Effects of surfactant supplement and end-expiratory pressure in lung-lavaged rabbits.

By using adult rabbits in which the lung surfactant had been reduced by repeated whole-lung lavage, we examined the effects of surfactant supplement and positive end-expiratory pressure (PEEP). The rabbits were anesthetized with pentobarbital sodium and mechanically ventilated with pure O2. Surfactant that was extracted from pigs' lungs was supplemented to the rabbits through the trachea. The PEEP was set at 4 cmH2O. The seven rabbits that were ventilated without the supplement and without the PEEP all died of hypoxia, hypercapnia, and reduction of lung compliance within 2 h. The 18 rabbits that were ventilated with either the supplement or the PEEP showed some elevations of arterial O2 tension, but 9 of them died within 4 h. After application of both the surfactant supplement and the PEEP, six out of the eight rabbits survived for more than 11 h with nearly normal values of blood gases and compliance. We concluded that the combination of the transtracheal surfactant supplement and ventilation with PEEP has favorable therapeutic effects on surfactant deficiency.

Hemoglobin values in adult women of various occupational groups in the community.

In 1966 and 1971, the authors studied hemoglobin values of about 2,000 women of 10 different occupational groups and different social classes in the same district. The mean values of hemoglobin and serum iron were highest in nursing students followed by urban housewives and nutritionists and lowest in 3 groups of farm wives and shoe factory laborers. The difference in the mean values of hemoglobin observed among occupational groups was in principle assumed to be due to the difference of frequency in levels of 11-12 g/dl. Hemoglobin levels between 1966 and 1971 tended to improve as a whole and the differences among occupational groups have been reduced. In this study, some kinds of social conditions as well as some physiological factors related to hemoglobin values were investigated. In the same occupational group, unmarried women had a higher level of hemoglobin than married women. Further, married women with infants showed a relatively lower level than those with no infants. This suggests that hemoglobin values of female workers may be significantly influenced not only by work conditions but also by social conditions including family circumstances.

A follow-up study on the lung disorders of rush (igusa) farmers exposed to "sendo" dust.

In the process of "tatami" mat production, from cultivating to weaving, rush farmers are exposed to "sendo" (dyer's earth) dust using it to prevent fading of the "tatami" mat color. It has been found that the workers tend to develop a relatively early stage of pneumoconiosis after about 20 years of this work. In 1980, 51 male (58.6 years old) and 37 female (54.4 years old) rush farmers, some of whom were previously examined in 1970, were re-examined for lung disorders. Chest X rays in the 1980 showed, 49.0% of males and 62.2% of females over 1/0 according to the 12 point profusion scale with irregular small opacities being dominant, a significant increase in the prevalence rates of pneumoconiosis compared with 1970 values (p less than 0.01 for both males and females). A relatively high prevalence of respiratory complaints and/or lung function impairment was also found. However, there were no severe cases and no significant changes were seen to have occurred during the seventies by those tests. It was also observed that increases in lung disorders during the decade were more common in the female workers. These findings suggest that the pathogenesis of rush farmer's pneumoconiosis might be in the histological changes of bronchial and/or alveolar walls by the deposition of "sendo" dust. The present study revealed that the lung disorders in the rush farmers may be associated with long-term inhalation of "sendo" dust, and the introduction of more suitable dust control measures in that processing must be actively pursued in the near future.

Mode of degradation of plasmid DNA with ozone.

The ozonization of pBR322 closed circular DNA showed the conversion to open circular DNA. The damaged site was investigated by restriction mapping. The results showed the damage and subsequent cleavage of the DNA strand of ccDNA by ozonization may occur at the region sensitive to nuclease S1.

Synthesis and properties of 2'-deoxy-8,2'-methylene-cyclopurine nucleosides.

A method was developed to synthesize 2'-deoxy-8,2'-methylene-cycloadenosine (1) and -cycloguanosine (2) which were new carbon-bridged cyclopurine nucleosides fixed in a high-anti torsional angle region. 3',5'-Di-O-acetyl-8-methanesulfonyl-2'-O-p-toluene-sulfonyladenosine+ ++ (3) or 2-acetamido-9- (3,5-di-O-acetyl-2-O-p-toluenesulfonyl-beta-D-ribofuranosyl)-6- ethoxy-8-methanesulfonyl-purine (9) was treated with sodium salt of ethyl malonate to give the cyclized nucleosides (4 and 10) in good yields, respectively. Subsequent decarboxylation and deblocking of 4 and 10 afforded 1 and 2 in crystalline form, respectively.

Synthesis of 8,2'-ethano-cyclo-2'-deoxyadenosines.

The radical- or photo-cyclization of protected 2'-iodoethyl or 2'-phenylthioethyl derivatives of 2'-deoxyadenosine afforded the 8,2'-ethano-cyclo-2'-deoxyadenosine, a new carbon-bridged adenine cyclonucleoside.

[Research on experimental cerebral infarction in cats with in vivo TMR (topical magnetic resonance) approach].

Metabolic changes in the ischemic brains of cats were investigated in vivo with high energy phosphate compounds as parameters by using a topical magnetic resonance (TMR) spectrometer. The experimental focal cerebral ischemia was made in four cats by modifying the method of O'Brien and Waltz. The stem of left middle cerebral artery was exposed and set the occlusive device 5-7 days before the in vivo measurement. 31P-NMR spectrum was taken under general anesthesia with Ketamine HCI and with decreased blood flow. The following points must be considered in obtaining 31P-TMR spectrum in a cat brain: Firstly, as much muscle as possible must be removed from the detective area because it contains phosphate compounds. Our experiment showed that bone and blood had little or no effect on the 31P-TMR spectrum. Secondly, although same procedure was repeated, it was difficult to obtain constant ischemic lesion; in site and size. The detective area in setting was not changed in the particular area. However there was a possibility of the detective area also including various non-ischemic regions. Thirdly, 31P-TMR spectrum had several peaks in a cat brain; which were sugar phosphate, inorganic phosphate, phosphodiesters, phosphocreatine, gamma-, alpha-, beta-ATP in the pre-occlusive conditions. These peaks did not appear in clear volume separation with one another. We drew the perpendicular line from through between two neighbour peaks to the horizontal base line and artificially divided two peaks. The area of each peak did not represent correctly the density of each component. Fourthly, unnecessary components were rationalistically excluded from the 'raw' spectrum.(ABSTRACT TRUNCATED AT 250 WORDS)
