Evaluation of an enzyme-linked immunosorbent assay for the detection of transmissible gastroenteritis virus antibodies.

An enzyme-linked immunosorbent assay (ELISA) using a detergent-solubilized antigen of purified virus was developed for detection of antibody against porcine transmissible gastroenteritis (TGE) virus in swine serum. The ELISA demonstrated antibody responses in pigs immunized intramuscularly with the attenuated TO-163 strain of TGE virus and in pigs orally infected with the virulent Shizuoka strain of the virus. The results of the ELISA were well correlated with those of the neutralization test. These results indicate the usefulness of the ELISA as a serological tool for TGE virus antibody.

Hemagglutination by pilus antigen 987P of enterotoxigenic Escherichia coli.

The hemagglutination (HA) by pilus antigen 987P of an enterotoxigenic Escherichia coli strain 987 was examined using fresh and glutaraldehyde (GA)-fixed erythrocytes (RBC) of various animals. Only when GA-fixed RBC was employed, a strain 987 exhibited striking HA activities. This was also demonstrated by using latex heads sensitized with the 987P antigen. The 987P-specific antiserum inhibited HA of strain 987 and 987P sensitized latex beads against GA-fixed RBC. We concluded that HA of strain 987 against GA-fixed RBC was specifically associated with the presence of 987P pilus antigen but do not exclude a possibility that adhesin is distinct from pili antigen.

Plaque assay of equine influenza virus.

ESK cells, a stable cell line derived from a swine embryo kidney, were found to be a good medium for plaque formation of the Prague and Miami strains of equine influenza virus. Factors influencing the plaque formation were investigated and a plaque assay for these viruses was worked out. The method is not only simple enough for routine use, but also is as sensitive as the egg inoculation method. The method was readily adapted for a neutralization test.

Comparative measurement of equine influenza virus antibodies in horse sera by single radial hemolysis, neutralization, and hemagglutination inhibition tests.

Single radial hemolysis (SRH), neutralization (NT), and hemagglutination inhibition (HI) tests were carried out on sera from horses immunized against the Prague and Miami strains of equine influenza virus. The HI and NT tests demonstrated good sensitivity; the sensitivity of the SRH test was somewhat lower. The NT titers of individual sera were correlated very closely with the HI titers, although the NT titers were higher. SRH zone diameters of individual sera also showed significant correlation with the NT and NI titers. The SRH test appears to be suitable for large-scale serological surveys and offers the advantages of rapidity and simplicity.

[Surgical treatment of the thoracic arterio-venous malformation with intraspinal and extraspinal niduses (author's transl)].

Recent advances in the technique of selective spinal angiography and microsurgery have provided us remarkable progress in diagnosis and treatment of spinal arteriovenous malformation. Although in some cases total extirpation is still difficult, various attempts have been performed for complete cure. In this paper, the authors reported a case of 6-year-old girl who complained of the progressive gait disturbance. In this case we found the thoracic AVM which had niduses not only in the intraspinal canal but also in the extraspinal space. Transthoracic ligature was performed in this case. That is, through a thoracotomy with rib resection on the right side, the enlarged anomalous feeding artery, i.e., intercostal artery with its branches, from which Adamkiewicz's artery should be derived was ligated and clipped. And the result was good. In Japan, we have never had a report of such surgical method for AVM. Besides such a case is very rare that the AVM had nidus extending to both intra- and extra-spinal canal space. So we discussed the treatment against such an AVM with the investigations of literatures. Further follow-up study for this case will be reported later. The paper was read by the authors at the 53rd Kyushu region meeting of Japan Neurological Society (February 28, 1976, Kagoshima).
