[Workers' subjective feeling of fatigue and attitudes towards work--effects of age and job difference].

The present study is an attempt to assess attitudes towards work and subjective feelings of fatigue. To discover the relationships between these factors, a field study was conducted in a large-sized electric company. The subjects were 1376 male workers. The results of the 30 questions concerning subjective feelings of fatigue published by the Japan Association of Industrial Health showed that the complaint rate of fatigue in younger workers (18-29 yrs.) was higher than that of older workers (30-66 yrs.). In the middle aged (44-49 yrs.), feelings of fatigue in the administrative group were lower than that of the non-administrative group. Finally, workers who had a high complaint rate of fatigue were less favourably disposed towards their work and felt an increase in boredom, loneliness and monotony. Overall, the results indicated that the difference of labor mode exerts an influence on the onset of self-reported stress.

Endodeoxyribonuclease from nuclei of bovine small intestinal mucosa: further studies on intranuclear localization and cleavage mechanism of the enzyme.

Most of the activity of endodeoxyribonuclease was extracted from isolated chromatin with buffer containing 0.6 M NaCl, indicating that the endonuclease is present as a chromatin-bound form. When nuclei or chromatin of calf thymus or rat liver was digested with the bovine nuclear enzyme in the presence of 2 mM EGTA, to suppress endogenous Ca, Mg-dependent DNase activity, discrete DNA bands with integral multiples of 200 base pairs in length were produced, but no acid-soluble nucleotide was detected. The enzyme made single-strand breaks in pBR322 DNA and degraded it to fragments of limited size. The size of the final products of DNA's of Micrococcus luteus, rat liver nuclei, and calf thymus nuclei was about 3,000, 200, and 160 base pairs, respectively, but the enzyme showed no base specificity. Thus the endonuclease seems preferentially to recognize AT-rich regions of double-stranded DNA and to make single-strand breaks.

Development of hand-eye dominance in relation to verbal self-regulation of motor behavior.

The development of the relation between hand-eye dominance and verbal self-control of motor behavior was examined with 4- and 5-year-olds. Performances of subjects falling into either crossed or consistent hand-eye dominance categories were compared on a two-choice button-pushing task. Children performed this with their dominant hand in one of two conditions: verbalizing and not verbalizing their button-pushing activity. Results showed that crossed hand-eye dominant children gave significantly poorer performances than consistent hand-eye dominant children and that the former used their self-instructions in a motoric manner. These findings were interpreted as supporting the view that the functions of cerebral hemispheres in children with crossed dominance are more immature than those in children with consistent dominance.

Purification and properties of a neutral endodeoxyribonuclease from rat small intestinal mucosa.

An endodeoxyribonuclease has been purified to near homogeneity from rat small intestinal mucosa by a procedure involving Con A-Sepharose affinity chromatography. During the initial steps of purification, the presence of 5 mM CaCl2 was essential for stability of the enzyme activity. The enzyme has a molecular weight of 32 000 and an isoelectric point of 4.7. NaCl, sulfhydryl reagents, and iodoacetate strongly inhibited the reaction, but tRNA did not. The enzyme required divalent cations for activity and had a pH optimum of pH 6.2 with Co2+ and pH 7.7 with Mn2+. In both optimum conditions, the enzyme hydrolyzed native DNA more rapidly than denatured DNA, and the average chain lengths of limit digestion products of native and denatured DNA were 8 and 10, respectively, at pH 6.2 and 9 and 11, respectively, at pH 7.7. The enzyme activity to produce acid-soluble fractions from linear DNA substrate was similar in the two optimum conditions, but the activity to nick double-stranded, superhelical circular DNA substrate was significantly higher at pH 6.2 than at pH 7.7. The endonuclease formed single-strand breaks making 5'-phosphoryl and 3'-hydroxyl termini, and deoxythymidine was present at the 5' termini with a frequency of about 50% in both optimum conditions. Bovine pancreatic DNase I antibody and G-action inhibited the enzyme activity. Thus this endonuclease is classified as a DNase I.

Nature of discriminating and categorizing functions of verbal labels on recognition memory for shape.

The purpose of this experiment was to investigate the hypothesis that the discriminating and categorizing functions of a label associated with an ambiguous shape can differentially influence delayed recognition memory for the shape. The temporal course of shape recognition was measured after the subjects were trained to associate the shapes with relevant verbal labels, irrelevant verbal labels, or no verbal labels. Either immediately or after delays of 15 min or 1 wk, subjects were given a shape recognition test and both free- and aided-recall tests for retention of the associated verbal labels. Results indicated that relevant verbal labels differentially affect memory for shape and that the discriminating function of the label operates effectively at the time of stimulus encoding. The categorizing function of the label affects the changes occurring during the storage stage of memory. These results are interpreted as supporting the view that the courses of shape recognition and relevant label recall are not independent.

Adverse affect of blood transfusions on survival of patients with gastric cancer.

The effect of perioperative blood transfusions on the survival rate of patients with gastric cancer was studied. The survival rate of the transfusion group was significantly lower than that of the nontransfusion group in each of the 5 postoperative years. When no adjuvant immunochemotherapy was performed postoperatively, the prognosis was definitely worse in the transfusion group than in the nontransfusion group. Furthermore, the survival rate of the transfusion group was lower than that of the nontransfusion group in both histopathologic classifications of gastric cancer, and it was lower to a statistically significant extent among the well-differentiated types. These results indicate that transfusions might adversely affect postoperative survival of patients with gastric cancer.
