[Effect of inflammatory factor and yishen huoxue xiezhuo decoction on growth of glomerular mesangial cells in rats].

OBJECTIVE: To explore the part of inflammatory factors (cytokines) and Yishen Huoxue Xiezhuo (YSHXXZ) Decoction on the proliferation of extracorporeal cultured mesangial cells (MCs) in rats. METHODS: The effects of lipopolysaccharide (LPS) and interleukin-6 (IL-6) as stimulating factors, its action on the proliferation of rat MCs were investigated by using the technique of 3H-TdR incorporation. Meanwhile, adopting serum pharmacology assay, the medicated serum of rat, containing YSHXXZ decoction was extracted and its effects on the growth of MCs were also studied. RESULTS: LPS and IL-6, in a dose-dependent and time-dependent manner, could markedly stimulate the MCS proliferation, while this stimulatory effects could be strongly antagonized by the serum containing YSHXXZ decoction. CONCLUSION: Mesangial cell is the main target cell of the action of YSHXXZ decoction, and the inhibition on MCs might be one of the mechanisms of the YSHXXZ decoction in preventing the progression of chronic glomerulonephritis.

[The relationship between bone mineral density and polymorphism of the estrogen receptor gene in Chinese healthy menopausal women].

OBJECTIVE: To investigate the possible relationship between bone mineral density (BMD) and polymorphism of the estrogen receptor (ER) gene in Chinese healthy postmenopausal women. METHODS: 237 unrelated healthy postmenopausal women were selected for bone mineral density determination with Dual energy X-ray absorptiometry (DEXA) and the polymorphism of estrogen receptor gene were detected with polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). RESULTS: It is shown that the distribution of ER genotype in the ER gene was as follows: PP 19.8%, Pp 44.3% and pp 35.9% respectively. ANOVA analysis indicated that BMD of trochanter major (Troch) was associated with polymorphism of ER gene (P = 0.0106). Multiple stepwise regression analysis showed that the relationship between polymorphism of ER gene and BMD of Troch (P = 0.0548) and lumbar vertebra 2 approximately 4 (P = 0.0998) was not statistically significant. CONCLUSION: It is suggested that there is a significant relationship between polymorphism of ER gene and Troch BMD in healthy postmenopausal women. p allele may be a protective factor for bone mass loss.

[Assessment of erythrocyte pyrimidine 5'-nucleotidase test for screening of occupational lead poisoning].

OBJECTIVE: To determine the diagnostic value of red cell P5'-N for detecting occupational lead poisoning. METHODS: Red cell pyrimidine 5;-nucleotidase, blood lead and urine lead levels, free erythrocyte protoporphyrin (FEP), zinc protoporphyrin (ZPP) and urine delta-aminolevulinic acid levels were determined in 157 lead exposed workers and 30 non-exposed healthy people. According to the golden standard; i.e. blood lead >/= 1.93 micromol/L and >/= 2.90 micromol/L, the diagnostic value of red cell P5'N and other indicators for detecting blood lead >/= 1.93 micromol/L and >/= 2.90 micromol/L was compared by receiver operative characteristic curve (ROC) software. After determining the optimal cut-points of the above indicators for detecting the blood lead >/= 1.93 micromol/L and >/= 2.90 micromol/L, the sensitivity of the parallel tests for detecting the blood lead >/= 1.93 micromol/L and >/= 2.90 micromol/L was calculated and compared by EPINFOR software. RESULTS: For detecting blood lead level >/= 1.93 micromol/L, the areas under ROC curve (AUC(ROC)) of P5'N was significantly larger than that of urinary lead and urinary delta-ALA (P < 0.05). The sensitivity of parallel tests with ZPP and P5'N was significantly larger than that of parallel tests with Upb and Udelta-ALA. For detecting blood lead level >/= 2.90 micromol/L, the areas under ROC curve (AUC(ROC))of P5'N was significantly larger than that of urinary lead, FEP and urinary delta-ALA (P < 0.05). The sensitivity of parallel tests with P5'N and ZPP was significantly larger than that of parallel tests with Upb and Udelta-ALA, and that of parallel tests with ZPP and FEP (P < 0.05). CONCLUSION: For detecting blood lead >/= 1.93 micromol/L and >/= 2.90 micromol/L, red cell P5'N is more accurate. It is worth being applied in detecting occupational lead poisoning. For detecting blood lead >/= 1.93 micromol/L and >/= 2.90 micromol/L, the parallel tests with P5'N and ZPP are more accurate.

[Endovascular embolization treatment of intracranial aneurysms].

OBJECTIVE: To evaluate the effect of endovascular embolization of intracranial aneurysms with detachable coil and detachable balloon and emphasize the mainpoint of embolic technique. METHODS: 194 patients underwent complete cerebral angiography using microcatheter under DSA imaging. 80 cases with 90 aneurysms were emboliced by MDS, 69 cases with 71 aneurysms were emboliced by GDC. 37 of 40 giant aneurysms were emboliced by detachable balloon, 2 aneurysms by GDC, 1 by MDS. RESULTS: 194 patients with 201 aneurysms were cured successfully; 191 cases recovered clinically, 3 patients died. 187 aneurysms achieved 100% embolization, 8 with 95% embolization, 4 with 90% embolization, and 1 with 80% embolization. Four aneurysms reputured during the embolization, 4 patients had cerebral infarction, one patient died by hard-treated cerebral vasospasm which was affected by microcoil escape. Two recurrent cases were cured by second GDC embolization. CONCLUSIONS: The method of endovascular embolization to treat intracranial aneurysm is safe, reliable and effective. Those cases with giant aneurysms will have high recurrence in the future. Patients who suffer from SAH repeatedly may have great possibility of aneurysm rupture during embolization.

[Adenosine in treatment of rats with spinal cord injury].

OBJECTIVE: To observe the changes of endogenous extracellular adenosine after spinal cord injury (SCI) to rats and the effect of exogenous adenosine on extracellular calcium after SCI and post-injury neurological function. METHODS: A ventral compression injury model of T13 spinal cord was used, and the extracellular fluids were collected consecutively every 20 minutes after injury by using microdialysis. Adenosine in the samples was analyzed using high-pressure liquid chromatography (HPLC) with u.v. detection. The rats received different doses of 2-chloroadenosine (2-CADO), a nonspecific agonist of adenosine receptors, by intrathecal injection 15 minutes before injury. The extracellular fluid was collected every 10 minutes immediately after injury and the calcium was measured using atomic absorption spectrophotometer. Neurological function score, inclined plane angle, and histology were observed 24 hours after injury. RESULTS: A significant increase of adenosine was found immediately after spinal cord injury. The concentration of adenosine peaked at one hour after injury and dropped down to the basal level. There was a positive relation between the increase of adenosine and the severity of SCI. High dose of 2-CADO can significantly significantly inhibit the decrease of extracellular calcium and improve the neurological function of injured rats. CONCLUSIONS: Adenosine could involve the pathological process of secondary spinal cord injury and might play a protective role in SCI.

[Intraperitoneal lymphatic chemotherapy for colonic cancer].

OBJECTIVES: To evaluate the effect of intraperitoneal lymphatic chemotherapy with carboplatin and sandostatin on colonic carcinoma and abdominal lymphatic metastasis in patients after radical resection. METHODS: Patients with colonic cancer were divided into two group: abdominal lymphatic chemotherapy group and control group. The lymphatic chemotherapy group was treated at the introduction stage of anesthesia with carboplatin (200 mg) injected intraperitoneally and sandostatin (0.1 mg x 2) injected subcutaneously, whereas the control group was just treated with carboplatin (200 mg) injected intraperitoneally. The platinum levels in abdominal lymphatic nodes were monitored during operation. The two groups were uninterruptedly given intraperitoneal lymphatic chemotherapy or intraperitoneal chemotherapy. Retroperitoneal lymphatic metastasis was evaluated in each patient by abdominal CT scan after operation. RESULTS: The platinum levels in abdominal lymphatic nodes increased more significantly at 2, 3 hours in the lymphatic chemotherapy group than in the control group (P < 0.01). There was no difference in the platinum levels of the portal vein between the two groups, and also no difference in the platinum levels between the lymphatic nodes with or without metastasis. The rates of retroperitoneal lymphatic metastasis after resection were significantly lower in the lymphatic chemotherapy group control group (P < 0.01). CONCLUSION: Intraperitoneal administration of carboplatin combined with sandostatin may elevate the platinum levels in the abdominal lymphatic nodes and reduce the abdominal lymphatic metastasis after radical resection of colonic cancer. It is an effective method for intraperitoneal lymphatic chemotherapy.

[Measurement of gastric emptying with Xianshizi in patients with resected esophageal cancer].

OBJECTIVE: To determine gastric emptying radiological in patients with esophageal cancer and to obtain a similar effect produced by radionuclide study. METHODS: A new product-Xianshizi (XSZ) made of the mixture of barium and silica gel was used quantitatively to evaluate gastric emptying. 17 patients with resected esophageal cancer before and after operation, and 7 healthy volunteers were studied. RESULTS: Preoperative patients had obviously delayed gastric emptying compared with the control group (P < 0.01). Postoperative patients had more delayed gastric emptying compared with themselves (P < 0.05). CONCLUSIONS: Postoperative gastroparesis is related to delayed gastric emptying. The results of this study were identical with those obtained by scintigraphy. Compared with isotope studies, X-ray exam after taking meal composed of XSZ may eventually contribute to detecting delayed gastric emptying quantitatively.

[Study on risk factors of breast cancer among urban women in China].

OBJECTIVE: Breast cancer is the most common malignant neoplasm in women all over the world. It is necessary to establish monitoring methods and to identify risk factors. METHODS: Six cities including Beijing, Shanghai, Tianjin, Chongqing, Wuhan and Guangzhou were chosen as the sample areas. 2 100 cases with 1:1 matched controls were interviewed with a uniformed questionnaire. Conditional logistic regression and cluster analysis were used for multivariate analysis. RESULTS: The common risk factors for the combined data of the six cities were high educational level (OR: 1.17), high body mass index (OR: 1.51), high intake of milk products (OR: 1.49), high intake of eggs (OR: 1.14), psychological depression (OR: 2.78), history of benign breast diseases (OR: 2.24), old age at marriage (OR: 1.42), dysmenorrhea (OR: 1.22), short duration of lactation (OR: 1.17). Among these factors, different cities had different ORs, as high educational level (OR: 1.20 - 1.61), high body mass index (OR: 1.40 - 22.36), high intake of milk products (OR: 1.64 - 3.39), depression (OR: 1.60 - 10.50), history of benign breast diseases (OR: 1.73 - 19.44). Some statistically significant correlations existed in different cities such as early age at menarche, old age at marriage, long menstrual periods, high intake of meat, eggs, sea food and passive smoking were risk factors. Some risk factors varied between premenopausal and postmenopausal women. Beijing, Wuhan and Chongqing were clustered as the same group in cluster analysis, so did Shanghai and Tianjin, but Guangzhou could not be grouped with other cities. CONCLUSION: Some common risk factors for breast cancer did exist in the six cities, but there were varieties in different areas which showed different places would have their own characteristics. Thus, careful study of risk factors, especially in the selection of variables and data analysis need to be carried on.

[Geographic information systems spatial analysis on transmission of schistosomiasis in China].

OBJECTIVE: To understand the epidemiologic status and geographical distribution of schistosomiasis in China. METHODS: Relevant detabases were set up after collection of data from two National Sampling Surveys on Schistosomiasis, in 1989 and 1995. Spatial analysis was undertaken after the database linked to the GIS software which was supported by Arc View 3.0a. Correlation analysis was performed to understand the relationship between rates of human infection and cattle infection. RESULTS: The epidemic areas of schistosomiasis with high risk are mainly distributed in the marshland along the Yangtze River and can be identified as five spatial distribution regions based on the results of spatial analysis. Both epidemic areas and positive rates of stool examination in cattle and water buffalo are much wider and higher than that in humans. The positive correlation was seen between infection rate in human and in cattle from data of both sampling surveys. CONCLUSION: The relevant strategy for schistosomiasis control in different spatial regions should be performed accordingly and the measures of control for cattle and water buffalo should be strengthened in the endemic areas.

[Effect of chrysotile and cigarette smoking solution on DNA damage and repair in human alveolar epithelial cells].

OBJECTIVE: To study the effect of chrysotile and solution of cigarette smoke separately or combined on DNA damage and repair. METHODS: Single-cells gel electrophoresis assay was used to detect break in DNA strand and to analyze its repair function in human alveolar epithelial cell line A549, which were exposed to chrysotile or cigarette smoke solution separately or combined for one hour and then immediately cultured for different time. RESULTS: Break in DNA strands was induced obviously in A549 cells after exposure to 40 microg/ml of chrysotile or 2.5 x 10(-4) cig/ml of cigarettes smoke solution for one hour. Repair for DNA break could be seen clearly in chrysotile-treated cells after continuing culture for three hours, and 30.6% had been repaired after culture for four hours. Repair could also be seen clearly in cigarette smoke solution-treated cells after continuing culture for two hours, and 65.2% had been repaired after culture for four hours. Repair of DNA in cells exposed to both chrysotile and cigarette smoke solution combined was similar to that exposed to cigarette smoke solution only. CONCLUSION: Damage to DNA caused by chrysotile is more difficult to repair than that induced by cigarette smoking solution.

[Studies on chrysotile-induced cell transformation and its intervention effect].

OBJECTIVE: To explore the effects of several kinds of chemicals on chrysotile-induced cell transformation in human embryo lung (HEL) cells. METHODS: After being soaked in aluminum citrate, mixed rare earth or sodium selenite solutions for one hour, chrysotile was incubated with HEL cells. Then, the number of focus of cell transformation was observed. RESULTS: Chrysotile could cause morphological transformation of HEL cells in a dose-dependent pattern, with (0.60 +/- 0.55), (1.20 +/- 1.31), (2.60 +/- 1.14) and (3.60 +/- 1.14) foci of HEL cell transformation per plate after treatment with 0.625, 1.25, 2.5 and 5.0 microg/ml of chrysotile, respectively, and there was significant difference between the groups treated with 2.5 and 5.0 microg/ml of chrysotile and control group. It indicated that chrysotile could cause morphological transformation of HEL cells. Number of focus of HEL cell transformation induced by chrysotile that was pretreated with the above-mentioned three kinds of chemicals reduced significantly, as compared with that in the group without pretreatment of it. Its inhibitory rate was 50% in the group treated with mixed rare earth, with significant difference. CONCLUSION: Pretreatment for chrysotile with these three kinds of chemicals could reduce its carcinogenicity.

[Studies on cell transformation and cell cycle in human embryo lungs induced by nickel compounds].

OBJECTIVE: To elucidate the association between nickel compounds and occupational lung cancer. METHODS: Human embryo lung cells (MRC-9 and IMR-90) were transformed by water-insoluble nickel oxide and nickel subsulfide, and changes in cell cycle of these transformed cells were analyzed by flow cytometric technique. RESULTS: Morphological transformation in MRC-9 and IMR-90 cells were induced after exposure to nickel compounds for 3 - 5 months, including formation of transforming foci, overlaying growth and epithelium-like changes in cells, etc. Some transformed cells could form colonies in soft agar media. Changes in cell cycle of the transformed cells were observed with flow cytometry. Proportions of cells at G(1) phase and G(2)/M phase were 62.61% and 16.37%, respectively, in the untreated MRC-9 cells and hyperploid cells were hardly seen in cell cycle plots. But, in the transformed MRC-9 cells, proportion of cells at G(1) phase reduced to about 50%, and that at G(2)/M phase increased to over 20% and that of hyperploid cells to over 10%. The same phenomenon was observed in the IMR-90 cells. CONCLUSIONS: Water-insoluble nickel compounds can induce morphological transformation of human embryo lung cells and those transformed cells appeared certain biological characteristics of tumor cells.

[The relationship of cellular DNA content with clinical stage and biological features of colorectal cancer].

The purpose of this study was to explore the relationship of cellular DNA content with clinical stage and biological features of cancer. Flow cytometry was performed on fresh specimens from 86 patients from 1997 to 1998. Forty-five (53.3%) specimens were found to contain cells with abnormal DNA (DNA nondiploidy). Although none of the sex, age, site, differentiation variables correlated with flow cytometric DNA ploidy, nondiploidy was associated with Dukes' stage and lymph node metastasis. Duke's A stage tumors were more frequently diploid than were more advanced tumors, but no difference between Duke's B, C and D stages were observed. These findings suggest that ploidy is associated with some pathological factors.
