Human yolk sac tumor serially transplanted in nude mice: its morphologic and functional properties.

Three human yolk sac tumors (TE, OE, and TT-1) were serially transplanted into nude mice. The histology of the transplanted tumors is moderately different from that of the original tumor; the transplants had mainly a tubular structure with an inconspicuous endodermal sinus structure due to a decreased mesenchymal element surrounding the vascular core. Ultrastructural features of the transplanted tumor cells, however, were identical to those of the original tumor. Immunochemical survey of the blood plasma of nude mice bearing these tumors demonstrated the production of adult type human plasma proteins including prealbumin, albumin (Alb), alpha 1-antitrypsin (alpha AT), transferrin (Tf), hemopexin, and alpha 1-fetoprotein (AFP). The types of adult plasma proteins that were produced varied, depending on the tumor line but were the same between serial passages within the same tumor line. The intracellular localization of AFP, alpha AT, Tf, and Alb was also investigated by immunoelectron microscopy. Plasma proteins of the adult type and AFP were localized mainly within the membrane systems of the endoplasmic reticulum and Golgi complex and occasionally on the microvilli. These results suggest that transplanted human yolk sac tumors have almost all of the protein-producing function possessed by their normal counterparts.

Purification and immunohistochemical localization of rat liver glutathione peroxidase.

Glutathione peroxidase was purified from the rat liver to give a single protein band in polyacrylamide gel electrophoresis. Rabbits were immunized with this purified enzyme, and a highly specific anti-glutathione peroxidase antiserum was obtained. Using this antibody, an immunohistochemical technique (the indirect method of peroxidase-labeled antibody) was applied to study the localization of the enzyme in the liver cells. On immunohistochemical observation, glutathione peroxidase was localized exclusively in the cytoplasm of hepatocytes, and a stronger 'immuno-staining' was exhibited in the peripheries of the hepatic lobules than in the central zone.

Immunoglobulins in cerebrospinal fluid--enzyme-immunoassay.

Using an enzyme-immunoassay (EIA) technique we established control values for IgA and IgM in cerebrospinal fluid (CSF). These values, together with IgG values determined by single radial immunodiffusion (SRID) technique, showed significant positive correlations with CSF total protein values. The CSF IgA and IgM levels were related to corresponding levels determined in serum. In addition, the values for all 3 immunoglobulin classes in CSF as well as CSF total protein values showed positive correlation with age of subjects, and IgG% and IgA% inreased with age. This new EIA procedure can be completed within 24 h and is sensitive enough for determining all 3 immunoglobulin classes using a small amount (100 microliter or less) of native CSF.

Testosterone-indued focal myocarditis in rats. II. Morphological and biochemical observations of the adrenal in relation to the pathogenetical mechanism of cardiac lesions.

Correlative morphological and biochemical studies were made on the adrenal of rats treated with daily subcutaneous injections of testosterone propionate in a dose of 50 mg/kg for 30 consecutive days. Histologically, there is a thinning of the zona fasciculata with a decrease of cell numbers, swelling of the cell body and the occurrence of large lipid droplets in the cytoplasm. Electron microscopic examination revealed swelling of mitochondria with reduction of cristae and hypertrophy of smooth-surfaced endoplasmic reticulum with cluster formation or whirl-like arrangement. In in vitro biochemical experiments, it was noted that conversion of deoxycorticosterone to corticosterone or 18-hydroxydeoxy-corticosterone was decreased to 45% of the control level in the testosterone-treated group.

Plasma protein production by human tumors xenotransplanted in nude mice.

For detection of plasma proteins produced by human malignant tumors, a survey of blood plasma obtained from nude mice bearing serially transplanted human tumors was performed by immunoelectrophoresis and the double immunodiffusion technique. Among 34 lines including 18 types of human tumors, human specific plasma proteins were demonstrated in the plasma of nude mice transplanted with two lines of renal cell carcinoma, one adenocarcinoma of the colon, and one squamous cell carcinoma of the maxillary sinus. These tumors can be designated as "ectopic" plasma protein-producing tumors since the organs or tissues from which they originated are not considered to be usual sites of plasma protein synthesis. Plasma protein production, as well as that of alpha1-fetoprotein, was also found in one line of hepatoblasotma and three lines of yolk sac tumors. The above tumors were shown to produce one or more of the following 10 of 20 plasma proteins examined: albumin, prealbumin, alpha1-antitrypsin, ceruloplasmin, alpha2-macroglobulin, hemopexin, haptoglobin, C3 and C4 component of complement, and transferrin. An immunochemical demonstration of human specific cancer products observed in human tumors xenotransplanted into nude mice may provide a new approach for investigating the metabolism of neoplastic cells.
