In vitro secretion of insulin-like growth factor-binding proteins from liver of striped bass, Morone saxatilis.

In vitro secretion of insulin-like growth factor-binding proteins (IGFBPs) from liver of striped bass (sb: Morone saxatilis) was studied using a simple organ-culture system. Liver cubes (1 mm3) were cultured in minimum essential medium with Earle's salts containing 0.1% bovine serum albumin and 100 U/ml penicillin in 5% CO2/95% O2 at 16 degrees. The amount of double-stranded DNA in these cultured liver cubes did not change by 192 hr in the culture, but decreased by 216 hr. Four IGFBPs (a 23- to 24-kDa protein, a 28- to 30-kDa protein, a 35- to 39-kDa protein, and an 85- to 90-kDa protein) were identified in striped bass serum by Western ligand blotting; two of these IGFBPs, 23-24 kDa (sbIGFBP-1) and 28-30 kDa (sbIGFBP-2), were consistently detected in culture media by Western ligand blot analysis. The intensity of the blot for sbIGFBP-2 was consistently greater than that of sbIGFBP-1, which was no longer secreted after 96 hr in culture. The effects of hormones and growth factors on IGFBP secretion by liver tissue were measured after 48 hr in culture. sbIGFBP-1 in the medium was significantly decreased by adding ovine prolactin (10 micrograms/ml), bovine insulin (100 micrograms/ml), and bovine IGF-I (100 ng/ml), but was increased by 17 beta-estradiol (E2: 5 and 50 ng/ml).(ABSTRACT TRUNCATED AT 250 WORDS)

Effects of sex hormones on oncogene expression in the vagina and on development of sexual dimorphism of the pelvis and anococcygeus muscle in the mouse.

Neonatal treatment of female mice with diethystilbestrol (DES) is known to induce ovary-independent persistent proliferation and cornification of vaginal epithelium. This irreversibly changed vaginal epithelium persistently expressed higher levels of c-jun and c-fos mRNAs, which was not altered by postpubertal estrogen. Sexual dimorphism was encountered in mouse pelvis and anococcygeus muscle. Postpubertal estrogen changed the shape of the pelvis to the female type and postpubertal androgen changed it to the male type. Neonatal exposure to DES and to the antiestrogen tamoxifen altered the developmental pattern of the pelvis, which contained lower concentrations of calcium and phosphorus than controls. The size of anococcygeus muscle was increased by postpubertal androgen but decreased by postpubertal estrogen. However, neonatal estrogen (DES) exposure permanently enlarged the anococcygeus muscle. Thus, neonatal treatment of mice with estrogen and antiestrogen results in irreversible changes in nonreproductive as well as reproductive structures.

Biochemical changes during growth and regression of pregnancy-dependent mammary tumors of GR/A mice.

The weights of pregnancy-dependent mammary tumors (PDMT) of GR/A mice continued to increase until parturition and decreased soon after delivery; however, mitotic indices in epithelial cells and stromal cells of PDMT reached a maximum plateau on Day 18-19 of pregnancy and decreased thereafter. Growth of PDMT in progesterone-treated mice on Day 15 of pregnancy was higher than that in 17 beta-estradiol-treated mice and no treatment controls. DNA fragmentation was observed in PDMT on Day 20 of pregnancy and just after parturition. Two-dimensional gel electrophoresis of PDMT extracts revealed that five and six protein spots appeared newly on Day 20 of pregnancy and just after parturition, respectively. N-terminal amino acid sequences of two of the protein spots were identical to that of alpha-lactalbumin. PDMT on Day 15 and 20 of pregnancy and just after parturition secreted matrilysin, one of the matrix metalloproteinases, which was identified by Western blotting. However, matrilysin was not found in hormone-independent autonomous mammary tumors of the mouse. Estrogen receptor and c-fos mRNA expression levels in PDMT were high on Day 15 of pregnancy but low on Day 20 of pregnancy and just after parturition. These findings suggest that regression of PDMT is caused by apoptosis, and new proteins expressed on Day 20 may participate in the process of regression.

[Studies on the bone metabolisms in either after natural menopause or surgical menopause: implications of IGF-IGFBP system for postmenopausal osteoporosis].

It is well established that accelerated bone loss occurs in association with estrogen deprivation as seen following the natural menopause and in premenopausal women undergoing surgical oophorectomy (i.e., surgical menopause). We have measured serum levels of bone biochemical markers after both natural menopause and surgical menopause. Circulating levels of insulin-like growth factor-I (IGF-I), which is considered to be the local regulator of osteoblast activity and one of its binding protein, insulin-like growth factor binding protein-4 (IGFBP-4) which binds to IGF-I and suppress its biological activity, were also measured. Bone mineral density measured by dual energy X-ray absorptiometry was decreased more rapidly after surgical menopause. A concomitantly higher rate of bone turnover as assessed by bone biochemical markers was observed after surgical menopause, and thus the levels of procollagen type I C-peptide, pyridinoline and deoxypyridinoline were increased. The serum levels of IGF-I were significantly reduced after natural menopause compared with that after surgical menopause. The levels of IGF-I were correlated with bone mineral density after natural menopause (r = 0.62, p < 0.001), but no significant correlation was observed between these two variables after surgical menopause. The binding activity of IGFBP-4 was significantly greater after surgical menopause than after natural menopause. A stronger inverse correlation existed between the binding activity of IGFBP-4 and bone mineral density after surgical menopause (r = -0.90, p < 0.001) compared to that after natural menopause (r = -0.29, p < 0.05). The simplest explanation is that whereas the loss of bone depends upon the decreased level of IGF-I after natural menopause, after surgical menopause it depends upon the increased level of IGFBP-4.

Factors affecting the incidence of death at home of residents of Hida district, Japan.

This study compared the percentage of residents who died from disease at home in Hida district, Japan, between the years 1979-1981 and 1989-1991, and found it decreased from 46.9% during the earlier period to 32.6% during the latter. It differed significantly by age and underlying cause of death. Residents aged 85 and over were almost twice as likely to die at home as were all other subjects. Patients who died of malignant neoplasms showed the lowest proportion of death at home. A greater percentage of residents who lived far from hospitals or those who had the causal disease for a prolonged time died at home. The proportion for 1989-1991 was lower than that for 1979-1981 after adjustment for age, gender, cause of death, period of illness, and availability of hospital. It was speculated that the increase of hospital beds affected the proportion of death at home.

O-antigenic lipopolysaccharide of Vibrio cholerae O139 Bengal, a new epidemic strain for recent cholera in the Indian subcontinent.

Lipopolysaccharide (LPS) from Vibrio cholerae O139 Bengal contained colitose (3,6-dideoxy-L-galactose) in addition to glucose, L-glycero-D-manno-heptose, fructose, glucosamine and quinovosamine in its polysaccharide and only glucosamine in lipid A, while perosamine, a characteristic component sugar of V. cholerae O1 LPS, was absent. 3-Hydroxydodecanoic, tetradecanoic and hexadecanoic acids as ester-bound fatty acids and 3-hydroxytetradecanoic acid as amide-bound fatty acid were identified in the lipid A. A very high serological specificity of O139 LPS distinct from that of O1 V. cholerae was demonstrated by passive hemolysis and passive hemolysis inhibition tests by using the LPS either as antigen for sensitizing sheep red blood cells or as inhibitor in the latter test.

N-3-hydroxypropionyl-alpha-D-perosamine homopolymer constituting the O-chain of lipopolysaccharides from Vibrio bioserogroup 1875 possessing antigenic factor(s) in common with O1 Vibrio cholerae.

A structural study was performed by 13C-n.m.r. spectroscopy and methylation analysis of the O-chain of lipopolysaccharide (LPS) from Vibrio bioserogroup 1875 possessing antigenic factor(s) in common with O1 Vibrio cholerae. It was demonstrated to contain a linear homopolymer of (1-->2)-linked N-3-hydroxypropionyl-alpha-D-perosamine [4-(3-hydroxypropanamido)-4,6-dideoxy-alpha-D-mannopyranose], which is very similar to, but not identical with, both (1-->2)-linked linear N-3-deoxy-L-glycero-tetronyl(S-2,4-dihydroxybutyryl)-alpha-D - perosamine homopolymer and (1-->2)-linked linear N-acetyl-alpha-D-perosamine homopolymer which constitute the O-chains of O1 V. cholerae and non-O1 V. cholerae bioserogroup Hakata LPS respectively.

Occurrence of 2-O-methyl-N-(3-deoxy-L-glycero-tetronyl)-D-perosamine (4-amino-4,6-dideoxy-D-manno-pyranose) in lipopolysaccharide from Ogawa but not from Inaba O forms of O1 Vibrio cholerae.

A structural study by GC-MS, methylation analysis, and 1H and 13C NMR was carried out on alpha (1-->2)-linked linear N-(3-deoxy-L-glycero-tetronyl)-D-perosamine homopolymer constituting the O-polysaccharide chain of lipopolysaccharide from O1 Vibrio cholerae Ogawa and Inaba O forms. Occurrence of 2-O-methyl-N-(3-deoxy-L-glycero-tetronyl)-D-perosamine was demonstrated at the non-reducing terminus of the perosamine-homopolymer of lipopolysaccharide from the Ogawa O form in contrast to the presence of N-(3-deoxy-L-glycero-tetronyl)-D-perosamine at the nonreducing terminus for the Inaba O form.

Morphometric analysis of the pelvis in mice treated neonatally with tamoxifen.

The pelves of male and female C57BL/Tw mice given five daily injection of 100 micrograms tamoxifen, 50 micrograms dihydrotestosterone (DHT), or 2 micrograms diethylstilbestrol (DES) from the day of birth were examined morphometrically and histomorphometrically. Total areas of the pelvis, ilium, ischium, and pubis were significantly smaller in neonatally tamoxifen-treated mice than in the controls. There was no significant difference in length of the ischium between tamoxifen-treated and control mice of both sexes. However, lengths of ilium and pubis, and widths of ilium, pubis, and ischium in tamoxifen-treated male and female mice were significantly smaller than in the respective controls. In contrast, neonatal treatment with DHT or DES did not affect the shape of the pelvis of either sex. In the neonatally tamoxifen-treated females, the number of osteoblasts and osteoclasts per 200 microns trabecular surface length and per 10,000 microns2 subperiosteal area of pubic bone section was smaller than in the controls. Inhibition of ossification persisted in the junction of the pubis and ischium of pelves treated with tamoxifen in vitro. These results suggest that neonatally administered tamoxifen mainly retards the growth of the ilium and pubis in mice by changing the activities of osteoclasts and osteoblasts, and that tamoxifen acts directly on the neonatal mouse pubis to inhibit its ossification.

Light and electron microscopic study of remodeling and maturation process in autogenous graft for anterior cruciate ligament reconstruction.

We evaluated the remodeling process of autogenous patellar tendon graft for anterior cruciate ligament (ACL) reconstruction by means of light microscopic (LM) and electron microscopic (EM) examinations from the biopsy specimens obtained at the time of second-look arthroscopy. Twenty-one patients were examined at various times postoperatively (from 6 weeks to 15 months, mean 9.5 months), and the results were correlated with the morphology of normal patellar tendon and normal ACL. Our study showed that the graft was revascularized in the early postoperative period, fibroblastic remodeling took place, and the graft obtained gross similarity to the original ACL on their arthroscopic and LM appearances at approximately 1 year postoperatively. However, EM study showed that at both approximately 6 months and 1 year postoperatively the grafts consisted equally of active fibroblasts with a higher cytoplasm-to-nucleus ratio compared with normal ACL. Collagen fibrils of these grafts were of uniformly small diameter compared with normal patellar tendon and ACL. Our results with ultrastructural study suggest that the grafts were still immature even at 1 year postoperatively.

Developmental changes in mouse placental cells from several stages of pregnancy in vivo and in vitro.

Developmental changes in mouse placentae from the 6th to the 18th day of pregnancy were studied in vivo and in vitro. Placental volume increased from the 6th to the 18th day of pregnancy; however, the total number of cells per placenta reached a plateau on the 14th day. Decidual cells were predominant in the placenta on the 6th day. Placentae obtained from the 10th to the 18th day contained decidual cells, trophoblastic (labyrinth and spongiotrophoblast) cells, and trophoblast giant cells. Decidual cells increased in number from the 6th to the 10th but decreased on the 14th day, whereas trophoblastic cells increased linearly until the 14th day. Two types of placental cells were distinguished in vitro: small fibroblast-like cells and large flattened cells containing 2-3 nuclei. The large cells reacted to anti-desmin antibody, indicating their decidual character. The small cells reacting to anti-keratin antibody appeared to be trophoblastic cells. Decidual cells from all days of gestation were nonproliferative, regressing with time in culture. 17 beta-Estradiol (E, 10(-9) and 10(-8) M), progesterone (P, 10(-10), 10(-9), and 10(-8) M), and a combination of E and P (10(-9) M each) stimulated proliferation of the trophoblastic cells only from the 6th and the 10th days. Keoxifene (2 x 10(-7) M), but not tamoxifen, significantly inhibited the E-induced proliferation of the trophoblastic cells.(ABSTRACT TRUNCATED AT 250 WORDS)

Immunocytochemical localization of progesterone receptor in the reproductive tract of adult female rats.

The distribution of the progesterone receptor (PR) was investigated immunocytochemically in female reproductive tracts of rats during the estrous cycle and early pregnancy through use of an anti-PR monoclonal antibody. PR was localized predominantly in the nuclei of epithelial, stromal, and muscle cells in the uterus and vagina during the estrous cycle. In the uterus, the nuclei of epithelial cells were stained intensively at diestrus, while the PR staining of the stromal cells was more intense at proestrus than at any other stage of the cycle. PR expression during the cycle in muscle cells of the myometrium was similar to that in the endometrial stromal cells. In the vagina, however, PR expression during the cycle was approximately the same among epithelial, stromal, and muscle cells, the nuclei of which were stained deeply at proestrus. Ovariectomy at various stages of the cycle altered the PR expression appearing in the uterus and vagina during the cycle. In ovariectomized rats, estrogen increased the PR immunoreaction of various types of cells examined in the uterus and vagina except for the uterine epithelial cells. The reaction of these uterine epithelial cells was decreased by estrogen but was increased by progesterone given after estrogen; however, progesterone given alone reduced the reaction. In the epithelial and stromal cells of the uterus, intensity of the staining was increased after mating, reaching maximum on Day 3 of pregnancy, and then decreased on Day 4 (day of implantation), while in epithelial and stromal cells of the vagina the staining remained weak during early pregnancy.(ABSTRACT TRUNCATED AT 250 WORDS)

Lipopolysaccharide isolated from a new O-antigenic form (O13) of Vibrio parahaemolyticus.

The chemical properties of a lipopolysaccharide (LPS) isolated from a new O-antigenic form (O13) of Vibrio parahaemolyticus were investigated. The LPS contained glucose, galactose, L-glycero-D-manno-heptose and glucosamine. 2-Keto-3-deoxy-octonate (KDO) was not detected in the LPS by the periodate-thiobarbituric acid test (Weissbach's reaction) under conventional hydrolysis conditions. Instead, phosphorylated KDO (X1 and X2) was found in its strong-acid hydrolysate. This sugar composition was identical to that of V. parahaemolyticus O3, O5 and O11 LPS, indicating that, based on the sugar composition, O13 LPS belongs to Chemotype III to which O3, O5 and O11 belong. In addition, structural study demonstrated the presence of KDO 4-phosphate in its inner-core region.

A chemotaxonomic study of Vibrio fluvialis based on the sugar composition of the polysaccharide portion of the lipopolysaccharides.

A chemotaxonomic study was carried out with a new serotyping scheme comprising 35 O-antigen groups of Vibrio fluvialis on the basis of the sugar composition of the polysaccharide portion of their lipopolysaccharide (LPS). A previously developed rapid method of preparing samples for compositional sugar analysis was employed. The 35 O-antigen groups were divided into 21 chemotypes. It is noted that a rarely occurring component sugar of gram-negative bacterial LPS, D-glycero-D-manno-heptose, and two kinds of uronic acids, i.e., galacturonic acid of a weakly bound type and glucuronic acid of a strongly bound type, were found in common in all the 21 chemotypes. A characteristic sugar component of gram-negative bacterial LPS, 2-keto-3-deoxyoctonate (KDO), was not detectable in any of the 21 chemotypes. Instead, three kinds of "KDO-like substances" were found, one in each of three chemotypes (III, XI and XVII). They were strongly positive in Weissbach's periodate-thiobarbituric acid test for KDO, but definitely not identical to it in high-voltage paper electrophoresis (HVPE); the substance present in chemotype XI was indicated by HVPE to be 3-deoxy-D-threo-hexulosonic acid which is a sugar constituent of Vibrio parahaemolyticus O7 and O12 LPS.

ED-110, a novel indolocarbazole, prevents the growth of experimental tumors in mice.

A new indolocarbazole compound, ED-110, which was obtained by glucosylating a microbial product (BE-13793C) and is a potent topoisomerase I inhibitor, showed characteristic inhibitory effects on the growth of 12 human tumor cell lines tested. The IC50 values of ED-110 against 9 of the 12 lines ranged from 11.5 micrograms/ml to 0.07 microgram/ml, while the remaining 3 lines were quite resistant (IC50, > 100 micrograms/ml). In in vivo experiments, i.p. treatment with ED-110 increased the survival period by more than two-fold in mice implanted i.p. with P388, L1210, L5178Y or EL4 murine leukemic cells. The minimum effective dose increasing the life-span of mice bearing P388 leukemia by 25% was < 2.5 mg/kg/day x 10 and the maximum tolerated dose was > 160 mg/kg/day x 10. ED-110 was also effective against the spontaneous metastasis of mouse Meth A fibrosarcoma cells and the growth of xenografted MKN-45 human stomach cancer cells as well as s.c. implanted mouse colon 26 and IMC carcinoma cells. These results indicated that ED-110 may have potential as a new antineoplastic agent with a large chemotherapeutic index and a wide range of effective doses.
