[Expression of intermediate filament proteins in MDCK cells cultured in the three-dimensional gel in the presence of cytokine HGF/SF].

An immunomorphologic study was made of expression of intermediate filament proteins--keratin and vimentin--in the structures formed in branching tubulogenesis of Madin-Darby Canine Kidney epithelial cells (MDCK, clone 20), cultured in three-dimensional collagen gel in the presence of the "scatter factor" (HGF/SF). An improved method of parallel immunostaining of the whole branching structures and of their cryostat cross-sections was used. It was shown that both the discoid epithelial cells, forming walls of tubules with inner clear spaces, and the fibroblast-like cells invading the collagenic gel coexpressed keratin and vimentin. Thus, in the studied system of simplified morphogenesis in vitro, some conservatism of intermediate filament protein expression was revealed which does not respond to changes in cell shape or cell localization.

[Dependence of keratin No. 17 distribution on the degree of malignancy of bladder tumors].

Keratin No 17 expression (Mab E3) correlated with the stage and degree of malignancy of transitional-cell bladder carcinoma (TCC). Antibodies to keratin No 17 stained only the basal layer of the tumour growth in the TCC I while in TCC II-III positively stained cells were found everywhere in the tumour and staining was diffuse. Immunostaining for the simple keratins No 7, 8, 18, 19 in TCC I-III was diffuse. Distribution of keratin No 17 as well as that of the simple keratins was not regular in the metaplastic and anaplastic forms of TCC. Thus, the expression of keratin No 17 and simple keratins may serve an objective criterion of the tumour progression stage.

[Monoclonal antibodies to the proteins of intermediate filaments and of basement membranes in the differential diagnosis of certain forms of human breast tumors].

Monoclonal antibodies to keratins No. 8 and 17 specific for lining epithelium and myoepithelium of the mammary gland, respectively, as well as to basement membrane laminin, entactin, collagen type IV and heparan sulfate proteoglycan were used to the immunohistochemical analysis of 77 benign and malignant human breast lesions and that of 38 cases in which an intraoperative biopsy diagnosis was difficult. Morphologically similar benign and malignant proliferations were distinguished by keratin expression. In benign lesions both keratins were present, while in malignant ones only keratin No. 8 was expressed. Basement membranes associated with a myoepithelial layer were intact in benign lesions and in situ structures, but they were absent around the vast majority of invasive tumor foci. Basement membrane loss was important in differential diagnosis of benign sclerosing adenosis and cystadenopapilloma from invasive tubular and papillary carcinoma, respectively. Diagnosis of microinvasion in ductal and lobular carcinoma was much easier when combination of antibodies to keratins and basement membrane proteins was used.

[The expression of keratins Nos. 8, 17 and vimentin in pleomorphic adenoma of the salivary glands].

In the normal salivary gland, the monoclonal antibody to keratin 8 immuno-morphologically identifies the epithelium that covers acini and ducts. The monoclonal antibodies to keratin 17 and vimentin detect normal myoepithelium. The two keratins are found both in the epithelioid and mesenchyma-like components of pleomorphic adenoma, suggesting a single epithelial nature of this tumor. In all the morphological components of the pleomorphic adenoma, there are cells that combine protein expression of intermediate filaments normally labelling different cell subpopulations. This fact provides support for the hypothesis that the pleomorphic adenoma originates from bipotent precursor cells. A particular phenotype of pleomorphic adenoma elements is realized under the control of the local density of cells and microenvironment, as a result of which the cells expressing vimentin predominate in the mesenchymal component, keratin 8 in the epithelial tubular one.

[Malignant myoepithelioma of the breast (histological, immunomorphological and electron microscopic study)].

A rare kind of a breast tumor was identified in a woman of 47. The disease ran a malignant course and presented difficulties for morphological verification. Histological appearance showed areas of spindle-shaped or lamellar cells occasionally encircled with homogeneous hyalin-like substance. Locally, there were structures of alveolar or glandular type with an irregular basal membrane and loose position of cells. The primary histological diagnosis suggested a tumor of mesenchymal origin. Followed by immunomorphologic (monoclonal antibody to KL 1) and electron-microscopic examinations, the diagnosis was verified providing evidence for epithelial nature of the tumor cells, loose, forming glandular-like structures and simulating stromal elements with ultrastructural features typical of myoepithelial differentiation. The evidence obtained justify the tumor definition as a rare neoplasm of the breast known in the literature as myoepithelioma.

[Isolation of monoclonal antibodies to horseradish peroxidase and their use in immunohistochemistry and immunoblotting].

Monoclonal anti-peroxidase antibodies (McAb) were generated by means of hybridization of BALB/c immune splenocytes with X-63.653 cells. Peroxidase-antiperoxidase (PAP) staining of fibroblast cultures with murine McAb against fibronectin was used for the detection of positive cultures. McAb AP-FC-2B4 were found to be highly efficient in different immunoenzyme tests, e.g., immunochemistry and immunoblotting. PAP method was used for histological diagnosis of invasive human breast cancer.

[Immunohistochemical research on human breast tumors using monoclonal antibodies to intermediate filament proteins. Cancer of the breast].

Immunomorphologic study of 29 breast cancer cases using monoclonal antibodies to proteins of intermediate filaments shown to differentiate the lining epithelium from myoepithelium in the non-proliferating epithelial structures of the mamma, has shown the cells in the majority of tumours (according to the International WHO Classification defined as infiltrating ductal, lobular, and tubular cancer forms) to contain prekeratin (PK) C12, specific for normal lining epithelium, but not for the myoepithelium. In cases of cancer with chondroid metaplasia (a malignant variant of the so-called "mixed tumour") the cells contained PK E3, vimentin and structural myosin, normally specific for myoepithelium. The cell heterogenicity in PK C12 content or its absence noted in the infiltrating cancers with predominance of a solid component can indicate a high degree of tumour anaplasia. It is concluded that usage of monoclonal antibodies to PK C12, invariably found in the cells of fibrotic invasion foci, can be a useful indicator for early diagnosis of infiltrative tumour growth.

[Immunomorphological research on human breast tumors using monoclonal antibodies to intermediate filament proteins. The proliferative epithelial structures in fibrocystic disease (dysplasia) and benign tumors].

Proliferating epithelial structures were studied immunomorphologically in 16 cases of fibrocystic disease and benign tumours. Monoclonal antibodies were used to prekeratin (PK) C12, normally specific for the lining epithelium, to prekeratin (PK) E3 and to the protein of intermediate filaments in mesenchymal cells--vimentin, normally specific for myoepithelium. The immunofluorescent analysis of the most common proliferating structures has shown that there are elements with a variable combination of PK C12, PK E3 and vimentin among the proliferating cells. While there are cells similar to normal lining epithelium (PK C12) or myoepithelium (PK E3) and/or vimentin), many cells have no analogues either among normal cells, or among cells from ductal, lobular and tubular tumour forms. Since in the most common forms of breast cancer only cells containing PK C12 were found, the immunofluorescent study with the application of monoclonal antibodies to PK C12, PK E3 and vimentin can be recommended in difficult and dubious cases for the recognition of carcinogenic or dysplastic nature of morphologically similar proliferating structures.

[Immunomorphological study of the distribution of prekeratin with a molecular weight of 49 kilodaltons in various epithelial cells in rats].

Cryostat sections of various tissues of rat were stained using an indirect immunofluorescent method with monoclonal antibody against individual prekeratin with the molecular mass of 49 kilodalton (PK-49). Connective tissue endothelial cells, neurons, glia, haematopoetic tissue and smooth muscles were completely negative in this test. 46 morphological variants of epithelial structures were investigated. PK-49 was absent from all the stratified epithelia (epidermis, hair folliculi, oesophagus) but was expressed in virtually all simple epithelia of endodermal origin (exceptions: squamous lung alveolar epithelium and germinative epithelium of testis). There were negative (kidney tubules) as well as positive (bladder, mammary, glands) cell elements among mesodermal and ectodermal simple epithelia. High specificity of individual PK in respect to morphological variants of epithelia points out to the important role played by prekeratin-type intermediate filaments in morphogenesis.

[Immunohistochemical study of human breast tumors using monoclonal antibodies to intermediate filament proteins (nonproliferating epithelial structures in breast dysplasia)].

An immunohistochemical analysis of nonproliferating epithelial structures was carried out in 10 samples of human breast dysplasia and in 4 samples of tissue surrounding mammary gland carcinoma. Monoclonal mouse antibodies against individual prekeratins of rat monolayer epithelial antibodies of clone C12 against rat prekeratin with the molecular mass 49 kilodalton and antibodies of clone E3 against rat prekeratin with the molecular mass 40 kilodalton-monoclonal antibodies against vimentin (clone 30), as well as polyclonal antibodies against smooth muscle myosin and against the basement membrane glycoprotein laminin were used. The lining epithelium of all glandular structures reacted only with C12 antibodies. Two variants of myoepithelial cells containing myosin were detected. Variant I contains myosin and vimentin and is localized in intralobular ducts. Variant 2 contains myosin and prekeratin, recognized by E3 antibodies and is found in extralobular ducts.

[Evolution of hyperplastic foci in the rat liver induced by diethylnitrosamine].

The immunohistochemical localization of ligandin (L), A-protein (A) and alpha-fetoprotein (AFP) was studied in the liver of rats treated intraperitoneally with diethylnitrosamine (DENA). In early foci of altered cells there was a consecutive change in the content of antigens. The uniform content of L and A in these foci changed for the uniform content of only one marker. Late foci of altered hepatocytes were characterized by a heterogeneous distribution of the markers. 12-14 weeks after the treatment the neoplastic nodules with different antigenic phenotypes dependent on the morphological structure were detected. Cells in neoplastic nodules were uniform as to the marker content. Alpha-fetoprotein was unavailable in foci of altered cells and in neoplastic nodules either. It is concluded that evolution of most of foci follows the way of postnatal maturation similar to that in the normal liver and neoplastic nodules represent a side branch of differentiation blocked at different stages.

[Immunomorphological analysis of the level of differentiation of hepatocellular tumors in rats].

Combinations of three molecular markers (alpha-fetoprotein, ligandin and A-protein) were investigated in individual cells of the rat hepatocellular tumours by means of indirect immunofluorescence. It was found that each subtype of trabecular carcinoma classified on the basis of cytomorphological appearance as well, moderately, and poorly differentiated hepatomas, exhibited its characteristic combination(s) of the molecular markers studied. Each marker pattern observed in the tumours had its counterpart in the marker combination specific to one of the stages of normal liver tissue differentiation. Thus immunomorphological examination of protein marker combinations in hepatocellular carcinomas allowed the authors to propose an objective classification of these tumours according to their level of differentiation.

[Ligandin localization in the gonadal cells of rats at various stages of ontogeny].

Ligandin, a protein binding some carcinogens, steroids and other substances in the rat liver, has been found by means of indirect immunofluorescence in the gonad cells of different types: embryonic and mature Leidig cells in testes, ovarian thecal cells at the maturation stages (theca interna, atretic follicles and interstitial cells) and luteal cells of corpus luteum at pregnancy. Ligandin is found, thus, in cells which belong to various lines of cell differentiation. The functional role of ligandin is discussed.

[Evolution of hepatocyte population in the process of chemical carcinogenesis].

By means of two different markers of differentiation, using immunofluorescene method, the authors have characterized changes in the population of hepatic cells 1-8 weeks following the start of 3'-methyl-4-dimethyl-aminoazobenzene or 2-acetyl-aminofluorene action. Alpha-fetoprotein served as a marker of embryonic hepatocyte differentiation; while ligandin-as a marker of high-differentiated mature hepatocytes. The toxic effect of the carcinogens on hepatic stem cells was accompanied with a decrease of ligandin content in centrilobular hepatocytes. Among newly proliferating elements "oval" cells, cells of bile tract epithelium and most of basophilic hepatocyte-like cells fail to contain either alpha-fetoprotein or ligandin. Small groups of basophilic hepatocyte-like cells would contein alpha-fetoprotein. In cells of high cylinder-shaped epithelium of intestinal type ligandin was found, but alpha-fetoprotein was not found. The latter was absent in oxyphilous hepatocytes of hyperplastic nodules. In terms of ligandin content three types of morphologically identical nodules were differentiated; a) ones not containing this protein, b) ones containing it in amounts common to normal mature hepatocyte, and c) hyperdifferentiated nodules containing abnormally high concentrations of ligandin. Within one nodule cell all cells were identical in ligandin content. Thus, it is shown that at early stages of chemical carcinogenesis there occure in the liver multiple foci of differentiation of various kind. These intensive processes are assumed to be essential for tumor evolution in the tissue.
