Use of an enzyme-linked immunosorbent assay to monitor the control of Staphylococcus aureus mastitis.

Conventional culture methods were used to evaluate the ability of an ELISA to identify Staphylococcus aureus IMI. The test was 96% accurate; sensitivity was 90%, and specificity was 97%. The test was used to screen preserved milk samples rapidly in 10 cooperator herds. Prevalence of IMI was greater than 10% in 6 herds at the first test. Average prevalence of cows scoring +2 (suspect) and +3 (positive) was 12.6%. Prevalence declined during the 12-mo study. Incidence of new IMI decreased from 7.9% at 6 mo to 3.6% at 12 mo. Rinsing teat cup liners with a 25-ppm iodophor or 100-ppm chlorine solution reduced the presence of S. aureus on the milking machine liners by 97%. Elevated scores were correlated with increases in lactation number. Milk antibody concentrations changed quadratically with increasing SCC. The SCC increased as milk antibody concentration increased. In 38 dairy herds, bulk tank antibody tests reflected herd prevalence of S. aureus infection. The average prevalence was 15.0% in 87 herds in which all lactating cows were tested.

The effect of cimetidine and ranitidine on the duration of action of succinylcholine.

A recent study suggested that cimetidine prolongs the duration of action of succinylcholine (SCh). We examined this reported interaction, and investigated whether it is the result of inhibition of plasma cholinesterase (PChE). We also studied the effect of ranitidine on the duration of action of succinylcholine. Thirty patients were randomly assigned to three groups to receive oral cimetidine 300 mg or ranitidine 150 mg or no H2-antagonist the night before surgery and again 1-2 hours prior to induction of anesthesia. Succinylcholine (1.5 mg/kg i.v.) was given to facilitate tracheal intubation after induction of anesthesia; neuromuscular function was monitored by evoked compound electromyogram. Pretreatment blood samples were analyzed for PChE activity. PChE activity and levels of appropriate H2-antagonists were measured in preinduction blood samples. No significant effect of either cimetidine or ranitidine on the duration of action of SCh or PChE activity was demonstrated. We conclude that H2-antagonists administered preoperatively do not prolong the duration of action of succinylcholine or alter PChE activity.

Analysis of errors in parameter estimation with application to physiological systems.

The accuracy of parameter estimation applied to physiological systems is analyzed. The method of analysis is applicable to procedures utilizing minimization of squared output error and a nonlinear dynamic system model. Three major sources of estimation error are described: 1) measurement error, 2) modeling error, and 3) optimization error. Measurement errors affect values used for the system output, the model input, and nonestimated parameters of the model. Modeling errors are due to failure to adequately describe the structure of the system and to numerical errors that occur in the digital computer solution of the model equations. Linearization by use of Taylor series expansions in the region of the nominal solution is used to obtain an expression for the covariance matrix of the parameter estimates in terms of the covariance matrix of each error source. The analysis is applied to the example of cardiac output estimation from respiratory measurements. The results demonstrate that an analysis of system identifiability is not sufficient to ensure usable estimates and that systematic error analysis is essential for assessing the usefulness of parameter estimation techniques.

Comparison of R-R interval prediction models.

Changes in the sequence or pattern of R-R intervals is an integral part of electrocardiogram detection of most clinically important arrhythmias. Moreover, there is inherent interest in these patterns and their method of study as basic physiological phenomena. Prediction of R-R interval changes based on past history and detection of departure from an established pattern have been utilized in automated monitoring strategies and algorithms. The studies described her further clarify the usefulness of Markov processes for this purpose, compare the accuracy of linear and nonlinear regression models, and indicate some advantages of both.
