The effect of protease pretreatment on the demonstration of hepatitis-B-surface antigen by immunofluorescence (IF) and the unlabeled peroxidase-antiperoxidase technique (PAP) in conventionally processed (formalin-fixed, paraffin-emmbedded) liver biopsy material was quantitatively assessed by microphotometry. Protease digestion significantly enhances the intensity of specific staining by both methods, and, in addition, suppresses non-specific background fluorescence. The sensitivity of the immunomorphologic test is significantly enhanced, and antigen in low amounts, for example hepatitis-B-surface antigen associated with liver cell membrane ('membrane' staining), is easily detected.
The influence of carbonyl iron treatment on different human mononuclear cell subsets was investigated. There is evidence that, in addition to removal of phagocytic cells, lymphocytes are depleted, B cells being more affected than other subpopulations.

In 241 paraffin-embedded liver-biopsies of HBSAg-positive, voluntary blood-donors hepacellular HBSAg was demonstrated with the immunomorphologic peroxidase-antiperoxidase (PAP)-method. In all sections, HBSAg-positive liver-cells were estimated, their distribution-pattern was assessed and different types of cytoplasmatic localisation of HBSAg were described. Despite high sensitivity of the PAP-method, 24% of seropostive cases were histologically negative. A correlation of HBSAg-content and histologic diagnosis was lacking. An association of high HBSAg-content with non-aggressive liver-disease and of low HBSAg-content with aggressive liver-disease in a single biopsy could not be found in our material in contrast to the literature. 34 patients were followed up through 4-6 years. Constant or improved liver-histology was associated with increased, deterioration with decreased HBSAg-tissue-content.
Erythrocytes of different species (chicken, sheep, man, mouse, rat, guinea pig) except rabbit erythrocytes strongly adhere to the marginal zone of mouse spleen follicles in frozen sections. This adherence reaction (AR) is not restricted to red blood cells but is also observed with human lymphocytes. Pretreatment of the tissue sections with trypsin, mercaptoethanol, periodate, chloroform/methanol, acetone, and heating the sections abolishes AR whereas neuraminidase (VCN) treatment of the sections has an amplifying effect. AR is inhibited by preincubation of the neuraminidase- or untreated sections with neuraminic acid (NA). Treatment of the erythrocytes with VCN completely abolishes AR whereas treatment with other enzymes (hyaluronidase, collagenase) is ineffective in this respect. Determination of NA in the erythrocyte membrane before and after VCN treatment reveals a positive correlation between the amount of NA and AR. Rabbit red blood cells have the lowest NA content in their membranes and, in addition, there is little effect of VCN treatment in further reducing it. It is possible that a lectin-like substance is responsible for AR. The biologic significance of AR is hypothetical, but since AR occurs in an area of the spleen playing a role in antigen trapping it is conceivable that this trapping may be mediated by interaction of NA and NA receptor(s).

Treatment of dewaxed sections of formalin-fixed paraffin embedded material with 0.1% pronase for 15 min at 37 degrees C prior to the performance of the unlabelled antibody-enzyme (PAP) procedure greatly enhanced its sensitivity. This was shown by detection of immunoglobulin within plasma cells in chronic gastritis specimens.
Pulmonary emphysema was induced in 8 bastard dogs by intratracheal instillation of Papain. Development of emphysema was documented by pulmonary function tests. The emphysematous animals underwent single lung homotransplantation. Postoperative bronchospirometric measurements revealed that up to the 6th week after transplantation there is no evidence of serious ventilation perfusion inbalance.

Homologous articular cartilage without subchondral bone instead of the usual osteochondral grafts was transplanted in sheep. The transplants were fixed to the host by a biological tissue adhesive based on fibrinogen clotting. Light- and electronmicroscopic studies showed viable transplants up to 7 months and a stable bony union with the host through endochondral bone formation. A additional external fixation of the joint after transplantation or the use of a conventional cyanoacrylate tissue adhesive failed to achieve positive results. The relations of these preliminary findings for restorative techniques of articular cartilage defects are discussed.

Twenty-three lymphomas of the eye, eyelid, and orbit were chosen study from biopsy material of the Department of Pathology, University of Kiel, and the Lymph Node Registry in Kiel. There were 12 malignant non-Hodgkin's lymphomas and 11 benign lymphomas. Catamnestic examination confirmed the histologic diagnosis in all 11 cases of benign lymphoma. Eleven of the malignant lymphomas represented immunocytomas and were therefore a low-grade malignancy. In contrast, there was only one case of malignant lymphoma of high-grade malignancy, which was diagnosed as centroblastic lymphoma. The immunocytoma was retrobulbar in one third of our cases and conjuctival in only 2 cases, whereas benign lymphoma had developed in the conjunctiva in 7 of 11 cases. Immunocytoma represents a tumor composed of lymphocytes and plasma cells or plasmacytoid cells. PAS-positive globular inclusions are usually found in the nucleus and/or cytoplasm of the plasma cells or plasmacytoid cells.
