[Combination chemotherapy with MOPP and intrathecal methotrexate in children with brain tumors].

Five children with brain tumors, who had had no prior chemotherapy, were treated with combination chemotherapy including nitrogen mustard, vincristine sulfate, procarbazine, prednisolone (MOPP) and intrathecal methotrexate. Four had progressive tumor or metastatic deposits, (one medulloblastoma, one malignant choroid plexus papilloma, and two were not explored) but responded to chemotherapy. One child with medulloblastoma underwent resection, which was considered to be macroscopically radical, and demonstrated objective improvement of neurological findings after chemotherapy. Toxicities were not severe enough to require termination of the chemotherapy.

Effects of cryosurgery in experimental carcinoma on lectin binding and keratin distribution.

Histochemical alterations of lectin binding and keratin distribution in experimental carcinomas of the hamster cheek pouch were obtained following cryotreatment. Cryotreated carcinoma cells showed a characteristic reduction in lectin binding and keratin staining shortly following cryosurgery. Tumor tissue, on the 2nd and 3rd days after cryotreatment, displayed destruction and necrosis with almost a complete loss of lectin binding and keratin staining. The remaining neoplastic cells located in the deeper layer showed positive reaction for both lectin binding and keratin, which is indicative of tumor recurrence. Histochemical staining of lectin binding and keratin proteins were useful markers in cryotreated tumor cells to identify either destruction and necrosis or vital activity of neoplastic growth.

Identification of vascular system in experimental carcinoma for cryosurgery--histochemical observations of lectin UEA-1 and alkaline phosphatase activity in vascular endothelium.

Histopathologic and histochemical changes in experimental carcinomas following cryotreatment were observed to detect alkaline phosphatase (ALP) activity and UEA-1 lectin binding. Experimental carcinomas were induced in the hamster cheek pouch by topical application of 0.5% DMBA acetone solution twice a week. The cryoprobe at -60 degrees C was directly attached to the tumor surface for 90 sec. Histochemically, the tumor tissue following cryotreatment was completely destroyed in the surface area by the direct freezing and such cryonecrotic tumor tissue lacks stainability. Soon after cryotreatment and before cryonecrosis takes place, it has been observed that there is an intense dilatation of capillary vessels. Histochemically, high ALP activity was limited to capillary endothelium and to inflammatory cells. Lectin UEA-1 staining was usually found in both normal and neoplastic epithelial cells which were confined to capillary vessels. At 1 to 3 hr after cryotreatment, lectin UEA-1 binding was also positive in dilated endothelial cells of the frozen tissue as well as viable remaining neoplastic epithelia by freezing. ALP activity and UEA-1 binding disappeared in capillaries of cryonecrotic area in tumor tissues. Those findings suggest that biologic membrane changes in capillary endothelium of tumor stroma occur following cryotreatment.

Are cat lingual muscles dually innervated by the hypoglossal nerve and facial nerve?

The origins of the somatic motor nerve innervating the cat lingual muscles were studied using the horseradish peroxidase (HRP) method. Following HRP injection into the lingual muscles, labeled neurons were found not only in the hypoglossal nucleus but also in the facial nucleus, ipsilateral to the injection side. HRP-labeled neurons in the facial nucleus were principally observed in the ventromedial and ventrolateral divisions of the nucleus. This study suggests that cat lingual muscles are innervated by both hypoglossal motoneurons and some of the motoneurons in the ventromedial and ventrolateral divisions of the facial nucleus.

Histologic and histochemical changes in experimental carcinomas following cryosurgery.

Histologic and histochemical changes in experimental SCCs following cryosurgery were reported. Effects of cryosurgery on SCCs of hamster cheek pouch were classified into 3 zones; the superficial zone was the cryodestructive layer on which the cryoprobe had been attached directly to the SCC; the zone beneath this layer was the indirect cryodestructive layer; the 3rd zone was unaffected tissue in which the critical low temperature was never reached. In the superficial cryodestructive layer, tumor cells were destroyed completely and bleeding was found in the stroma. In the zone beneath it, neoplastic cells also showed morphologic and enzymatic changes indicating incomplete cellular destruction; dilatation of capillary vessels was also found. LDH isozyme pattern displayed a high level of LDH 5 in the non-treated SCC, and following cryosurgery, the high level of LDH 5 decreased and revealed an approximately normal LDH 5 pattern.

Studies on deficiency of Schwann cell basal lamina and deformation of collagen fibres induced by beta-aminopropionitrile in cultures of developing rat peripheral neurons.

The morphological analysis in culture of superior cervical ganglia and dorsal root ganglia showed the following: (1) collagen fibres external to the neurite bundles were larger than those inside the neurite bundles in both control and BAPN-treated cultures; (2) there was much more irregularity of shape in external than in internal fibres in dorsal root ganglia with BAPN; (3) there was reduced basal lamina formation by Schwann cells in both types of ganglia (Okada & Bunge, 1981); (4) basal lamina formation in the superior cervical ganglion was reduced as the days of treatment with BAPN increased (from 94.7% with one day of treatment to 66.7% with 8 days of treatment; (5) in the dorsal root ganglion preparation, the myelin internode was shorter in BAPN (105.1 micron +/- 34.5 micron) than in control media (127.8 micron +/- 32.8 micron: P less than 0.001); (6) there was no significant difference in thickness of myelin between controls (2.9 micron +/- 0.9 micron) and those treated with BAPN (2.7 micron +/- 0.7 micron: 0.05 less than P less than 0.1); (7) the node of Ranvier was more elongated in BAPN (5.1 micron +/- 4.1 micron) than in control media (2.5 micron +/- 0.8 micron; P less than 0.01). The relationship between connective tissue abnormalities caused by BAPN and of the basal lamina formation of myelin-forming cells affected by BAPN is discussed.

[Myelin formation of the dorsal root ganglion cells of the senile rat in long-term culture].

Using the dorsal root ganglia (DRG), long-term cultures were done in neonatal (control) and senile (experimental) rats. The morphological analysis on myelin sheath formation was carried out by light microscopy, electron microscopy and image analyzing apparatus. The results are as follows: The neurites of senile DRG cells appeared 7 days later than the neurites of neonatal DRG cells. While the myelinations were observed in neonatal DRG culture by the third week, myelinations of senile DRG were also formed 7 days later. The myelin sheaths in both neonatal and senile DRG cultures looked much like those which were formed in vivo, but the number of myelinations in senile DRG culture was fewer than that of the neonatal DRG culture. In observations by electron microscopy, myelin lamellae numbered about 20 in both neonatal and senile DRG cultures with normal-appearing axon structures as seen in vivo. The discontinuities in the external lamina of Schwann cells were not observed in both the neonatal and senile DRG cultures. In the morphological analysis of neonatal and senile DRG cultures, the internodal length, diameter of myelin sheath and the length of nodal gap (distance between the rounded end of the compact myelin) were measured by image analyzing apparatus (MAGISCAN-I, Joyce Loebl LTD.) with Sudan black B stain. Although the internodal length of myelin sheath in senile DRG culture (144.9 +/- 40.7 microns) proved to be significantly longer than the neonatal DRG culture (131.6 +/- 26.2 microns) (p less than 0.01), the diameter of myelin sheath showed no significant difference between neonatal (2.9 +/- 0.7 micron) and senile (2.8 +/- 0.7 micron) DRG.(ABSTRACT TRUNCATED AT 250 WORDS)

Observations on the fine structure of the phrenic nucleus in the cervical spinal cord of the cat with special reference to its dendritic bundles.

The structure of the cat phrenic nucleus in the cervical spinal cord was studied by light and electron microscopy. Neurons of this nucleus are medium sized and fusiform and are arranged longitudinally. The most unique feature of this nucleus is the extremely well organised longitudinal dendritic bundles. In the dendritic bundles there are dendrodendritic casual appositions without specialised structures and with desmosome-like contacts. Gap junctions, however, are not found at the site dendrodendritic contact. Neurons of the phrenic nucleus possess many axodendritic synapses whereas axosomatic synapses are infrequent. In most axon terminals there are round, clear, synaptic vesicles, but on rare occasions terminals containing either cored or flat vesicles are observed. The significance of these features of the nucleus, especially of the large dendritic bundles, is briefly discussed.

Ribonucleotide reductase and thymidine kinase activities in various cultured cell lines derived from hematologic malignancies.

The activities of ribonucleotide reductase and thymidine kinase, and the thymidine incorporation rate were measured in 16 cultured human hematologic malignant cell lines with different cell proliferation rates. Thymidine kinase activity was significantly higher in myeloid and monocytoid cell lines than in other cell lines, but ribonucleotide reductase activity presented as CDP reductase activity was similar in the different cell lines. The ratio of thymidine kinase to CDP reductase activity was high in monocytoid cell lines. A close correlation was found between the cell proliferation rate and CDP reductase activity, but not thymidine kinase activity or the thymidine incorporation rate. The ratio of thymidine kinase to CDP reductase activity was high in slowly growing cell lines and low in rapidly growing cell lines. These results indicate that in cultured human malignant cells a high potential for proliferation may depend mainly on the de novo pyrimidine pathway of DNA biosynthesis.

[Noninvasive and quantitative evaluations of the severity of isolated pulmonary valvular stenosis using two-dimensional pulsed Doppler echocardiography].

The severity of isolated pulmonary valvular stenosis (PS) was evaluated noninvasively and quantitatively using two-dimensional pulsed Doppler echocardiography (2D-PDE). The subjects consisted of 17 patients with PS diagnosed by cardiac catheterization (3 to 15-year-old with a mean age of 7.9 years) and 28 healthy children (3 to 12-year-old with a mean age of 7.2 years). Flow signals by 2D-PDE were recorded in the right ventricular outflow tract just below the pulmonary valve and in the main pulmonary artery. The following three parameters were measured from the flow signals in the right ventricular outflow tract: the right ventricular pre-ejection period (RPEP) from the onset of the QRS in the electrocardiogram to the beginning of RV ejection in 2D-PDE, the acceleration time from the onset of RV ejection to peak velocity (AcT), and the right ventricular ejection time (RET) from the onset to the end of right ventricular (RV) ejection. Acceleration time index (AcT/RET), and systolic time intervals including RPEP/RET, RPEP/square root RR and RET/square root RR in PS were compared with those of the controls. The correlations between the above-mentioned parameters and pressure gradients and RV systolic pressures in PS were examined. Data were expressed as mean value +/- SD. AcT/RET ranged from 0.37 to 0.53 (0.45 +/- 0.04) in the controls and from 0.54 to 0.76 (0.62 +/- 0.07) in PS. The mean AcT/RET was significantly greater in PS than in the controls (p less than 0.001). There was a highly positive correlation of AcT/RET with pressure gradients (r = 0.94) and RV systolic pressures (r = 0.93). RPEP/RET ranged from 0.32 to 0.47 (0.37 +/- 0.04) in the controls and from 0.24 to 0.35 (0.29 +/- 0.03) in PS, and the mean was significantly lower in PS than in the controls (p less than 0.001), and this was caused by the short RPEP and prolonged RET. RPEP/square root RR ranged from 102 to 160 msec (122 +/- 16 msec) in the controls and from 86 to 136 msec (107 +/- 16 msec) in PS, and the mean was significantly shorter in PS (p less than 0.05). RET/square root RR ranged from 300 to 345 msec (323 +/- 13 msec) in the controls and from 308 to 417 msec (363 +/- 27 msec) in PS, and the mean was significantly longer in PS (p less than 0.001). The pressure gradient and RV systolic pressure in PS did not correlate with RPEP/RET, RPEP/square root RR and RET/ square root RR.4+ 102 to 160 msec (122 +/- 16 msec) in the controls and from 86 to 136 msec (107 +/- 16 msec) in PS, and the mean was significantly shorter in PS (p less than 0.05).(ABSTRACT TRUNCATED AT 400 WORDS)
