[Cerebokrast as a corrector of postischemic phenomena in acute transient cerebral ischemia].

Acute cerebral ischemia in cats (both carotid arteries occlusion during 30 min after permanent occlusion of both vertebral arteries) was accompanied by postischemic hypoperfusion and hypo-oxygenation of the cerebral tissue. Intravenous infusion of cerebrocrast (1 micrograms.kg-1.min-1 during 60 min) prevented manifestation of the postischemic phenomena. Antihypoxic effect of cerebrocrast involved the cerebral blood flow increase, brain oxygen consumption lowering and Hb-O2-affinity decrease.

[Antioxidant effects of antihypoxic drugs in cerebral ischemia].

Cerebral ischemia in rats (both carotid arteries occlusion) during 30 min, 3 hours and recirculation (1 hour) after ischemia (30 min) stimulated diene conjugates and fluorescent products accumulation in brain tissue. Intraperitoneal injection of sodium hydroxybutyrate (100 mg/kg), bemitil (50 mg/kg), ethomersol (50 mg/kg) reduced brain lipid peroxidation and did not yield in this respect to emoxypin (5 mg/kg). In contrast to emoxypin, sodium hydroxybutyrate, bemitil and ethomersol had no antiradical activity.

[The evaluation of the efficacy of antihypoxic agents lowering hemoglobin oxygen affinity in acute cerebral ischemia].

Influence of natrii hydroxybutyrate (100 mg/kg), ascorbate (100 mg/kg), cavinton (5 mg/kg), bemitil (50 mg/kg), ethomersol (50 mg/kg) on Hb-O2 affinity and cortex PO2 after both carotid artery occlusion in rats was investigated. Correlation (r-0.87; P less than 0.05) between lowering of Hb-O2 affinity and antihypoxic effect was demonstrated in the line of these drugs.

[Effects of ethomersol on cerebrovascular blood circulation and oxygen metabolism of the brain in acute transient ischemia and recirculation].

Intravenous infusion of ethomersol (50 mg/kg during 60 min) decreased cerebral tissue hypoxia at the end of acute cerebral ischemia in cats (30-min occlusion of both carotid and both vertebral arteries). In recirculatory period ethomersol prevented the postischemic hypoperfusion and especially hypo-oxygenation. Ethomersol-induced reduction of Hb-affinity to O2 was demonstrated.

[Antihypoxic and antioxidative properties of bemitil].

The authors discovered antihypoxic properties of the bemitil (pretreatment injections 50 mg/kg intraperitoneally) in the experiments on rats with the circulatory or hypoxic hypoxia. There was limitation of pO decrease and diene conjugates and Schiff bases production increase with the drug in the circulatory hypoxia conditions. Bemitil restricted malondialdehyde accumulation in the rat brain homogenate under the activation of free radicals processes. In the mitochondrial suspension incubation similar effect of the medicine was accompanied with limitation of organelle degradation. Bemitil showed no antiradical activity.

[Effect of dibazol on cerebral blood supply and EEG in experimental intracerebral hemorrhage].

A hemorrhage into the right internal capsule in vigilant cats is accompanied by a drop of focal cerebral blood flow in the sensomotor cortex, in the thalamus and reticular formation of the middle brain of both hemispheres, an appearance of slow-wave high amplitude in the EEG activity and in some cases of convulsive discharges. Such changes of the blood flow and bioelectrical activity are preserved in survived cats after 24 hours following hemorrhage. An intracarotid singular injection (Img/kg) and especially a 30 minute infusion of 0.5% solution of dibasol brings about in most of the cases an increase of the blood flow in the studied brain structures, and is accompanied by a partial normalization of bioelectrical activity.

[The mechanisms of the ethomersol correction of postischemic hypoperfusion].

In rat experiments during brain ischemia (30-min carotid artery occlusion), ethomersol administered intraperitoneally in a dose of 50 mg/kg before occlusion or at the end of ischemia eliminated postischemic hypoperfusion. The effect of the drug was due to its spasmolytic and antiaggregatory activities. An analysis of the vasodilating action of ethomersol revealed its capacity to block potential-dependent calcium channels and partially intracellular calcium mobilization when the adrenergic and serotoninergic receptors were activated. The antiaggregatory activity of the drug appeared as inhibited platelet aggregation, which was induced by ADP, serotonin, arachidonic acid, thrombin, and as enhanced antiaggregatory activity of the vascular wall.

[Use of isothiobarbamin in the correction of disorders of the vital activities of the brain in intracerebral hemorrhage].

Isothiobarbamin injected intravenously to cats in the phase of circulatory hypoxia 1-2 days after intracerebral hemorrhage improves the functional activity, blood supply, oxygen supply to the brain, prevents excessive accumulation of lactate in the brain tissue. The drug exerts the stimulating effect on the disordered local blood flow: the blood supply to the ischemic areas increases, and that of the hyperemic ones decreases. The mechanism of the vasodilative effect of isothiobarbamin is due to inhibition of Ca2+ delivery through the potential-dependent and serotonin-activated channels of the smooth muscle plasmic membrane. The drug activity increases with a rise of the initial vascular tone.

[Effect of glycerin on several indices of intracranial hemodynamics and the electroencephalogram of wakeful cats with experimental intracerebral hemorrhage].

An intravenous infusion of glycerin to cats 24-48 hours after an intracerebral hemorrhage contributes to normalization of the EEG, Intensifies the local blood flow in the visual cortex, thalamus and reticular formation of the midbrain in both hemispheres (predominantly in the regions of the right affected hemisphere). The drug produces a fall in the absolute values of the intracranial pressure without its secondary rise after infusion is discontinued and causes a moderate rise of the perfusion pressure. The intravenous infusion of glycerin is more effective than its single injection.
