Organization and expression of the transforming region from the European elk papillomavirus (EEPV).

The nucleotide sequence of the early (transforming) region from the European elk papillomavirus (EEPV) double-stranded DNA has been determined together with flanking regions. The established sequence, which is 5732 bp long, shows that the genome of EEPV is closely related to the previously sequenced bovine papillomavirus type 1 (BPV-1) and deer papillomavirus (DPV) genomes. Seven open reading frames (ORFs), designated E1-E7, were identified in similar positions as in the BPV-1 genome. The E1 and E5 regions were best conserved. The strong homology between the E5 ORFs of EEPV, BPV-1 and DPV is interesting in the light of the recent proposal that these ORFs encode a major transforming function (Schiller et al., 1986; DiMaio et al., 1986). A set of mRNAs, comprising six size classes, were identified in EEPV-transformed cells. At least two different promoters appear to control EEPV transcription in transformed cells.

Quantitative hematological characteristics in MZ and DZ twins.

The level of several biochemical hematological characteristics was determined--using colorimetric methods--in a group of 111 twins, aged 16-22 years. Significant variance differences between MZ and DZ twin pairs were observed in metabolites and serum enzyme levels. The intrapair differences of the variance of ions and protein levels were comparable in MZ and DZ twins. The validity of quantitative determination of heterability of biochemical characteristics is discussed.

Stability studies on human pituitary prolactin.

Differently charged isomers of human pituitary prolactin were subjected to stability studies under various conditions. The study was guided by analytical electrophoresis and radioimmunoassay. In alkaline medium (pH 10) it was found that each isohormone was converted into faster-migrating components, probably due to deamidation. A quantitative study of this alteration was made by measuring the rate constant for this conversion assuming first-order kinetics. The result indicated that the rate constant decreased with increased acidity of the examined prolactin components. Furthermore, this alteration was also found to be paralleled by a decreased immunoactivity of the components. At neutral pH the conversion reaction was studied both in 0.9% NaCl and in human serum. For the least acidic isohormone the rate constant in serum was calculated to be about four times higher than that in the saline solution which in turn was comparatively low. It was concluded that the alteration observed at pH 10 might be attributed to non-enzymic deamidation. The conversion of prolactin that occurred in human serum indicated that this reaction could be caused by enzymes. A study of the immunoactivity of human prolactin as a function of time at various storage conditions is also included in this work. The stability of the hormone was found to be strictly concentration dependent and also affected by buffer ionic strength. In the presence of ethylene glycol (50%, v/v) at -20 degrees C and at a protein concentration in the range of 0.1-2 mg/ml the hormone was found to be both immunologically and electrophoretically stable for several years.

Usefulness of 99mTc pyrophosphate bone scintigraphy in the survey of dialysis osteodystrophy.

Fogelman's score (FS) was used to determine the usefulness of 99mTc pyrophosphate (Tc-PP) bone scintigraphy in the evaluation of dialysis osteodystrophy. FS correlated well with bone 47Ca accretion rate. It remained stable after six months in patients treated with 1 alpha (OH)D3 and increased significantly in a randomised group of untreated patients. It decreased after two years of 1 alpha (OH)D3 therapy while serum calcium increased and iPTH and alkaline phosphatases decreased. Patients with low FS, treated by 1 alpha (OH)D3, rapidly developed hypercalcaemia. In cases of spontaneous hypercalcaemia, parathyroidectomy did not normalise serum calcium in patients with low FS despite a significant decrease in serum iPTH. Lower FS were associated with a higher increase in serum aluminium after desferrioxamine (DFO) administration and in two cases of proven aluminium osteomalacia, DFO therapy was followed by a dramatic increase in FS.

Multi-dimensional complexes of skull traits.

The simultaneous incidence of sutural bones was studied by a multidimensional analysis (Wanke method). On a medieval sample of 249 male and female skulls from Lower Austria the presence of two main complexes of sutural and fontanelle bones was observed. The interaction of possible genetic factors and developmental growth incongruences in causing these phenomena are discussed. The necessity of further studies considering such basic biological aspects with minor skeletal traits is stressed.

Characteristics of donor semen and cervical mucus at the time of conception.

With freshly ejaculated semen from 15 donors, 1017 inseminations were performed in 247 women. One hundred conceptions were achieved. The degree of crystallization of the cervical mucus and the characteristics of the donor semen were examined at each insemination. One insemination only from one single ejaculate was performed in each cycle. This procedure made it possible to describe the quality of the cervical mucus and the sperm sample characteristics on each occasion of conception. The conception rate was significantly reduced (P less than 0.001) when the degree of crystallization of the cervical mucus was impaired. The conception rate was low when the sperm density was less than 20 million/ml, the percentage of abnormal sperm was above 50, and the degree of motility was graded 2 or less. The in vitro capillary sperm penetration test gave reliable information on the progressive motility of the sperm, showing a good correlation between penetration ability and conception rate.

A study of parental attitudes after donor insemination (AID).

Artificial donor insemination (AID) is important in the treatment of infertility when the couples' wish for a family is denied because of male sterility. However, very little is known regarding the continued development of AID families and their attitudes to insemination after the child is born. The study is based on a questionnaire anonymously completed by 92 couples returning to obtain a second child by insemination. None of the parents regretted his or her decision regarding insemination and the majority were prepared to recommend the method to other couples in the same situation. Apart from one couple the parents intended to withhold the truth from the child regarding its origin. The majority of parents chose not to inform anyone else regarding the child's conception. The parents were therefore anxious that case notes and details concerning insemination were treated with the utmost discretion and confidentiality. The parents in this study clearly indicated they preferred insemination to adoption. A large proportion of the parents who already had a child as a result of insemination returned to the clinic for a second child.

[Methodological contributions to craniometric studies of some early medieval populations of the upper Donau region. II. Comparison of early medieval populations on absolute skull measurements].

On the basis of the means of 10 cranial measurements of 7 medieval populations different methodical approaches of the two commonly used distance estimation methods are compared with respect to their relevance. In addition also the results obtained by the same methods but based on different data (5 indices or 10 measurements, respectively) were compared. In spite of the fact that (DD)2 distances and the dendrograms (after Creel) give better results than the other methodical and graphic variants, the choice of qualified characters is more important than a certain distance method according to the authors' opinion. Thus the groupings and the clustering on the basis of the means of only 5 cranial indices show the best accordance with the ethnogenetic and historical conditions. An additional examination of the segregative values of cranial measurements revealed that the omission of the smallest frontal breadth and probably also the nasal height and breadth does not result in a reduction of the segregative accuracy.

[Methodological contribution to craniometrical studies of various early mediaeval populations of the upper Danube region. I. The most usual cranial indices as a basis for population comparisons (graphic and approximation methods)].

Using the means of five cranial indices of three early medieval groups from Austria and four comparative series from neighbouring regions (Southern Germany, CSSR and Hungary) distance measures and similarity relations were computed and the applied methods were compared. In addition, the attempt for a better consideration of the biological conditions was made by combining two known methods at a time (Dendrogram by means of (DD)2 distances; population-composition on the basis of adjacent secured comparative series). The results of this study reveal, that using the above cited simple methods on the only basis of the means of five cranial indices a correspondence with the attributions of the three Austrian series cited in the literature and basing on extensive investigations was obtained. Thus a very strong relation -- especially concerning the males -- resulted between the series from Pitten and the Slavic ones, and between the groups of Zwölfaxing -- more expressed in the females -- and the Avaric series. Of special interest are the results of the mixed population of Zwentendorf were the females resemble quite strikingly the Slavic, the males, however, the Bajuvaric comparative series.

[Quantitative mathematical evaluation of growth studies from the viewpoint of exogenously or genetically influenced variability].

Firstly, the ideas are sketched which serve as the basis for the phenomenologic-mathematical kind of modeling of the body length growth process of man. For proving the biological relevance of the spurts analyzed by numerical procedures one has to consider the social and the biological circumstances in which the growth process takes place. An analysis of longitudinal data series of the body length of (monozygotic) twins will give further hints to the possible meaning of the growth spurts by way of separation of exogenous and of genetic determined endogenous agents on the growth process. The data available for our examinations cover the time interval of the praepubertal and the postpuberal development as well as the time interval in which the puberal growth spurt takes place. The way to proceed in evaluation these time series will be presented.
