Analysis of protein antigens as a potential marker for Streptococcus pneumoniae.

Protein antigens of the five more common Streptococcus pneumoniae serotypes in Spain-1, 3, 4, 5 and 23 -were studied by crossed immunoelectrophoresis and sodium dodecyl sulfate polyacrylamide gel electrophoresis. Most protein antigens defined by crossed immunoelectrophoresis appeared to be common to the five serotypes studied since the degree of cross-reaction was high. Using gel electrophoresis a considerable degree of similarity in the patterns of protein bands was also observed. Thus, protein antigens would not appear to be epidemiological marker for typing pneumococci.

Staining of xylem parenchyma mitochondria with photo-oxidized 3,3'-diaminobenzidine.

A photo-oxidized solution of 3,3'-diaminobenzidine (DAB) is used to stain xylem parenchyma mitochondria in specimens prepared from lupin hypocotyls fixed with glutaraldehyde and osmium tetroxide and embedded in Epon. No other subcellular components, including plastids, nuclei, vacuoles or cell walls were stained when xylem parenchyma cells were exposed to this reagent for 1 hr. This reaction was stable for 20 min at 80 C, inhibited by KCN, and insensible to 3-amino-1,2,4-triazole. The outstanding sensitivity of this reaction to inhibition probes suggests that this stain is analogous to the previously described DAB/cytochrome c/cytochrome oxidase reaction in plant mitochondria, although the incubation of lupin sections with freshly prepared DAB solution (free of auto-oxidized DAB) did not result in staining. These results draw attention to the unreliability of DAB oxidation for demonstrating electron transport in plant mitochondria. However, we do recommend photo-oxidized DAB as a direct ultrastructural stain for plant mitochondria without reference to its oxidative capacity.

Polyvalent inhibitory action of fusidic acid in isolated rat liver cells.

The steroidic antibiotic fusidic acid showed a polyvalent action on isolated rat liver cells. It displayed a strong inhibitory capability on protein synthesis in intact cells even stronger than that previously reported in cell-free extracts. Also, it inhibited basal gluconeogenesis and promoted an increase of the membrane permeability to trypan blue. However, the effects on both protein synthesis and basal gluconeogenesis were observed at doses smaller than those required to reduce the cell viability.

[Chemical composition and biological value of tortillas and bread produced on the commercial level in Costa Rica].

A study was conducted to evaluate the nutritive value of tortilla, wheat bread, and tortilla meal. Six types of tortillas, five breads and two tortillas meal were thus studied. Chemical composition of each product was determined, and biological trials in laboratory rats were also carried out with each of them. Results demonstrated that the corn products had a better protein quality than the wheat products. Furthermore, little variability in nutrient content and protein quality (NPR) between the different corn and wheat products was found. The utilizable protein content of the wheat products was higher than that of the corn products, while the gross energy content was similar for both. An economical-nutritional analysis revealed that the corn-soybean tortilla meal is the most nutritive and economic product to provide basic nutrients (protein and energy) to the population.

Isolation and partial characterization of a protein fraction from the opossum (Didelphis marsupialis) serum, with protecting property against the Bothrops jararaca snake venom.

Two separated methods were used to purify a fraction from the opossum (Didelphis marsupialis) serum able to protect mice against Bothrops jararaca venom. The first of them included an initial batch DEAE-Cellulose ion-exchange of the serum, followed by another ion-exchange chromatography on a Carboxymethyl Sepharose column. The second method was a column ion-exchange chromatography on DEAE-Sephacel. These techniques allowed to obtain a protein fraction which resulted homogeneous in cellulose acetate and conventional polyacrylamide gel electrophoresis. The obtained protein fraction proved to be a glycoprotein according to the positive staining with periodic acid Schiff. Sodium dodecylsulfate polyacrylamide gel electrophoresis of the B-mercaptoethanol-reduced fraction showed heterogeneity and allowed to estimate molecular weights in the range of 42,000 to 58,000 daltons. The obtained serum fraction could effectively block the lethal effect of B. jararaca venom when jointly injected to laboratory mice by peritoneal route.

Alkaline phosphatase, gamma-glutamyl transferase, lactate dehydrogenase and creatine kinase in bronchial aspirate from neoplastic and normal pulmonary tissue.

The activities of 4 enzymes, i.e. alkaline phosphatase, gamma-glutamyl transferase, lactate dehydrogenase and creatine kinase were studied in bronchial aspirates and serums from two groups of subjects, the first one was composed of 14 subjects without active bronchopulmonary pathology and the other of 20 patients with lung cancer. The results showed a statistically significant decrease of the activities of alkaline phosphatase and beta-glutamyl transferase in bronchial aspirate from patients with bronchogenic malignant tumors in relation to normal subjects. This finding could be explained by the 'fetalism' principle, which states that the quantitative pattern of enzymes of immature human tissues resembles those of neoplastic tissues.

[Serum LDH and myeloma. Correlation with the degree of bone marrow infiltration].

The authors study the serum lactate dehydrogenase (LDH) activity in multiple myeloma patients in relation to survival, plasma cells bone marrow infiltration, and creatinine and paraprotein serum values. Twenty five patients seen between 1979 and 1983 were studied considering two groups, one with LDH values higher than 100 IU/1 and the other with LDH values lower than 100 IU/1. Patients having more than 100 IU/1 showed a more marked bone marrow infiltration. Paraprotein concentration was higher in patients having less than 100 IU/1. No correlation was observed for the other factors considered.

Lack of induction of serum gamma-glutamyltransferase in diabetes mellitus.

Some authors have suggested that carbohydrates can induce hepatic microsomal enzymes, resulting in increased serum gamma-glutamyltransferase (GGT) in diabetes mellitus. Previously we demonstrated the lack of serum GGT increases in patients with acute diabetic crises. In this work we studied serum GGT activity, blood glucose levels and glycosylated hemoglobin levels (HB A1) in 35 patients with diabetes mellitus and 27 healthy volunteers. We did not see differences in the serum GGT activity among controlled (25.05 +/- 2.72 U/l) and uncontrolled (26.44 +/- 4.05 U/l) diabetics and the control group (22.51 +/- 2.95 U/l). Also, there was no significant correlation between serum GGT and HB A1 levels in diabetic patients (r = 0.279). We think that our observations may be relevant because they support the hypothesis that hyperglycemia does not act as an enzyme-inducing agent in chronically uncontrolled diabetics and, furthermore, they indicate that in the presence of abnormal serum GGT levels in diabetics it is necessary to investigate other associated diseases.
