Effect of smoking on semen quality of infertile men in Shandong, China.

AIM: To study the effect of smoking on the semen quality in infertile men in Shandong Province, China. METHODS: Adult non-drinker males attending the infertility clinic, including 110 non-smokers and 191 smokers, were recruited for the study. Sixty-one fertile, non-smoker and non-drinker males, who had one or more children, served as the controls. The smokers were divided into subgroups according to the amount and duration of smoking. Semen parameters (semen volume and sperm density, viability, motility, and morphology) were examined and seminal plasma contents of Zn , Cu and superoxide dismutase (SOD) determined. RESULTS: The semen volume and acidity, and the sperm density, viability and forward progression, as well as the seminal plasma contents of Zn, Cu and SOD were much lower in the medium, heavy and long-term smokers than in the non-smokers (P < 0.01). The sperm density, viability and forward progression, and the seminal plasma Zn, Cu and SOD levels were negatively correlated with the amount and duration of cigarette smoking (P < 0.01). CONCLUSION: Medium, heavy and long-term smoking adversely affected the semen quality in a population of men visiting the infertility clinic in Shandong, China.

[Studies of multidrug resistance gene expression and apoptosis in stomach carcinoma].

OBJECTIVE: To investigate the influence of multidrug resistance gene (MDR) expression and apoptosis on stomach cancer. METHODS: A total of 80 cases of paraffin embedded stomach cancer tissues was stained immunohistochemically for P-gp, GST-pi and TOPO II using monoclonal antibodies and apoptosis (in terms of apoptotic index, AI) in tissues was musured by the TUNEL method. The results were correlated with tumor type, clinical stage and prognosis. RESULTS: Normal stomach tissue expression of P-gp was associated with better survival, but higher level of P-gp expression in tumor tissues was associated with poor prognosis. The expression of GST-pi didnot show a relationship with cell differentiation of stomach cancer and clinical stage of the disease, but positive correlation did exist between intensity of expression and survival time. Although the expression of TOPO II was associated with tumor cell differentiation, it did not affect prognosis. Apoptosis was demonstrated in 73% of the cases and was related to clinical stage of stomach carcinoma. CONCLUSION: While P-gp expression in tumor cells is indicative of poor prognosis, its expression in normal tissue has the opposite effect. Positive correlation exits between GST-pi expression intensity in tumor cells and survival. MDR and apoptosis are related to the biologic behavior of the tumor.

[Experimental study on an animal model of axonal form Guillain-Barre syndrome].

OBJECTIVE: To investigate an animal model of axonal form Guillain-Barre syndrome (GBS). METHODS: A strain of Campylobacter jejuni (Cj PENO: 19) from stools of a patient with axonal form GBS was isolated. After the lypopolysaccharides (LPS) were extracted from the strain, they were inoculated into 26 rabbits, 10 micrograms per week. RESULTS: Three rabbits showed paralysis, with an incidence rate of 11.54%. For the three rabbits, LPS was inoculated two, three and three times, respectively. Pathological examination showed serious axonal degeneration in the sciatic nerve. The electrophysiological features showed motor axon damage. CONCLUSION: The results suggested the presence of an axonal form GBS. LPS in Cj could play an important role in causing axonal form GBS.

[Study on alkaline hydrolysis of polyglutamate for de-esterification].

This paper reports the alkaline hydrolysis of poly(methyl glutamate), poly (benzyl glutamate) and copoly(methyl glutamate-benzyl glutamate-glutamic acid) for de-esterification. The results showed that in this process of alkaline hydrolysis the demethylation of poly(methyl glutamate) was faster, but debenzylation of poly(benzyl glutamate) was hardly processed. An increase in the methyl glutamate content of starting copolymers, in the alkaline concentration and the time of alkaline hydrolysis, and a decrease in the film thickness, would lead to a raise in the degree of alkaline hydrolysis, that is, an increase of the glutamic acid segment content in product of alkaline hydrolysis.

[Alteration of banked blood cholinesterase level and its significance in emergency treatment of acute organophosphorus pesticide poisoning].

OBJECTIVE: To observe the relationship between the preservation days of banked blood and the alteration level of plasma cholinesterase with the aim of making proper selection of banked blood in emergency treatment of acute organophosphorus pesticide poisoning (AOPP). METHODS: We selected at random the banked blood that has been stored for different length of time before blood transfusion so as to determine the plasma cholinesterase value. The normal plasma cholinesterase value was determined on blood samples in the Blood Center which served as a control group. The cholinesterase value was determined with a kit of the BM Company and the Cobes-Fara II automatic analysor of the Roch Company. RESULTS: It was found that there was no significant difference of plasma cholinesterase value between the one-day banked blood and the normal control group (P > 0.05), but the plasma cholinesterase values of the other experiment groups were all significantly lower than that of the control group (P < 0.05). As compared with the normal control group, the relative ratio with fixed base was about from 80 percent to 16 percent. It seems that the preservation days are in negative correlation with the cholinesterase value (r = -0.7929, P < 0.01). The curvilinear regression equation is Y = -1,823.3 Ln(X) +/- 6,229.4. CONCLUSION: With the increase of the banked blood preservation days after blood sampling, the plasma cholinesterase value decreased gradually. So in the emergency treatment of severe AOPP, it is essential to use the fresh blood collected within one day, so as to avoid missing the chance of first-aid treatment. This study provides the experimental basis for emphasizing transfusion of fresh blood and ensuring transfusion of highly active cholinesterase in treating AOPP.

[Preparation of hepatitis C virus nonstructural region antigens and detection of their relative antibodies].

OBJECTIVE: To investigate the relationship between HCV NS3 and the pathogenesis of HCV and research a method to assess the therapeutic effect of interferon (lEN) in clinic. METHODS: In a retrospective research, we used HCV-NS3 antigens of different terminals expressed in E. coli which contained the recombinant plasmids involving amino terminal region (N') or carboxyl terminal region (C') of HCV-NS3, to detect the antibodies by Western blot in sera of 63 hepatitis C patients who were treated with IFN. RESULTS: It proved that antibodies against HCV-NS3 N' and/or HCV NS3 C' terminals in hepatitis C patients may predict the treatment result of IFN, especially in the patients with chronic hepatitis C. CONCLUSIONS: The detection of antibodies against HCV-NS3 N' and/or C' terminals may be used for evaluation of IFN treatment, especially in chronic hepatitis C ones.

Protective effects of Ginkgo biloba extract on gastric mucosa.

AIM: To study the protective effects of Ginkgo biloba extract (GbE) on gastric mucosa. METHODS: By means of restaint-cold stress (RCS) in rats and 100% ethanol gavage in mice, the index of gastric mucosal injury was evaluated. The gastric juice was collected using pyloric ligation, and the volume and acidity of juice, and activity of pepsin were determined. The content of malondialdehyde (MDA) was measured by thiobarbituric acid (TBA) method. RESULTS: GbE (25, 50, and 100 mg/kg, bid x 5 d, ig) inhibited dose-dependently the gastric mucosal injury induced by RCS and 100% ethanol gavage. The index of gastric mucosal injury after RCS in groups pretreated with GbE was 58%, 43%, and 31% of control group respectively. The index of gastric mucosal injury induced by ethanol in groups pretreated with GbE was 62%, 36%, and 26% of the control group, respectively. And GbE enhanced the protective effects of cimetidine (Cim) on gastric mucosa. But it did not obviously influence the volume and acidity of gastric juice as well as the activity of pepsin. One hour after the administration of ig 100% ethanol, the contents of MDA in gastric mucosa and serum in mice increased (P < 0.01) vs the control group. But pretreatment with GbE (25, 50, and 100 mg/kg, ig) could inhibit this increase of MDA both in gastric mucosa and in serum. CONCLUSION: GbE had protective effects on gastric mucosa and GbE plus Cim possessed the synergism in the treatment of acute gastric mucosal lesions.

[Effect of different nitrogen forms on growth of Fraxinus mandshurica seedlings].

The effect of NO3(-)-N and NH4(+)-N and their different supply ratios on the growth of Fraxinus mandshurica seedlings was studied by sand culture in greenhouse. The highest seedling growth was obtained at a NH3(-)-N/NH4(+)-N ratio of 75/25 in nutrient solution. The growth of seedlings declined with the increasing ratio of NH4(+)-N to NO3(-)-N. Excessive NH4(+)-N inhibited root growth, and the root/shoot ratio of the seedlings significantly decreased as the NH4(+)-N/NO3(-)-N ratio was over 50%. Increasing NH4(+)-N in nutrient solution led to the decrease of net photosynthetic rate, the increase of phosphorus content in plant tissues, and the decrease of Zn and Fe contents. The pH value of cultural medium was affected markedly by nitrogen form, which was increased with increasing NO3(-)-N and decreased with increasing NH4(+)-N in nutrient solution. The pH value was up to 7.22 in the medium contained NO3(-)-N alone, while decreased to 4.20 only with NH4(+)-N.

Polymorphisms of TAP, LMP and HLA-DM genes in the Chinese.

OBJECTIVE: To investigate the distribution of antigen-processing genes transporter associated with antigen processing (TAP), low molecular weight proteasome (LMP2) and HLA-DM in the Shanghai population and to explore the possible role of these genes in the genetic susceptibility to autoimmune diseases IgA nephropathy(IgAN) and multiple sclerosis(MS). METHODS: Eighty normal Shanghai Chinese, 60 patients with IgAN and 21 patients with MS were randomly selected and genotyped for antigen-processing genes (TAP1, TAP2, LMP2, HLA-DMA and -DMB) using polymerase chain reaction-sequence specific oligonucleotide (PCR-SSO) and polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Two-locus linkage disequilibrium analysis was performed. RESULTS: Four TAP1, 6 TAP2, 2 LMP2, 3 DMA and 4 DMB alleles were observed. Statistically significant associations were found between TAP1B-TAP2A, TAP1B-LMP2H and TAP2D-HLA-DMA* 0101 (Pc < 0.01). There was no association between antigen-processing genes and either IgAN or MS. CONCLUSION: The polymorphisms of antigen-processing genes in this Shanghai population are similar to those observed in the other ethnic populations. No association was found between IgAN or MS and any antigen-processing genes tested in Shanghai population.

Macrophage activation of lymphoma-bearing mice by liposome-mediated intraperitoneal IL-2 and IL-6 gene therapy.

OBJECTIVE: To investigate the antitumor mechanism of interleukin-2 (IL-2) and interleukin-6 (IL-6) gene therapy. METHODS: Liposome encapsulated IL-2 DNA and IL-6 DNA were intraperitoneally (i.p.) injected into mouse lymphoma cell line (EL-4) lymphoma-bearing mice. Macrophage function (M phi) from the mice was assessed. RESULTS: Cytotoxicity, major histocompatibility (MHC) II expression and IL-1 and TNF secretion of the macrophages all augmented after i.p. injection of liposome encapsulated IL-2 DNA or IL-6 DNA. More efficient activation of macrophages was observed in mice treated with liposome encapsulated IL-2 DNA than IL-6 DNA. IL-2 gene therapy combined with IL-6 gene therapy showed the maximal activation of macrophages in the lymphoma-bearing mice. CONCLUSION: IL-2 and IL-6 gene therapy can relieve the suppression of macrophages of the lymphoma-bearing mice, and efficiently activate the antitumor immune responses.

[The study of T helper cell subsets and relationship to relative cytokines and ventilatory function in asthmatic patients].

OBJECTIVE: To investigate the changes of T helper cell(TH) subsets and relationship to the relative cytokines, serum total IgE, eosinophil count and ventilatory function in asthmatic patients. METHODS: 20 asthmatic patients and 10 healthy subjects were selected. The counts of the TH subsets, the level of IL-4 and IFN-gamma in the supernatant of peripheral blood mononuclear cell(PBMC), serum total IgE concentration and eosinophil count in peripheral blood were determined in acute attack and stable patients with asthma and healthy subjects. RESULTS: The TH2/TH1 ratio in the acute attack and the stable patients with asthma were increased significantly, compared with that in normal group (P < 0.01). The relation analysis showed that the value of TH1 had significantly positive relation with that of IFN-gamma, the value of TH2 had significantly positive relation with that of IL-4. The total serum IgE had significantly positive relation with the level of IL-4. FEV1 had significantly positive relation with that of TH1, negative relation with that of TH2, and TH2/TH1 significantly. The total serum IgE and the number of eosinophils had significantly positive relation with that of TH2. CONCLUSION: The balance of TH in asthmatic patients deviates significantly. The deviation of balance of TH has relation with severity of asthma.

[Correlation of asymptomatic bronchial hyperresponsiveness with serum specific IgE and eosinophilic cation protein].

OBJECTIVE: To explore the correlation of asymptomatic bronchial hyperresponsiveness with serum specific IgE(sIgE) and eosinophilic cation protein(ECP). METHODS: Histamine inhalation provoking test (quantitative method) was used in a survey for bronchial responsiveress in 1,123 students aged between 11 to 14 years old, 41 individuals with asymptomatic bronchial hyperresponsiveness received further test for sIgE and ECP. 31 students with normal bronchial responsiveness were randomly selected as the control. RESULTS: 4%(41/1,123) of the students showed bronchial hyperresponsiveness. The incidence of bronchial hyperresponsiveness with family history of asthma (11/14, 79%) was higher than that without a family history (30/1,109, 3%), with statistical significance (P < 0.005). The positive rates for sigE (90%, 37/41, P < 0.01) and ECP (17%, 7/41, P < 0.05), sIgE q value (14.3 +/- 12.1) micrograms/L, (P < 0.01) and ECP value (44.3 +/- 60.2) micrograms/L, (P < 0.01) in asymptomatic bronchial hyperresponsiveness group were higher than those of the control with statistical significance. His PD20-FEV1 was negatively related to aIgE q value (P < 0.05) and not related to ECP (P > 0.05). CONCLUSION: The asymptomatic bronchial hyperresponsiveness was closely related to allergy but not to ECP. Those with a family history of asthma, or with positive sIgE should be followed.

[Preoperative chemotherapy and operation for invasive Masaoke stage III and IV a thymoma].

OBJECTIVE: To assess the effect of preoperative chemotherapy on invasive thymoma. METHODS: Fourteen patients with invasive thymoma (12 cases in Masaoka stage III and 2 cases in stage IV a) were treated with 3-4 cycles of CAVP (cyclophosphamide 600 mg/m2 D1, adriamycin 30 mg/m2 or epi-adriamycin 40 mg/m2 D1, vincristine 0.6 mg/m2 D1 or vindestine 3 mg/m2 D1, D8, cisplatin 30 mg/m2 D1, 2, 3). Following chemotherapy, patients were operated within 1-3 months. In 10 patients, sternotomy was performed and in 4 patients, anterolateral thoracotomy was performed. Radiotherapy was given with a total dose of 50-60 Gy in all patients except in those who were pathologically in complete remission. The patients were followed up for 6 months to 3 years. RESULTS: After chemotherapy, complete response was observed in 5 patients (35.7%) and partial response in 9 patients (64.3%). Nine patients received radical tumor resection and 5 patients received partial resection. Histologic/examination of the surgical specimens showed fibrosis of the remnant thymus in 5 patients. All but two patients survived in the follow-up period. Patient died from distant metastases at 18 and 24 months after treatment, respectively. CONCLUSION: Preoperative chemotherapy helps increase the resectability of stage III and IV a invasive thymoma. A longer follow-up period and more patients are needed to ascertain the impact of this treatment strategy on long-term survival.
