[Introduction of human leukocytes in cold anabiosis (-20 degrees C) under an exponential program].

Investigation of effective and accessible method for administration of human blood leukocytes in the cold anabiosis condition at am oderately low temperature of minus 20 degrees C by the exponential program with cryopreservative which does not require washing from the bio-object after its thawing, has been proposed. Morphological and functional properties of the leukocytes after different periods of conservation in the state of cold anabiosis were studied. The data obtained suggest that optimal conservation of the morpho-functional properties of cells with the aid of this method equals 3 weeks.

Ability of L-histidine to decrease desensitization of the myometrium to epinephrine.

Experiments were performed with 62 longitudinal strips of the uterine horns from 10 nonpregnant rats. The ability of continuously infused epinephrine in high concentration (10(-6) g/ml, 30 min) to suppress spontaneous contractions due to activation of beta-adrenoceptors progressively decreased, which was associated with receptor desensitization. Histidine in a concentration of 3 x 10(-11) g/ml had no effect, while in concentrations of 3 x 10(-8), 3 x 10(-7), and 3 x 10(-6) g/ml decreased the degree of desensitization. Our results indicate that histidine not only potentiates beta-adrenoceptor activation, but also prevents the development of desensitization. These data should be taken into account during therapy with beta-adrenoceptor agonists.

Phagocytic activity of neutrophils and lymphocytes at various periods of storage in the state of cold anabiosis (-40 degrees C).

We studied changes in phagocytic activity of neutrophils and lymphocytes subjected to cold anabiosis. The proposed effective method for introduction of human blood leukocytes into cold anabiosis at moderately low temperature (-40 degrees C) in the presence of a cryopreserving agent does not require washout of the biological material after defrosting.

Effect of cryohypobiosis on conservation of human blood leukocytes.

Morphological and functional indicators were investigated of donor leukocyte concentrates mixed at a ratio of 1 to 1 with three versions of cold-protective and kept at an undercooling temperature (-10 degree C) for 24 hours, and with a cryoprotector of optimal composition for 12 days,. An optimal version of a non-freezing cold-protective solution was found. It was determined that 76+/-4 % of the neutrophils retain there ability of phagocytosis among the viable cells (81+/-5 %) after a 12 day hypobiosis. Thus the results testify to a high morphofunctional safety of human blood leukocytes which experienced cold-hypobiosis (-10 degree C) in an original non-freezing solution not requiring washing after re-warming.
