Cyclic peptides. XIX. Synthesis, conformation, and biological activity of cyclo(-Pro-Val-Pro-Val-) with L-L-L-L and L-D-L-D sequences.

cyclo(-L-Pro-L-Val-L-Pro-L-Val-) (1L) and cyclo(-L-Pro-D-Val-L-Pro-D-Val-) (1D) were synthesized by the conventional method for peptide synthesis. Conformations of 1L and 1D in solution were studied. Compound 1L has a cis-trans-cis-trans backbone conformation with C2 symmetry in CDCl3. This conformation is slightly different from that in crystalline state and in DMSO-d6 solution. Compound 1D has a cis-trans-cis-trans conformation in DMSO-d6 and an all-trans conformation in trifluoroethanol-d3. Compound 1L retarded stem-growth of rice seedlings and, in contrast, compound 1D promoted root-growth of rice seedlings.

[Clinical value of the sequential study of the uric acid level in the CSF in patients with postoperative meningitis].

In this study we discuss the clinical value of the sequential measurement of the CSF uric acid and CSF cells in patients with postoperative acute meningitis. CSF was investigated for uric acid and cells in 10 cases with postoperative meningitis and 12 cases without it for the control group. The results were as follows; The CSF uric acid level was markedly increased in direct proportion to neutrophilia in the CSF in the acute stage of postoperative meningitis. Some cases with postoperative meningitis had biphasic increasing pattern of the CSF uric acid level. In 12 cases without postoperative meningitis the CSF uric acid level had been progressively reduced and then normalized until the fourth day after operation. The factors contributing to the increase of the uric acid in CSF under the condition of postoperative meningitis were thought to be 1) increased permeability of the blood-CSF barrier, 2) neutrophilia in the CSF and increase of the nucleic acid due to it, 3) increase of hypoxanthine, xanthine or xanthine oxidase activity in the central nervous system and 4) dysfunction of the CSF dynamics.

Sequence specificity of ozone-degradation of bases in supercoiled plasmid DNA.

It was found that ozone reacted preferentially with thymine and guanine residues located in the specific region in pBR322 DNA. The sequence analysis of the region including the cleavage site produced by ozonization of ccDNA showed that ozone-modification proceeded in the single stranded region formed by cruciform-formation in supercoiled DNA.

Effect of terminal base pairs on the CD spectra of oligodeoxynucleotide duplexes.

Oligodeoxynucleotide duplexes having terminal C:G base pairs and recognition sequences for restriction endonucleases Bgl II and Sau 3Al were synthesized by the solid or liquid phase phosphotriester approach. Among them, the self-complementary oligonucleotides with C or CC at the 5'-end and G or GG at the 3'-end exhibited unusual CD spectra in contrast to those of the usual B-DNA type CD spectra of the oligonucleotide with G or GG at the 5'-end. The spectra are different to those of the Z-DNA, and regarded to the terminal base effect of the C:G pairs.

Synthesis of deoxyoligonucleotides containing 7-deazaadenine: recognition and cleavage by restriction endonuclease Bgl II and Sau 3AI (nucleosides and nucleotides Part 55).

Deoxydecanucleotides having a recognition sequence of Bgl II and Sau 3AI, and their 7-deazaadenine analogs were synthesized. The decanucleotides containing 7-deazaadenine in place of adenine were partially resistant to the hydrolysis by Sau 3AI and strongly resistant to that by Bgl II. A new hypothesis on the mode of recognition and cleavage of specific nucleotide sequences by Bgl II, recognizing one strand and cleaving the other strand, is presented.

Acute leukemia with two cell populations of lymphoblasts and monoblasts.

A 46-year-old man had acute leukemia with two cell populations of lymphoblasts and monoblasts (L1 and M5-b in FAB classification, respectively), which were characterized by morphological, cytochemical and cell marker studies. At the time of diagnosis about 80% blasts were terminal deoxynucleotidyl transferase (TdT) positive lymphoid cells, while the rest were TdT negative monocytoid cells. After induction chemotherapy of vindesine and prednisolone for 15 days, almost all blasts were TdT negative monocytoid cells. Therefore, an additional course of the chemotherapy with the protocol for acute nonlymphocytic leukemia was given and one month later the patient achieved complete remission.

A sensitive and specific radioimmunoassay for 1-beta-D-arabino-furanosylcytosine.

In order to determine the blood level of 1-beta-D-arabinofuranosylcytosine (Ara-C), an antileukemic agent, a sensitive and specific radioimmunoassay (RIA) system using anti-Ara-C serum, [5-3H] Ara-C and a dextran-coated charcoal method has been developed. The anti-Ara-C serum obtained from a guinea pig was hardly cross-reactive with 1-beta-D-arabinofuranosyluracil (Ara-U), tetrahydrouridine (THU) and other Ara-C analogues. The RIA system for Ara-C could detect concentrations as low as 60 pg/ml in plasma. Average of the intra- and inter-assay variancies at 5, 10, 20 ng/ml were 4.3% and 5.6%, respectively. Ara-C in blood samples obtained from human patients who orally received N4-palmitoyl-1-beta-D-arabinofuranosyl-cytosine (PL-AC) was determined by the present RIA system.

Ergopeptine-sensitive calcium-dependent protein phosphorylation system in the brain.

We studied a protein phosphorylation system that is regulated by the dopamine-mimetic ergot bromocriptine. Bromocriptine was found to inhibit selectively the endogenous phosphorylation of a threonine residue(s) in 50,000- and 60,000-dalton proteins in a synaptosome fraction. The bromocriptine-sensitive phosphorylation is stimulated by calcium and by calmodulin, and occurs predominantly in the brain. The inhibitory effect of bromocriptine was not mimicked by 3,4-dihydroxyphenylethylamine or by any of the neurotransmitters and related agents tested, but was mimicked, although less effectively, by other ergots that contain peptide moieties. In the hippocampus, the brain region with the highest content of the 50,000- and 60,000-dalton proteins, the ergopeptine-sensitive protein phosphorylation appears to be localized to interneurons or cell bodies whose axons synapse outside the hippocampus. The results raise the possibility that some of the bromocriptine- and ergopeptine-induced pharmacological effects in the CNS may be mediated by the inhibition of the calcium/calmodulin-dependent phosphorylation of these specific proteins.

Further evidence for the presence of subepithelial nerve cells in the rat ileum--an immunohistochemical study.

Earlier we observed electronmicroscopically an unidentified but neuron-like subepithelial cell in the crypts of the rat ileum. We have now studied some immunocytochemical characteristics of similar cells in the fluorescence microscope by indirect immunofluorescence. Three markers for neuronal tissues were used: 1) Monoclonal Thy-1-antibodies, demonstrated previously to recognize surface antigens of thymus-derived cells and nervous tissue; 2) Tetanus toxin, which binds to the GT1 ganglioside receptor of nerve cells, and to some other neuronal receptor; and 3) Anti-Protein I-IgG, which is monospecific for Protein I located in synaptic vesicle membranes. The various antisera used for secondary incubations all reacted with leucocytes and other connective tissue cells in the villous cores. When the specific primary antisera of Thy-1 and Protein I, and tetanus toxin, were included, a small number of spindle-shaped cells with two processes, extending just beneath the basal lamina of the epithelium, was observed. These cells had a similar location and appearance as the neuron-like cells observed electronmicroscopically and may be identical with these cells. These results give further evidence that cells with neuron-like characteristics are present in the mucous layer of the small intestine.
