Stereo (C7)-dependent topoisomerase II inhibition and tumor growth suppression by a new quinolone, BO-2367.

A new antimicrobial quinolone (-)BO-2367, (-)-7-[(1R*, 2R*, 6R*)-2-amino-8-azabicyclo[4.3.0.]-non-3-en-8-yl]-1- cyclopropyl-6,8-difluoro-1,4-dihydro-4-oxo-3-quinolinecarboxylic acid, strongly inhibited both mammalian and bacterial topoisomerase II. The IC50 values of (-)BO-2367 against the DNA relaxation activity of L1210 topoisomerase II and the supercoiling activities of Escherichia coli gyrase and Micrococcus luteus gyrase were 3.8, 0.5, and 1 microM, respectively. This compound enhanced double-stranded DNA cleavage mediated by topoisomerase II not only with purified enzyme, but also with intact L1210 cells. All these activities of (-)BO-2367 were more than 2-fold stronger than those of VP-16. Intriguingly, (+)BO-2367, which has an enantiomeric substituent at the C7 position of (-)BO-2367, did not affect the activity of the mammalian topoisomerase II, while it inhibited E. coli gyrase. Intraperitoneal injection of (-)BO-2367 at 0.08 mg/kg increased the lifespan of CDF1 female mice bearing ascitic L1210 leukemia by 2.4 times, and subcutaneous injection at 1.25 mg/kg completely inhibited the growth of colon 26 carcinoma implanted subcutaneously. These results suggest that (-)BO-2367 is a potent antitumor agent which targets topoisomerase II. These enantiomers should be a useful tool for studying drug-topoisomerase II interactions.

Epidermal growth factor receptor levels in reproductive organs of female mice exposed neonatally to diethylstilbestrol.

Binding of epidermal growth factor (EGF) to membrane preparations of vagina, uterus, ovary, oviduct, and liver was examined in mice treated neonatally with diethylstilbestrol (DES) and compared with that in untreated mice. Binding in the vagina (12.5 +/- 0.73 fmol/mg protein) was somewhat higher than in the uterus (8.0 +/- 0.34 fmol/mg protein). Level of specific binding was of the order: liver (18.4 +/- 1.09 and 16.0 +/- 1.53 fmol/mg protein) > vagina (12.5 +/- 0.73 and 8.2 +/- 0.57 fmol/mg protein) > uterus (8.0 +/- 0.34 and 6.8 +/- 0.56 fmol/mg protein) > ovary (6.8 +/- 0.36 and 8.0 +/- 1.05 fmol/mg protein) > oviduct (2.1 +/- 0.32 and 1.7 +/- 0.05 fmol/mg protein) in control and neonatally DES-exposed mice, respectively. Thus, neonatal DES exposure significantly lowered the binding site level only in the vagina, without modifying the binding affinity (Kd = 5.4 x 10(-9) M in controls vs 4.6 x 10(-9) M in DES-exposed mice). Reduction of EGF receptor level in the vagina correlates with ovary-independent persistent proliferation and keratinization of the vagina induced by neonatal DES exposure. EGF receptors were immunohistochemically demonstrated in epithelial cells of vagina, uterus, and oviduct and in stromal cells in uterus and oviduct using a polyclonal antibody to human EGF receptor protein.

Altered mammary responsiveness to estradiol and progesterone in mice exposed neonatally to diethylstilbestrol.

Mammary glands from ovariectomised neonatally diethylstilbestrol (DES)-exposed (0.1 microgram daily for the first 5 days of life) mice seem morphologically indistinguishable from those of ovariectomised controls. However, administration of exogenous hormones reveals a differential response. In DES-exposed mice, estrogen implantation resulted in greater incidence of dilated ducts along with greater incidence of dilated ducts along with greater incidence and severity of terminal ductal hyperplasia and greater severity of cystic alveolar adenosis; combined estrogen and progestin treatment resulted in greater severity of terminal duct hyperplasia and less alveolar formation, and progestin treatment resulted in lower incidence and degree of lateral budding. Thus, mammary sensitivity to sex steroids is altered by early exposure of mice to DES.

Comparative study of sexual dimorphism of the innominate bone in rodents and amphibians.

Sexual dimorphism in the shape of the innominate bone was demonstrated in the rat, mouse, Chinese hamster (Cricetulus griseus), frog (Ranna nigromaculata), and newt (Cynops pyrrhogaster) with a computer-assisted morphometric technique. In the rat, mouse, and hamster, ratios of ischium and pubis widths to the innominate bone length (INL) were larger in the males than in the females; however, the ratio of pubis length to INL was larger in the females than in the males. In the frog, the ratio of distance between the upper edge of the acetabulum and the lower edge of the pubis to INL was larger in males than in females. In the newt, ratios of width of ischiopubis and the longitudinal length of the ischiopubis to INL were larger in females than in males.

Effects of sex steroids on the development of sexual dimorphism in mouse innominate bone.

The sexual dimorphism of the innominate bone was examined in 14 strains of mice. In female mice of all strains, the pubis was significantly longer and thinner than that in the strain-matched males. In 13 of 14 strains, the ischium in the male was longer and thicker than in the female. In the testicular-feminized male (Tfm) mouse, the ischium was longer and thinner than that in the wild-type male, resembling that of the wild-type female. The pubis of the Tfm mouse was longer than in the wild-type males. The pubis width in the Tfm mouse was between those of the wild-type male and female. Gonadectomy at ages of 5, 10, 20, 30, and 60 days in both sexes showed that the ischium develops as the female type when sex hormones are absent. In contrast, postnatal testicular androgen induces the male-type ischium. Gonadectomy at 60 days had a slight effect on the pubis, indicating that sexual dimorphism of the pubis was determined before 60 days of age. Estrogen receptors (ER) were immunohistochemically demonstrated in bone cells of 0- to 60-day-old mice. ER was found exclusively in the periosteum of the pubis at the day of birth; however, it appeared in bone cells of all parts of pelvis at 10-60 days. These results indicate that sexual dimorphism of the pubis is consistent for the 14 mouse strains examined, and that the shape of the pubis is determined by sex steroids before 60 days of age. Since ER exist in the bone cells, morphogenesis of the pelvis may be regulated by these sex steroids.

Terminal deletion of the short arm of chromosome 3.

A boy with growth and mental retardation, flat occiput, high and broad forehead, blepharoptosis, narrow palpebral fissures, low set, malformed ears, short neck, anal atresia, deep sacral dimple is reported. High-resolution banding analysis showed terminal deletion of the short arm of chromosome 3 (46,XY,del(3)(p25.3)). Deletions of the short arm of chromosome 3 are relatively rare. The clinical features of the patient are compared with those of 19 previously reported cases.

Effects of antiestrogens on adult and neonatal mouse reproductive organs.

Estrogenic potencies of various antiestrogens, including keoxifene (Kx) and trifluoperazine (Tfp), on reproductive tracts of ovariectomized adult mice, and effects of neonatal Kx and Tfp on reproductive organs were studied in C57BL/Tw mice. In adult ovariectomized mice, weight, DNA, and protein of the uterus and vagina were increased by 3 daily injections of 100 micrograms clomiphene, tamoxifen (Tx), and nafoxidine, and of 1 microgram estradiol-17 beta (E), but not by Kx. Antiestrogenic potency of Kx was studied in adult mice given injections of E. Kx significantly suppressed the E-induced increase in weight, DNA, and protein in the uterus and vagina. Tfp (20 micrograms), known as a tranquilizer and an antiestrogen, had no estrogenic effect on either organ. Male and female mice given 5 daily injections of Kx or Tfp from the day of birth were examined at 30, 40, and 60 days of age. Weights of testis, epididymis, and seminal vesicle in neonatally Kx-treated mice were significantly lower than in controls at 30 and 40 days. Spermatozoa were not formed in the seminiferous tubules of Kx-treated mice, although spermatogenesis occurred at 60 days. In neonatally Kx-treated females, weight of the uterus at 60 days and of the vagina at 40 and 60 days was significantly lower than in controls. Corpora lutea were absent from the ovaries of Kx-treated females. In neonatally Tfp-treated mice of both sexes at all ages examined, no differences were found in organ weights or histology, other than lower spermatogenic indices at 40 and 60 days of age.(ABSTRACT TRUNCATED AT 250 WORDS)

[Chemical and serological study of lipopolysaccharide isolated from Shigella flexneri 88-893 possessing a new type-antigen].

The O-specific polysaccharide chain which represents a new type-antigen in lipopolysaccharide (LPS) of Shigella flexneri 88-893 was investigated. The O-polysaccharide chain was found to be composed of repeating units comprising rhamnose, N-acetylglucosamine and glucose (3:1:2). In the passive hemolysis test, group-6 antiserum of S. flexneri exhibited a high hemolytic titer (50% hemolysis titer: 7,900) against sheep red blood cells (SRBC) sensitized with intact 893 LPS, but virtually no hemolytic activity against SRBC sensitized with alkali-treated 893 LPS. None of the type-specific antisera (I-VI), showed any significant hemolytic titer against SRBC sensitized with either intact or alkali-treated 893 LPS. Thus, 893 LPS contained both the group-6 antigen and a new type-antigen which is distinct from any known type-antigen of S. flexneri.

[The clinical and pharmacokinetic evaluation of two-route chemotherapy with cisplatin and sodium thiosulfate in combination with angiotensin II].

Thirty-six courses of chemotherapy with cisplatin (CDDP) and sodium thiosulfate (STS) were performed in 31 patients with gynecologic cancer 2 weeks after operation under the hypertensive condition induced by angiotensin II (AT-II). One hundred-fifty mg of CDDP/body was intraperitoneally administered while the usual systolic blood pressure was increased to 130-140% by AT-II. The hypertension was maintained for 15 minutes after finishing CDDP infusion, then 8g of STS was intravenously infused immediately after the cessation of AT-II. The urinary Pt level measured 15 minutes after finishing CDDP infusion was extremely low at 1.37 +/- 0.47 micrograms/ml, in spite of the significantly high levels of plasma total Pt (7.83 +/- 0.85 micrograms/ml) and filtrable Pt (4.02 +/- 0.55 micrograms/ml). This suggests that, during renal vasoconstriction induced by AT-II, the renal blood flow as well as renal uptake of CDDP was decreased. Plasma filtrable Pt levels were measured at 15, 30, 60 and 120 minutes after intraperitoneal administration of CDDP. At 15 and 30 minutes a significantly higher blood concentration was found than in the control group (p less than 0.05). However, CDDP-induced toxicities of the bone marrow, liver, kidney and alimentary tract were not increased. The extreme vasoconstriction in the kidneys and other organs induced by AT-II might have protected these organs from the toxicities of CDDP despite the fact that STS infusion was delayed by 15 minutes after CDDP infusion. STS infused immediately after the cessation of AT-II could neutralize the CDDP preventing the occurrence of toxicities.(ABSTRACT TRUNCATED AT 250 WORDS)

Postnatal vaginal nodules induced by prenatal diethylstilbestrol treatment correlate with later development of ovary-independent vaginal and uterine changes in mice.

Pregnant ICR/JCL mice were given 4 daily subcutaneous injections of 0.2-2000 micrograms diethylstilbestrol (DES) starting on day 15 of gestation. Offspring of mothers given DES were killed at 1-10 days of age and examined for nodules of enlarged polygonal cells under the epithelium of the Müllerian (upper) vagina. Some offspring were ovariectomised at 30 days and killed at 120 days. The nodules which appeared in the prenatally DES-exposed mice (2-2000 micrograms/day) at 3-7 days were not connected with the epithelium of the sinus vagina and reacted positively to an antibody to epidermal growth factor. Nodule formation may prove to be prodromic of later ovary-independent vaginal changes in the DES-exposed mice. Epithelial stratification (2-2000 micrograms/day) and downgrowths and/or pegs (20-2000 micrograms/day) occurred in vaginae of ovariectomized mice exposed prenatally to DES; however, adenosis-like lesions occurred only in the offspring of mothers given the highest prenatal injections of 2000 micrograms DES. Wolffian remnants and hypospadias (2-2000 micrograms/day) were also encountered in the DES-exposed mice. Ovary-independent stratification of the uterine epithelium (20-2000 micrograms/day) and disorganization of the circular musculature (2-2000 micrograms/day) were also observed in the DES-exposed mice. None of these changes was found in ovariectomised 0.2 micrograms DES-exposed and control mice.

Expression of chicken integrin beta 1 subunit in rat PC12 cells.

We transfected rat pheochromocytoma (PC12) cells with a cDNA encoding chicken integrin beta 1 subunit. The chicken integrin beta 1 subunit produced in stable transfectants associated with two major alpha subunits of rat integrins to form interspecific chimeric receptors. These receptors mediated cell spreading and initial neurite outgrowth on laminin as did corresponding endogenous integrins, although they were slightly less effective in inducing cell adhesion to laminin. These results indicate that chicken integrin beta 1 may functionally substitute for beta 1 subunit of rat integrins in PC12 cells. Apparently, the structure of the integrin beta 1 subunit is highly conserved in the evolution of these species.

Repression of integrin beta 1 subunit expression by antisense RNA.

A quail cell line (QT6-c) was co-transfected with pTEX vector expressing RNA complementary to chicken integrin beta 1 subunit mRNA (Anti-Int) and pRSVneo vector by a calcium phosphate method. Transfectants showing reduced expression of quail integrin beta 1 subunit were selected with an immunoblot assay, and a few positive clones were examined in detail. Northern blot and immunoblot analyses revealed that the Anti-Int caused a clear reduction of the transcript encoding integrin beta 1 subunit depending on culture conditions. The number of cell surface integrins also decreased in proportion to the decrement of the total amount of integrin beta 1 subunits. When one transfectant (QA23) was cultured in a serum-free medium, cell shape changed from fibroblast-like to neuron-like morphology accompanied by a low growth rate, and the cells did not form focal contact on fibronectin. A similar morphological change occurred in QT6-c cells when the cells were infected with Rous Sarcoma virus, which could produce the Anti-Int. The QA23 cells did not attach to fibronectin as efficiently as did the original QT6-c cells. These data suggest that reduced expression of integrin beta 1 subunit affects cell growth as well as cell morphology by disordering the interaction between integrins and matrix proteins and/or cytoplasmic proteins.

[Measurement of renal index on X-CT].

Morphological measurements of the kidney were made using X-CT images. Anteroposterior width of the kidney as well as the renal sinus on both right and left sides and the transverse width of the first lumbar vertebral body were measured from CT images. Changes according to age in anteroposterior width of the kidney and the transverse width of the lumbar vertebral body were investigated. As to the ratio of the anteroposterior width of the kidney (both right and left) to the transverse width of the lumbar vertebral body, and the ratio of the anteroposterior width of the kidney to the anteroposterior width of the renal sinus, the changes due to aging and the differences between male and female were studied. 1) In actual CT measurements, the average anteroposterior width of the right kidney was 47 mm, while that of the left kidney was 49 mm. The left kidney was slightly larger than the right kidney with statistical significance. In subjects up to 19 years of age, both the anteroposterior width of the kidney and the transverse width of the lumbar vertebral body showed correlation with age, while after the age of 20, there was no age-related correlation. 2) The average ratio of the anteroposterior width of the kidney to the transverse width of the lumbar vertebral body was 1.13 for the right kidney and 1.19 for the left kidney. The difference between those two items was statistically significant and the changes due to aging were slight. During the age between 4 and 64 years, they were constant and no significant change was noticed. No difference between male and female was found. From the age of 65 years, a decreasing tendency was noticed. 3) The ratios of anteroposterior width of the kidney to the anteroposterior width of renal sinus for both right and left kidneys in males and females were found to be inversely correlated with age. They were similar to the changes due to aging in renal functions of the cortex system. As seen on CT image, the ratio of the anteroposterior width of the kidney to the transverse width of the lumbar vertebral body, and the ratio of the anteroposterior width of the kidney to the anteroposterior width of the renal sinus could be the index of both the size of the kidney and the thickness of the renal parenchyma respectively.

Growth of mouse endometrial luminal epithelial cells in vitro: functional integrity of the oestrogen receptor system and failure of oestrogen to induce proliferation.

Normal endometrial luminal epithelial cells isolated from ovariectomized approximately 40-day-old BALB/cCrgl mice were purified by Percoll density gradient centrifugation and grown as primary cultures in collagen gel matrix and serum-free medium. Cells increased threefold in number during the 9-day culture period. Deletion of insulin, epidermal growth factor or bovine serum albumin resulted in decreased growth. Addition of any single factor to the unsupplemented medium had no effect. Relatively high levels of cytosolic oestrogen receptors and progestin receptors were demonstrable in the cultures. Addition of oestrogen did not enhance epithelial cell proliferation. On the contrary, all doses of oestrogen (180 fmol/l to 218 nmol/l) were inhibitory. Continuous exposure to oestradiol-17 beta (1.8 nmol/l) for 9 days in serum-free medium resulted in a decrease in cytosolic oestrogen receptors with an associated nuclear accumulation of oestrogen receptors. A corresponding increase in cytosolic progestin receptors was also observed, indicating that no qualitative modification of the oestrogen receptor system had occurred. Thus, as previously reported for vaginal epithelial cells, oestrogen, despite its stimulation of specific product synthesis (progestin receptors), did not increase proliferation of endometrial luminal epithelial cells in this culture system.

Quantitative analysis of the development of genital organs from the urogenital sinus of the fetal male mouse treated prenatally with a 5 alpha-reductase inhibitor.

The role of 5 alpha-dihydrotestosterone (DHT) in the development of the genital organs and in the differentiation of the genital tract into prostate, coagulating gland (CG), bulbo-urethral gland (BUG) and seminal vesicle (SV) in male mice exposed prenatally to the 5 alpha-reductase inhibitor 6-methylene-4-pregnene-3,20-dione (6-MP) has been examined quantitatively. Female ICR mice were given 7 daily s.c. injections of the inhibitor (400 mg/day) starting on day 12 of gestation and the experiment was terminated on day 19 when the fetuses were removed by Caesarean section. In the prenatally 6-MP-exposed male mice the anogenital distance was significantly shorter than in the controls. Feminization of the nipples and hypospadias of the phallic urethra were noted. Development of prostate, CG and BUG was significantly suppressed. SV and testis development were not affected. These results lend further support to the conclusion that DHT is necessary for the development of the urogenital sinus (prostate, CG and BUG) and penis, and for the regression of the nipples in male mice. Reproductive abnormalities were not found in 90-day-old mice of both sexes exposed to 6-MP in utero. The 6-MP-exposed male and female mice had a normal reproductive capacity when mated with normal mice. These results show that 6-MP-induced growth retardation of reproductive organs is evident on day 19 of gestation, but that such retardation is no longer apparent in the adult.
