Immunohistochemical detection of mammary tumor virus antigens in sweat and sebaceous glands of mice.

Mammary tumor virus (MTV) antigens in both sexes of GRS/A, SHN and C3H mice were examined in the sweat and sebaceous glands by immunoperoxidase technique using antiserum against gp52, envelope protein, or p27, core protein. Balb/c mice were used for reciprocal foster nursing with these inbreds to discriminate the expression of endogenous MTV from that of exogenous MTV. Both antigens were first detected around the age of 4 months in the sweat glands of mice with endogenous GR- or SHN- MTV. A linear staining of gp52 was seen along the luminal borders of glandular cells, and the reaction products for gp52 were demonstrated on the apical cell membranes, where no virion could be seen ultrastructurally. A diffuse staining of p27 was found in the cytoplasm of some glandular cells, where MTV particles could not be detected. In the sebaceous gland of the same mice, however, only p27 was first detected at the age of 60 days. A dot-like staining of p27 was found in the perinuclear region of some glandular cells, where an aggregation of intracytoplasmic A particles could be seen under an electron microscope. These positive stainings were unrelated to sex. In such skin appendages of all examined C3H mice and Balb/c mice with GR- or SHN- MTV, no antigen expression could be seen up to the age of 500 days. Therefore, some genes might be able to regulate the expression of endogenous MTV antigens in the skin appendages, while their glandular cells would have no receptor for exogenous MTV, namely the so-called "milk factor".

Eosinophilic gastroenteritis. Immunohistochemical evidence for IgE mast cell-mediated allergy.

A surgical case of a middle-aged Japanese with eosinophilic gastroenteritis is described. This 50-year-old man presented protein-losing gastroenteropathy, iron-deficiency anemia, and marked elevation of serum IgE, in whom the lesion formed a diffuse thickening of almost the entire portion of the stomach. Microscopically, there was an intense infiltration of eosinophils throughout the whole thickness of the gastric wall, interspersed with plasma cells and lymphocytes. On the affected area, an increased number of IgE-positive plasma cells and surface IgE-positive mast cells, some of which are degranulated, were identified. These immunohistochemical findings provide a strong evidence for IgE mast cell-mediated allergy concerning this disorder.

Mechanism of growth enhancement of 7,12-dimethylbenz[a]-anthracene-induced mammary tumors in rats given high polyunsaturated fat diet.

The mechanism of growth enhancement of 7,12-dimethylbenz[a]anthracene (DMBA)-induced mammary tumors by high fat diet was investigated in 169 female Sprague-Dawley rats fed a semisynthetic diet containing 0, 8 or 40% corn oil by weight. The animals, most of which were intubated with 10 mg of DMBA twice during the 8th week of age, were fed an unsupplemented or fat-supplemented diet for 2 months from the 12th to the 21st week of age and finally killed during the 21st week of age. Compared to the rats fed a fat-free diet, a significantly increased yield of mammary adenocarcinoma with a shorter latency was observed in the rats fed a 40% corn oil diet. DNA synthesis of the biopsied mammary tumors more than 1 month after diet switch-over was estimated in terms of the rate of [3H]thymidine incorporation into the tissue. Significantly higher DNA synthesis of the mammary tumors biopsied from proestrous hosts was found, as compared with the tumors in similar hosts at other stages of the estrous cycle. The greatest enhancement of DNA synthesis in small and rapidly growing tumors was observed in the 40% corn oil diet group. Feeding with 40% corn oil diet always resulted in elevation of the weight percentage of linoleic acid in both simple lipids and phospholipids of the mammary tumors. These data indicate that the effects of dietary fat on mammary tumor growth might be mediated by the enhancement of tumor response to certain hormones, and that the enhanced responsiveness of tumors was associated with increased linoleic acid in tumor lipids.

[An autopsy case of malignant mesenchymoma of the diaphragm].

An autopsy case of a huge tumor in the right thorax is reported. A 71-year-old man complained of dyspnea and died after three months of hospitalization. The tumor, consisting of a circumscribed mass measuring 24 X 24 X 8 cm and weight 3.7 kg, originated from the right diaphragm and had no metastasis. Histologically, the tumor comprised multiple mesenchymal elements of fibrosarcoma, rhabdomyosarcoma, osteosarcoma and chondrosarcoma, and there was no distinct border between these different components. This tumor fulfills the criterion of malignant mesenchymoma (Stout). The histogenesis of the tumor is discussed with a survey of literature.

[An operative case of spermatic cord rhabdomyosarcoma--with a review of similar cases in Japan].

A 17-year-old Japanese boy was admitted with the complaint of painless, progressive enlargement of the left scrotal contents during the preceding two months. The tumor was firm and nontender without transillumination. Orchitectomy with high ligatation of the spermatic cord was performed. The tumor, weighing 13.1 g, was gray, solid, located at the distal part of the spermatic cord, and separated from the testis and epididymis. The pathological diagnosis showed left spermatic cord rhabdomyosarcoma of the combined embryonal and alveolar type. All of the retroperitoneal lymph nodes dissected were negative for metastasis. The patient was treated with actinomycin D and vincristin postoperatively, and is well three years after the operation. Ninety-two cases of intrascrotal rhabdomyosarcoma reported in Japan are reviewed.

Benign and malignant epithelial tumors of the rete testis in mice.

Two papillary cystadenomas and one papillary cystadenocarcinoma in the rete testis were found in two of 500 very old male JCL:ICR mice; the hosts had no other tumors. One mouse had bilateral cystic tumors. A 5 x 3 x 3 mm papillary cystadenoma was observed in the rete of the right testis. A 7 x 3 x 3 mm tumor with a mixed pattern of papillary cystadenocarcinoma and papillotubular adenocarcinoma was found in the rete of the left testis; the latter extended from the mediastinum to the ductuli efferents and the caput epididymis. The other mouse had a 9 x 5 x 5 mm papillary cystadenoma of borderline malignancy in the rete of the right testis. The mice had no clinical signs.

[Acute traumatic hydrocephalus].

We have experienced two rare cases of acute traumatic hydrocephalus which developed within several hours after head injury. Case 1. A 4-year-old boy was stuck by an automobile. After losing consciousness for a few minutes, he became agitated and vomited. He was brought to our hospital 3 hours after accident. On admission, he scored 8 points on the Glasgow Coma Scale. CT scan on admission disclosed slightly enlarged ventricles, but there were no findings indicating cerebral contusion nor intracranial hemorrhage. His state of consciousness gradually deteriorated, thereafter right sided convulsive seizures occurred. 7 hours after accident, CT scan was repeated and demonstrated a marked enlargement of the ventricles. Continuous ventricular drainage was performed. The ventricular fluid was slightly bloody. After the ventricular drainage, the state of consciousness improved rapidly. 6 days after the accident, ventricular drainage was discontinued. Afterward, size and shape of the ventricles became nearly normal and the patient was discharged 36 days after the head injury without any neurological deficits. Case 2. A 3-year-old girl was buried under a burden. 30 minutes after the accident, her consciousness began to deteriorate with vomiting. She was brought to our hospital 3 hours after the accident. On her arrival, she scored 13 points on Glasgow Coma Scale. CT scan, which performed 4.5 hours after the accident, showed slightly enlarged ventricles and intraventricular hemorrhage in the third and fourth ventricles. Her state of consciousness gradually worsened with generalized seizures. Repeated CT scan 8 hours after the accident, demonstrated a marked enlargement of the ventricles. Following ventricular drainage, the patient's state of consciousness improved rapidly.(ABSTRACT TRUNCATED AT 250 WORDS)

Immunohistochemical studies on carcinoembryonic antigen in adenocarcinomas of the uterus.

In order to distinguish endocervical adenocarcinoma from endometrial adenocarcinoma, an immunoperoxidase stain for carcinoembryonic antigen (CEA) was tried. All of 10 endocervical adenocarcinomas revealed CEA and an adenosquamous carcinoma in the uterine cervix also showed it, while a mesonephroid adenocarcinoma in the uterine cervix did not. The immunohistochemical reaction products for the antigen were not observed in the glandular structures of 20 endometrial adenocarcinomas, although CEA was detected in all foci of squamous epithelial metaplasia occurred within 7 endometrial adenocarcinomas. CEA was detected in the endocervical type of glandular epithelium within a special endometrial adenocarcinoma containing predominantly endocervical type glandular epithelium. The immunoperoxidase staining pattern for CEA in the endocervical adenocarcinoma was related to the degree of histological differentiation of tumors, as follows; in the well differentiated glandular structure CEA was located on the luminal surface, while it was detected in the whole cytoplasm of tumor cells within the moderately and poorly differentiated areas. In conclusion, the immunoperoxidase stain for CEA would be useful for estimating malignancy of glandular structures within the uterus, distinguishing endocervical adenocarcinoma from endometrial adenocarcinoma, and grading of histological differentiation of endocervical adenocarcinoma.

Ultracytochemical observations on intracytoplasmic lipids by enzymic digestive methods: histochemical trials.

The penetration of Rhizopus arrhizus lipase into ethionine fatty livers was examined histochemically. A free-floating, 30 microns thick Vibratome section of the livers perfused with aldehydes was shown to be suitable for ultracytochemical analysis by the lipase digestive method. Accessibility of phospholipids in the outer membrane of erythrocytes to Naja naja phospholipase A2 was observed under a light microscope, and intact red cells were used for the ultracytochemical analysis by the phospholipase digestive method. Ultracytochemical pictures of cytoplasmic lipids in these materials subjected to the enzymatic digestive method are presented.

Contiguous malignant astrocytoma and Wilms'-like tumor in the brain.

A case of primary brain tumor composed of two contiguous neoplasms in presented. At operation, a nodular tumor was embedded in the infiltrating tumor within the brain parenchyma. With light and electron microscopy, the nodular tumor was similar to Wilms' tumor (nephroblastoma). The infiltrating tumor was malignant astrocytoma. Autopsy revealed, besides the recurrence of malignant astrocytoma in the brain and its subarachnoid dissemination, extracranial metastases to the abdominal cavity, liver, lung, and bone marrow. Recurrent and metastatic tumors were glioblastoma multiforme, which was more malignant than the surgical specimen. The possibility of metastatic Wilms' tumor from the kidney was completely ruled out by extensive autopsy survey. The authors present an extremely rare tumor including detailed observations on the electron microscopic appearance of the tumor; the histogenesis of these tumors is briefly discussed.

Inhibition by early treatment with bromocriptine of spontaneous mammary tumour development in rats with no side-effects.

Temporary inhibition by CB-154 (bromocriptine) of pituitary prolactin secretion, which induced a decline in mammary gland DNA synthesis for 4-11 weeks of age, resulted in a marked suppression of spontaneous mammary tumour development in rats after one year of age. The treatment had no effect on reproduction and subsequent lactation immediately after the CB-154 injection or growth and function of the mammary glands and pituitary prolactin secretion in later life.

Histologic architecture of submucosal connective tissues in human eustachian tube with supplemental reference to the effects of aging.

Serial sections of 74 human eustachian tubes from 37 autopsy cases were examined microscopically to determine the histologic architecture of submucosal connective tissues. Abundant elastic fibers ran just beneath the mucosal epithelium of the eustachian tube. Two kinds of layers of collagenous fiber were observed an inner circular layer and an outer longitudinal layer. An "elastic fiber band" perfectly encircled the tubal lumen at the pharyngeal part of the eustachian tube. This band became less sharply defined at the osseous part. The two layers of collagenous fiber were contiguous, especially in the upper part of the tube, and ran from the pharyngeal orifice to the isthmus. Submucosal connective tissues were found to be undeveloped in fetal eustachian tubes. In children as well, elastic fibers were not fully developed, and circular banding of collagenous fibers was less marked. With advancing age, elastic fibers increased in number and became large and brittle. Both layers of collagenous fibers became thicker. The present data indicate that the submucosal connective tissues may play an important role in the elasticity of the eustachian tube itself and in the maintenance of the tubal framework. Gerontologic changes in the eustachian tube stroma have been considered pathophysiologically.

Rapid enlargement of ventricles within seven hours after head injury.

We report a case of acute enlargement of the ventricles within 7 hours after head injury that was documented by repeated computerized tomography. It is suggested that the pathophysiological mechanism for this rapid enlargement of ventricles may be due to the raised intracranial pressure (ICP) that results from obstruction of cerebrospinal fluid pathways by subarachnoid hemorrhage, in addition to the elevation of ICP aggravated by frequent focal convulsive seizures.

Prophylaxis of spontaneous mammary tumorigenesis by temporal inhibition of prolactin secretion in rats at young ages.

The effects of temporal prolactin suppression during the limited periods of early age on spontaneous mammary tumorigenesis at advanced ages were examined in rats. Daily s.c. injections of 0.5 mg 2-bromo-alpha-ergocryptine mesylate, a potent suppressor of pituitary prolactin secretion, into virgin rats for 7 weeks beginning at 4 weeks of age resulted in almost complete prevention of mammary tumor appearance by 20 months of age. The incidence of tumors in this group (Group 1) was 3.3% (1 of 30), significantly smaller than that of the control [Group 2, 47.6% (10 of 21)]. Treatment given between 11 and 18 weeks of age inhibited mammary tumor incidence to a lesser extent [Group 3, 20% (6 of 30)], although tumor incidence was still significantly smaller than that in the corresponding control [Group 4, 41.2% (7 of 17)]. Serum prolactin level was decreased significantly by 2-bromo-alpha-ergocryptine mesylate; the levels on the evening of proestrus in the last week of injection were 18 +/- 3 (S.E.), 326 +/- 37, 15 +/- 2, and 460 +/- 55 ng/ml in Groups 1, 2, 3, and 4, respectively. Body weight change and pattern of estrous cycles were not affected by the treatment.
