The assay kit of Toxoplasma latex agglutination (LA) microtiters (Toxotest and Toxocheck "Eiken") was applied for the diagnosis of Toxoplasma infection in pig, cat, dog and some rodents. Frequency distribution of the LA titers of pigs and cats showed a through at 1:32. The starting point of the reaction for positive diagnosis was 1:64 dilution in these animals and the kit was found to be practical and useful. Application of the kit to the diagnosis of dogs was not yet successful. The LA titer of experimentally infected mice and rabbits with Toxoplasma was detected from 1--2 weeks after infection. In wild rats caught in Sapporo City 26.1% had LA titers of 1:4 or more.

Fluorescence quenching immunoassay of serum cortisol was established. The minimal amount of cortisol detected was 3.1 ng/tube and serum concentration of 3.1 micrograms/dl to 100 micrograms/dl of cortisol could be measured. Intra- and inter-assay coefficient of variation were 7.7-10.5% and 10.7-13.3%, respectively. Cortisol values determined by the present method correlated well with those determined by radioimmunoassay (r = 0.97, y = 0.89 x + 0.88, n = 33). This fluorescence quenching immunoassay satisfied the standard criteria of dilution and accuracy, and is a rapid and simple method requiring no antibody-bound and free separation.

Direct fluorescence polarization immunoassay of serum cortisol was established. Non-specific binding of fluorescent-labelled cortisol to serum proteins was successively eliminated by sodium dodecyl sulfate. The minimal amount of cortisol detected was 0.1 ng/tube and serum concentration of 1.0 microgram/dl to 100 microgram/dl of cortisol could be measured. This fluorescence polarization immunoassay satisfied the standard criteria of accuracy and precision. The values correlated well with those obtained by radioimmunoassay.

Investigation of the mechanism of the lowering effect of hydrochlorothiazide, which was administered orally, in the determination of estriol from the pregnancy urine has been carried out. The isolation and determination of the structure of a copolymer formed from estriol and formaldehyde have suggested the mechanism by which, during the acid hydrolysis process of the glucuronide conjugate of estriol, some copolymers of estriol and formaldehyde generated from hydrochlorothiazide, were formed, thus lowering the observed values of the estriol contents in the urine.
A new method for the determination of total and conjugated bilirubins in serum with use of sodium dodecyl sulfate (SDS) is described. In the presence of SDS, the reaction of bilirubin with diazotized sulfanilic acid proceeded smoothly and was completed within 15 min at room temperature, thus resulting in a clear solution having a maximal absorbance at 565 nm which was linear up to a level of 200 mg/l bilirubin concentration. The advantages of this method are: (1) a clear reaction mixture results in any case; (2) the result is not affected by hemolysis, lipemic and turbid sera. The proposed method offers a new general application for the measurement of total and conjugated bilirubins in serum.

With the parameters of a flow-volume and a volume-time curve, the discriminant analysis of bronchial asthma is described. The subjects were classified into three groups (healthy adults, mild asthmatic patients and moderates ones). The difference of the mean vectors of the parameters of the three groups was made clear by the selection methods of the discriminant analysis between any two of the groups both with 6 parameters (%FVC, FEV1.0%, peak flow rate (PF), flow rate at 50% of FVC (V50), flow rate at 25% of FVC (V25), and V50/V25) and with 8 (6 parameters mentioned above and V75, V10). Forced expiratory volume in 1 second percent (FEV1.0%) or V50 was selected at the first step with 6 parameters, and V75 was selected at the first step with 8 parameters. Probabilities of misclassification with 8 parameters were lower than those with 6 ones and the probability of misclassification at the discriminant analysis between healthy adults and mild asthmatic patients with 8 parameters was 15.75% at the final step.
