0 microgram/dl. Intra- and inter-assay coefficients of variation were 8.3% (n=6) and 9.8% (n=5), respectively. 11-deoxycortisol values determined by the present assay correlated well with those determined by radioimmunoassay. The present assay is particularly suitable for estimating the conditions of the pituitary and adrenocortical functions.

[Experimental examinations and toxicokinetic analysis of the absorption and excretion of 1,1,1-trichloroethane by the lung].

Of fifteen dogs each five of them inhaled 1,1,1-trichloroethane (1,1,1-TCE) at the levels of 700, 1500 and 2,000 ppm respectively for one hour. Changes in 1,1,1-TCE concentrations in the expired air, arterial blood and venous blood of dogs were measured from the beginning of exposure until one hour after the end of exposure. The results obtained were as follows. 1. Cumulative uptakes of 1,1,1-TCE in one hour exposure were 27.3 mg/kg at 700 ppm, 44.7 mg/kg at 1,500 ppm and 71.2 mg/kg at 2,000 ppm. 2. Excretion ratios of 1,1,1-TCE in one hour after the exposure were 66-71% at all the three levels of exposure. 3. During the exposure, highly significant negative correlation was seen between arterial/venous blood concentration ratio and expired/inspired air concentration ratio. In the post-exposure period, highly significant positive correlations were seen between expired air concentration and venous or arterial blood concentration. 4. The mean ratio of the total absorption value to the total inspired value was 14% at all the three levels of exposure. 5. From these results, the authors examined the multiple regression equations with two independent variables x and z which are shown as follows. a) During the exposure period, Y1: Cumulative absorption solvent value by the dog (mg/kg) X1: Time of exposure (min) Z1: Level of the solvent value in the ambient air (microgram/ml) log Y1 = -0.526 + 0.622 log X1 + 0.905 log Z1 b) Post exposure period, Y2: Cumulative excreted solvent value by the dog (mg/kg) X2: Time after the end of exposure (min) Z2: Cumulative absorbed solvent value by the dog at the end of exposure (mg/kg) log Y2 = -2.300 + 0.609 log X2 + 0.858 log Z2 We assumed that both equations are very fit and significant.
