[External erythromycin therapy of acne].

24 patients suffering from papulo-pustular acne were treated with mono-therapy with 2% erythromycin-containing alcoholic solution for eight weeks. Pustular lesions responded promptly, followed by inflammatory papules. Microbiological investigations revealed a lack of propionibacteria as well as reduction of pathogenic staphylococci. Patients after extended local erythromycin treatment did not show any erythromycin serum levels.

Tomographic gallium-67 citrate scanning. Useful new surveillance for metastatic melanoma.

Conventional gallium scans are not useful to evaluate patients with metastatic melanoma. We evaluated a new method of tomographic gallium imaging. One hundred fourteen tomographic scans were obtained in a prospective surveillance study of 67 patients over a 3-year period. Scans were evaluated and compared to findings of independent clinical evaluations. Sensitivity of gallium identification of tumor involving peripheral lymph nodes and soft tissues, abdomen, mediastinum, and osseous sites was 68% to 100%; overall sensitivity of this technique is 82% with specificity of 99% in 570 organ system assessments. Analysis of discordant findings when a site was clinically occult but gallium-positive showed gallium uptake to be true-positive in six of seven lymphatic sites, three of three lung and mediastinal sites, six of six abdominal sites, but in no brain or bone sites. Gallium lesions identified by computed tomographic scans proved to be false-positive at one lymphatic and one bone site, and false-negative at four otherwise clinically evident lymph node and soft tissue sites, seven pulmonary sites, and four brain sites. Gallium tomographic scanning provides a composite assessment of melanoma and may eliminate the need for other studies.

Lecithin:cholesterol acyltransferase activity and HDL composition in serum of patients with kidney transplants.

In comparison with a control group, lecithin:cholesterol acyltransferase activities and HDL-cholesterol concentrations in serum of patients with kidney transplants were unchanged, while apolipoprotein A and HDL-triglyceride concentrations were increased. This constellation shows that the cholesterol metabolism in patients with kidney transplants differs from that of patients undergoing chronic haemodialysis.

[Surgical treatment of the elbow joint in chronic polyarthritis].

Compared to the frequency of manifestation of rheumatoid arthritis in the elbow joint the quota of surgical intervention in this area is low. In most cases pain and disability are only moderate, especially if the reduced range of motion is compensated by maintained function of the equilateral shoulder and wrist joint. Surgical treatment is indicated if instability increases, the independency of the patients in daily life is endangered and inflammation increases acutely. A persistent compression of n. ulnaris has to be operated as soon as possible. Often the preferred early synovectomy is impossible because of priority of other joints. By late synovectomy, however, good results can be expected regarding stability, function and extent of pain. In our cases the joint was opened from radial and ulnar in combination with ventralization of the ulnaris nerve. The replacement of the head of radius by a silastic-spacer seems to reduce the secondary irritation in the radial part of the joint and to increase stability. The own method for resection-arthroplasty in modification of the method of Hass with interposition of the middle part of triceps tendon is described and discussed. Because of imminent loosening prostheses are implanted very rarely.

Abnormalities in the composition of serum high density lipoprotein in renal transplant recipients.

Levels of HDL-cholesterol, HDL-triglyceride and apolipoprotein A were determined in the sera of 26 female and 44 male renal transplant recipients. In comparison with controls, no significant abnormalities in HDL-cholesterol values could be found, but apolipoprotein A and HDL-triglyceride concentrations, were increased significantly. The increased apolipoprotein A/HDL-cholesterol ratio and the decreased apolipoprotein A/HDL-triglyceride and HDL-cholesterol/HDL-triglyceride ratios in these patients demonstrate abnormalities in HDL-composition. These results permit the conclusion that, in renal transplant recipients, the levels of HDL-cholesterol and particularly the changes in HDL-composition should be taken into account when considering the reason for the high incidence of cardiovascular disease in these patients.

Cloned fragments of human adenovirus type-12 DNA.

The following restriction endonuclease fragments of human adenovirus type 12 (Ad12) DNA have been cloned in plasmid or bacteriophage lambda vectors using standard protocols: the EcoRI-A*, -B, -D, -E, and -F fragments, the BamHI-B, -C, -D, -F, -G, -H, and -I fragments, the HindIII-F and -I fragments, and the PstI-A, -D, -F, -G, and -H fragments. The EcoRI-A* fragment comprises the right terminal 5 kb of Ad12 DNA including the terminal 143 bp.

Integration of adenovirus type 2 DNA at a limited number of cellular sites in productively infected cells.

Several experimental findings supported the notion that adenovirus type 2 DNA is covalently linked to cellular DNA in productively infected human cells. Although the significance of this linkage for the process of viral replication is unknown, the recombination of viral DNA with host DNA may simply reflect the efficacy of the recombination systems in mammalian cells. In this report, evidence is presented for the existence of selective sites of recombination between viral and host DNAs. These sites are presumably located in repetitive sequences of cellular DNA. All parts of the viral genome appear to be represented in the recombinant form.

DNA methylation and viral gene expression in adenovirus-transformed and -infected cells.

The level of DNA methylation in adenovirus type 2 (Ad2) and type 12 (Ad12) DNA was determined by comparing the cleavage patterns generated by the isoschizomeric restriction enzymes HpaII and MspI. As previously reported virion DNA of Ad2 and Ad12 is not methylated. Parental or newly synthesized Ad2 DNA in productively infected human KB or HEK cells is not methylated either, nor is the integrated form of Ad2 DNA in productively infected cells. Hamster cells and Muntiacus muntjak cells are abortively infected by Ad12. We have not detected methylation of Ad12 DNA in hamster or Muntiacus muntjak cells. An inverse correlation between the level of methylation and the extent of expression of viral DNA in Ad12-transformed hamster cells has been described earlier. A similar relation has been found for the EcoRI fragment B of Ad2 DNA which is not methylated but is expressed as the Ad2 DNA-binding (72K) protein in the Ad2-transformed hamster line HE1. Conversely, the same segment is completely methylated in lines HE2 and HE3, and there is apparently no evidence for the expression of the 72K protein in these cell lines.
