Estimation of acute toxicity of ammonium sulphate to the fresh water catfish, Heteropneustes fossilis I. Analysis of LC50 values determined by various methods.

The acute toxic effects of ammonium sulphate to fresh-water catfish, Heteropneustes fossilis (H. fossilis) have been studied by determining LC50 values with 95% confidence limits, by the graphic method, the logistic method, the Spearman and Karber method and the trimmed Spearman-Karber method. The trimmed Spearman-Karber method was found the most ideal for ammonium salt toxicity test. The flaws in the trimmed Spearman-Karber method are also discussed.

Estimation of acute toxicity of ammonium sulphate to the fresh-water catfish, Heteropneustes fossilis. II. A histopathological analysis of the epidermis.

The toxicity of 4000 ppm (96 h LC50 value) of the inorganic fertilizer ammonium sulphate on the epidermis of Heteropneustes fossilis (H. fossilis) at different intervals of time has been studied. The destruction induced by the ammonium salt is massive. Secretion of a copious amount of slime from the goblet mucous cells leading to the exhaustion and/or shedding and subsequent disappearance is perhaps the first reaction to the toxicity of the irritant. Later, the polygonal epithelial cells of the outermost layer show cyclic stages of necrosis and sloughing followed by their regeneration and repair. The contents of the club cells show enormous shrinkage and condensation with subsequent replacement of their areas with a hazy substance. In the earlier stages of exposure, regeneration takes place quickly, side by side with the degenerative changes at different stages of experimentation. Later after 8 to 10 d, the degenerative changes slow down and the epidermis appears identical to that of the normal skin. Goblet mucous cells also showed several cyclic increases followed by decreases in number and activity.

Phase II trial of PALA in combination with 5-fluorouracil in advanced pancreatic cancer.

Phosphonacetyl-L-aspartate (PALA), in inhibitor of aspartate transcarbamylase that depletes uridine nucleotide pools, selectively potentiates the antitumor activity of 5-fluorouracil (5-FU) in preclinical models. Due to the promising results we obtained using PALA/5-FU in colorectal cancer, we performed a phase II trial in patients presenting with advanced pancreatic cancer. PALA was given intravenously at 250 mg/m2 on day 1, followed 24 h later by 2,600 mg/m2 5-FU given by 24-h infusion. Treatments were repeated weekly. A total of 41 patients who had not previously undergone chemotherapy were entered in the trial; of these, 35 were evaluable for response. Toxicity was generally mild to moderate; neurotoxicity (13/35) and diarrhea (8/35) predominated. Among the 35 patients, 1 achieved a complete response and 4, a partial remission, for an overall response rate of 14%. The median survival was 5.1 months. Pretreatment with PALA alone was not sufficient to enhance the activity of 5-FU in pancreatic cancer.

Acute toxic effects of mercuric chloride on the mucocytes of the epithelial lining of the accessory respiratory organ and skin of the air breathing catfish Heteropneustes fossilis (Bloch).

Acute toxicity of 0.3 ppm mercuric chloride on the mucocytes of the branchial diverticulum and skin of Heteropneustes fossilis results in cyclic increases followed by decreases in the density, area occupancy and volume at different intervals of exposure. The alterations in the two tissues do not follow the same path perhaps due to different modes of action of the mercury salt: The skin comes under direct contact effects, while the branchial diverticulum may be affected by hormonal imbalance caused by a stress effect.

Histopathological changes induced by acute toxicity of mercuric chloride on the epidermis of freshwater catfish--Heteropneustes fossilis (Bloch).

The toxic effects of 0.3 ppm (96-hr LC50 value) of mercuric chloride solution on the epidermis of Heteropneustes fossilis at different time intervals have been studied. The epidermis reacts instantaneously by secreting a profuse amount of slime, especially due to hyperactivity of the goblet mucous cells which subsequently degenerate and get lost. Later, all other cell types also get entangled in the degenerative process. The damaged cells, especially the club cells, get sloughed at the surface, forming a thick crust of debris from the degenerating cells. Side-by-side regeneration of the epidermis also continues from the lower layers of the intact tissue with the addition of all the cell types. However, even before attaining maturity, these budding cells, especially the club cells, show signs of degeneration, their degeneration occurring faster than their regeneration. Thus most of the space left behind by the degenerated club cells gets quickly filled with haphazardly arranged polygonal epithelial cells, giving the epidermis an altogether different morphology.

Eastern Cooperative Oncology Group study 1879: mitotane and adriamycin in patients with advanced adrenocortical carcinoma.

From November 1979 to July 1986, 52 patients (27 women and 25 men; median age 52 years) with advanced adrenocortical carcinoma entered a prospective, nonrandomized study evaluating moderate-dose mitotane and doxorubicin hydrochloride (Adriamycin). Thirty-two tumors (62%) were well differentiated and evidence of hormone production was present in 24 patients (46%). Patients with well-differentiated or functional tumors received mitotane, 6 gm daily; patients for whom mitotane failed or those with poorly differentiated, non-hormone-producing tumors received Adriamycin, 60 mg/m2 every 3 weeks. Initially, 36 patients were treated with mitotane and 16 patients with Adriamycin. Eight patients (22%) responded to mitotane and three (19%) responded to Adriamycin. No response was noted in the 15 patients for whom mitotane failed and who then received Adriamycin. Severe toxicity occurred in 36% of patients who received mitotane and in 26% who received Adriamycin. Overall median survival after onset of treatment was 14 months. We conclude that mitotane or Adriamycin used initially can induce tumor regression in about 22% and 19% of selected patients, respectively. However, Adriamycin is ineffective as second-line chemotherapy for patients with well-differentiated or functioning tumors for whom mitotane is ineffective.

The histochemistry of urea-unmasked glycosaminoglycans in the skin of the eel, Anguilla japonica.

In the connective tissues of the dermis and subcutis of the eel skin, the histochemistry of urea-unmasked glycosaminoglycans has been studied by means of combined staining and enzyme digestion procedures. The staining procedures employed were alcian blue (AB) pH 1.0, AB pH 2.5, aldehyde fuchsin (AF), periodic acid-Schiff (PAS), AB pH 2.5-PAS, high iron diamine (HID) and low iron diamine (LID) methods, whereas the enzymes used were Streptomyces and testicular hyaluronidases, chondroitinases ABC and AC and keratanase. The results obtained have shown that a substantial amount of dermatan sulfate and a relatively small amount of hyaluronic acid, chondroitin, chondroitin sulfate A and/or C were the glycosaminoglycans involved in the connective tissues of the eel skin and that the tissues were devoid of keratan sulfate.

Unusual manifestations of multiple cranial nerve palsies and mandibular metastasis in a patient with squamous cell carcinoma of the lip.

A 70-year-old man who had squamous cell carcinoma of the lip later presented with right mental neuropathy, ipsilateral progressive involvement of all three divisions of the 5th, the 7th and 8th cranial nerves, and complete ophthalmoplegia. Biopsy proven metastasis to the mandible was demonstrated. Although repeated studies of CT scan of the head and cerebral angiography were negative, CSF examination revealed positive cytology, elevated protein and low sugar content. Although vascular dissemination from the primary lip lesion to the mandible and brain stem cannot be ruled out, extension through perineurial space via the 5th cranial nerve have been speculated. This case demonstrates two unusual features of squamous cell carcinoma of the lip, namely metastasis to the mandible and meningeal spread with multiple cranial neuropathies.

Histochemical analysis of urea-unmasked glycosaminoglycans in the skin of the rat and mouse.

Histochemical analysis of urea-unmasked glycosaminoglycans has been performed in connective tissues of the rat and mouse skin by means of combined staining and enzyme digestion procedures. The staining procedures used were Alcian Blue pH 1.0, Alcian Blue pH 2.5, Aldehyde Fuchsin, periodic acid-Schiff (PAS), Alcian Blue pH 2.5-PAS, high iron diamine and low iron diamine methods. The digestive enzymes employed were Streptomyces and testicular hyaluronidases, chondroitinases ABC and AC and keratanase. The results obtained indicated that the major components of the glycosaminoglycans in the connective tissues of the skin were hyaluronic acid, dermatan sulphate and chondroitin sulphate A and/or C, whereas the tissues were devoid of keratan sulphate.

Chemotherapy drug dose alteration due to radiation therapy in an adjuvant situation in breast cancer.

Fifty-six patients with T1-T3 and T2-T3 N0 medial lesions of breast carcinoma were randomized after modified radical mastectomy to receive either cytoxan, methotrexate, 5-fluorouracil (5-Fu) (CMF) chemotherapy for 1 year (group A) or CMF for 1 year and postoperative radiation therapy (group B). Thirty-two patients received chemotherapy alone and 25 patients received both chemotherapy and radiation therapy. Twenty-five of 32 group A patients (median age 52) and 20/24 group B patients (median age 50) were evaluable. Leukopenia was the major cause of drug dose reduction in both groups. In group A, 4/25 patients (16%) had leukopenia at less than the 2500 level, whereas 8/20 (40%) group B patients had leukopenia at the same level. If 70% of all three drug dosages are considered as adequate chemotherapy, 21/25 (84%) patients received adequate chemotherapy in group A, and 10/20 (50%) in group B (p approximately equal to 0.029 from contingency table). It appears that radiation therapy in postmastectomy patients hinders adequate drug dose delivery in an adjuvant setting.

Phase II study of dibromodulcitol in colorectal, kidney, and other carcinomas.

In a phase II study dibromodulcitol (DBD), an alpha-omega dibrominated hexitol, was used to treat 99 previously treated patients with colon, rectal, kidney, and other tumors. Six patients were ineligible and 10 patients were nonevaluable for response. Aside from thrombocytopenia toxicity was moderate. Twenty-three patients had platelet nadirs of less than 50,000/mm3 and there were three thrombocytopenic-associated drug deaths. One of 21 rectal and 1 of 13 kidney cancer patients entered a remission. All but seven patients had received previous treatment with one or more cytotoxic agents. Previously treated patients with colorectal and kidney cancer appear to be resistant to DBD.

Histochemistry and the structure of the epidermis of a fresh-water teleost Noemacheilus botia (Hamilton-Buchanon) (Cobitidae, Pisces).

Functional organization and the histochemical nature of the various cellular components of the epidermis of Noemacheilus botia are described. The various histochemical techniques reveal the basic proteinaccous nature of the outer free margins of the polygonal cells of the most superficial layer of the epidermis. These cells remain metabolically active as revealed by their healthy nuclei and are not sloughed off at the surface. the lateral cell membranes of these cells are fused together forming a continuous barrier which plays important role in water proofing the skin. In addition the polygonal cells in the most superficial layer also undergo the process of mucogenesis synthesizing sulphated acid mucopolysaccharides which may ultimately form a part of the contents of the protective extracellular cuticular coat.

A histochemical study of the epidermis of the climbing perch, Anabas testudineus (Anabantidae, Pisces).

A histochemical investigation has been made to localize and characterize various lipid, protein, carbohydrate and enzyme constituents present within the different cell types of the epidermis of Anabas testudineus. The polygonal cells contain glycogen, the amount of which gradually increases as the cells move towards the surface until they reach the most superficial layer where the amount of glycogen slightly decreases indicating the metabolically active state of these cells. The basal cells, which frequently undergo cell proliferation, contain no glycogen. The polygonal cells give strong reactions for SDH, alkaline phosphatase, cholesterol esters and nonsulphated acid mucopolysaccharides, moderate reactions for acidic lipids, phospholipids and free cholesterol and weak reactions for neutral mucopolysaccharides, protein bound NH2 groups, mucoprotein, tyrosine, tryptophan and cysteine bound sulphydryl groups. These cells in the outermost layer give stronger reactions for acidic lipids, phospholipids and cholesterol esters and weaker reactions for SDH and alkaline phosphatase activities. The above findings reveal that the polygonal cells remain metabolically active throughout the epidermis. The mucous cells are numerous and secrete mixture of neutral mucopolysaccharides, sulphated acid mucopolysaccharides and nonsulphated acid mucopolysaccharides. The contents of the sacciform granulated cels are mainly proteins. A thick coat of slime over the body surface containing mucopolysaccharides, lipids and proteins is important in keeping the skin moist and may facilitate the survival of the fish while it is on land. The melanophores in the epidermis may playing important role in preventing the colinization by parasites, fungi and bacteria over the body surface, act as macrophages.

Chondrosarcoma of the breast. Report of two cases.

The clinical and pathologic findings in two cases of pure chondrosarcoma of the breast are presented. As with other sarcomas, both tumors were large (5.5 and 25 cm in greatest diameter), did not invade the skin or axillary lymph nodes, and occurred in women more than 40 years old. Infiltrating margins, cellular atypism, and high mitotic rates adversely affected the prognosis.
