A series of 2-arylidene-4-cyclopentene-1,3-diones and 2-arylideneindan-1,3-diones, as well as mono- and bis(arylidene substituted)cycloalkanones, was synthesized and examined for antitumor activity against ascites sarcoma-180. All the 2-arylidene-4-cyclopentene-1,3-diones and one arylideneindan-1,3-dione (where the arylidene group was either a hydroxybenzylidene or substituted hydroxybenzylidene) exhibited a high degree of activity. Among both types of 1,3-diones and 3-methoxy-4-hydroxybenzylidene derivatives were found to possess the greatest potency, while all the mono- and bis(arylidene)cycloalkanones were found to be inactive.
Experiments were performed on dogs with hypothalamic lesions as well as on intact and hypophysectomized dogs. Adrenal venous blood samples were collected and analyzed for 17-hydroxycorticosteroids (17-OHCS). In intact dogs, a marked increase in adrenal 17-OHCS secretion was observed after an i.v. injection of histamine and following an i.v. infusion of cyanide. Adrenal 17-OHCS secretion in response to histamine was markedly reduced but not completely abolished by hypophysectomy and was partially impaired by anterior median eminence lesions and by posterior median eminence lesions, but was not significantly impaired by supramammillary lesions. Adrenal 17-OHCS secretion in response to cyanide was completely abolished by hypophysectomy, markedly reduced by posterior median eminence lesions and partially impaired by anterior median eminence lesions and by supramammillary lesions. Thus, a partial dissociation of the adrenocortical secretory responses to histamine and cyanide was observed in dogs with hypothalamic lesions, suggesting that the mechanism and the pathway to the anterior pituitary involved in the adrenocortical secretory response to histamine and cyanide are somewhat different.
Twenty-nine adult patients with skeletal Class III malocclusion were treated by combined surgical-orthodontic management. A thorough analysis enabled us to point out the exact location and degree of the deformity and was of great help in determining the appropriate types of treatment. Corrections were made in 12 patients by the mandibular body ostectomy in the premolar region; the curved oblique osteotomy, in which the ascending ramus was cut obliquely on a curved line from the anterior border of the ramus to the angle, was used for the treatment of the remaining 17 patients. The resulting profile and occlusal improvements were satisfactory, Preoperative orthodontic treatment such as leveling of the dental arches, expansion of the upper dental arch to accommodate the maxilla to the mandible, correction of the upper and lower incisal inclinations, extraction of the premolars to decrease horizontal discrepancy or alignment of malpositioned teeth, and postoperative adjustment were effective in providing definite changes in profile and a stable occlusion-an important factor in the prevention of postoperative relapse.

Adrenal venous blood samples were collected from conscious and anesthetized dogs before and after an injection of pilocarpine and were analyzed for 17-hydroxycorticosteriods (17-OHCS). In conscious dogs the adrenal 17-OHCS secretion rate increased from a pre-injection secretion rate of 0.11 plus or minus 0.014 to 1.58 plus or minus 0.069 (mean plus or minus SE) mug/kg/min following an intravenous injection of pilocarpine (0.3 mg/kg); there was a return to the baseline level by 120 min. In anesthetized dogs a marked increase in adrenal 17-OHCS secretion was also observed, which was completely abolished by hypophysectomy A noticeable impairment of the adrenocortical response to pilocarpine was found to be produced by radio-frequency-current lesions of the anterior median eminence. A partial depression of the response was also observed in dogs with the anterior ventromedial hypothalamic lesions. Lesions destroying other hypothalamic areas were found to be without significant effect upon the adrenal cortical secretory response to pilocarpine.
In dogs with hypothalamic lesions as well as in intact and hypophysectomized dogs, adrenal venous blood samples were collected and analyzed for 17-hydroxycorticosteroids (17-OHCS). In intact dogs a marked increase in adrneal 17-OHCS secretion was observed following the i.v. infusion of cyanide as well as after the i.v. injection of pilocarpine; this was completely abolished by hypophysectomy. The adrenocortical response to pilocarpine was markedly impaired and that to cyanide partially suppressed after lesioning of the anterior median eminence. However, after lesioning of the posterior median eminence, the adrenocortical secretory response to pilocarpine was almost the same as in intact dogs, whereas the response to cyanide was markedly impaired. Thus, a dissociation of adrenocortical secretory responses to cyanide and pilocarpine was demonstrated in dogs with hypothalamic lesions.

A case of Leber's amaurosis congenita is presented. The patient was bilaterally and congenitally blind. She had nonrecordable ERG responses and a striking fundus appearance with multiple irregularly shaped yellowish white flecks deep in the retina, peripheraly distributed over an area extending from behind the equator to the ora serrata. No flecks were present in narrow bands on both sides of the retinal vessels and in the whole nasal portion of the fundus. Fine powdery granules of pigment were diffusely spread in the areas that were devoid of flecks. The macula and the optic disc showed no gross abnormality. Although this fundus appearance resembles that of the flecked retina syndrome and Oguchi's disease, retinal function in this case is more profoundly affected than in either of the two.
