Liver cuts were performed on 21 Mini- Lewe pigs and then closed with suture in one case, and by ultrasonic welding of Fimomed - Gelaspon conglomerate in the other. Macroscopic assessment in situ and the histological examination of cut out liver areas regularly showed perifocal inflammatory peritoneal reactions and abscess formation for the sutured regions. From a histological point of view the liver tissue in the regions subjected to ultrasonic treatment reacted more strongly than after suture, particularly in the form of lobule structure disturbances, focal hepatitis, hepatic duct proliferation and destruction, and cholangitis. All of these were limited to the immediate vicinity of the conglomerate with its fibrous demarcation, which remained identifiable until the end of the experiment after a year and a half. The experiments resulted in the sucessful closure of the liver wounds in all cases where ultrasonic welding of Fimomed - Gelaspon compound was used. Application to human medicine is, however, not yet justified.
Particulate membrane fractions from Volvox carteri catalyze the transfer of mannose from GDP-mannose to dolichyl diphosphate-[14C]chitobiose to form lipid-linked oligosaccharides up to a dolichyl diphosphate-chitobiose-(mannose)5 structure. Mannosylation of the chitobiosyl lipid requires divalent cations and detergents as solubilizing agents. Depending on the nature of the detergent, the oligosaccharide pattern differs markedly: With deoxycholate or the zwitterionic detergent 3-14 a lipid-linked trisaccharide accumulates. The nonionic Triton X-100, however, gives rise to a spectrum of compounds up to a heptasaccharide. Enzyme digestion of the tri- and pentasaccharide structure, obtained after mild acid hydrolysis of the corresponding [14C]glycolipids, revealed that the first mannose is bound via a beta-glycosidic linkage to the chitobiosyl core, whereas the outer mannose residues are linked as alpha-mannosides. Our studies indicate that, in agreement with recent findings in other organisms, the innermost alpha-mannosidic residues are donated directly from GDP-mannose. The structure of oligosaccharides synthesized by Volvox membranes is thus consistent with results from other eucaryotic species, suggesting a common pathway of N-glycosylation of glycoproteins.

The development of a new enzyme-linked immunoassay for pregnancy-associated alpha-2 glycoprotein (alpha 2-PAG) has provided an opportunity to reassess the value of this protein as a tumour marker. Serum samples from 800 healthy individuals and patients with various benign and malignant diseases were assayed. There was a very wide range of alpha 2-PAG levels in normal females (1- greater than 100 mg/l), and although the levels found in normal males were lower and better defined, this intrinsic variation between individuals makes a single determination in a tumour-bearing patient meaningless. Also, the levels of alpha 1-PAG in patients with advanced cancer were not significantly different from levels in localized cancer, benign disease or even healthy controls, and furthermore, levels before and after successful cancer treatment did not show a significant change. Our results therefore indicate that alpha 2-PAG is unsuitable for use as a tumour marker as there was no apparent relationship between the levels of alpha 2-PAG and either tumour burden or response to treatment.
A conglomerate of homologous bone-chips and n-butyl-cyanoacrylate fixes the fragments in the ultrasonic osteosynthesis. The sound pressure leads to the forced penetration of the monomer into preformed gaps of the bone surface (Volkmann and Haversian canals) up to maximum 300 microns during the polymerisation forced by ultrasound. A toothing effect between the conglomerate and the cortex takes place in this way, this one leads to a higher mechanical resistance and to specialities in the regenerative process. The term welding is founded scientifically by the mechanism of the connecting formation. On the other hand the application of the ultrasound is sensible in order to force the polymerisation.

Absorption of ultrasound leads to reversible or irreversible changes in biological tissues in dependence on the sound intensity. The ultrasound causes under appointed conditions in skin, muscles, vessels, nerve tissue, and bone histopathological changes in a different scale which must be observed in the ultrasonic surgery. 200 tissue specimens from 40 guinea pigs were taken from defined localisations after an ample ultrasound exposition and were evaluated microscopically by hematoxylin-eosin staining technique. Skin and vessels showed very vehement reactions. Intact osseous tissue reacted with periosteal callus formation. Practical conclusions can be draws for the ultrasonic surgery by the discussion on histopathological changes.

The effects of various doses of synthetic neurotensin on regional blood flow in different tissue layers of the stomach, small bowel, colon, pancreas, brain, kidneys, adrenal gland, and heart of six dogs was studied using an isotope microsphere technique. Infusion of high doses (20, 40 pmol/kg . min(-1)) of exogenous synthetic neurotensin caused an increase of blood flow in the "muscularis" of duodenum, jejunum, ileum, and colon. Neurotensin infused in a dose (2.5 pmol/kg . min(-1)) raising neurotensin plasma levels to concentrations comparable to those observed after a meal caused an increase of blood flow in the muscular layer in ileum. Our results suggest that one of the physiologic actions of neurotensin may be the regulation of blood flow in the muscular layer of the ileum.
