2-Amino-5-methoxy-2'(or 3')-methylindamine and 2-amino-5-methoxy-2'(or 3')-methylindoaniline were isolated from a coloring matter formed by oxidative condensation of 2,5-diaminotoluene with 2,4-diaminoanisole, and their structures were elucidated on the basis of nuclear magnetic resonance and mass spectral data. These two main components of the oxidation product formed at the early stage exhibited a positive result in the mutagenicity test on the thin-layer chromatography plate. The mutagenic activities of purified aminoindamines and aminoindoanilines against Salmonella typhimurium TA 98 were as strong as that of 2-acetamidofluorene. It was also demonstrated that 5-methoxy-2'(or 3')-methylindamine was formed in 10% yield under the conditions used for hair dyeing and the yield was dependent upon the molar ratio of 2,5-diaminotoluene to 2,4-diaminoanisole.
The effect of RES blockade on the survival of heat-damaged and antibody coated red blood cells was evaluated in C3H mice. RES function of C3H mice was determined by the survival of 59Fe-labeled, heat-damaged red blood cells at 49 degrees C for 30 min. When animals were treated with an intravenous injection of non-labeled, heat-damaged red blood cells for the blockade of RES, red blood cell destruction was depressed in non-splenectomized and splenectomized mice. In the survival or organ distribution of 59Fe-labeled, antibody-coated red blood cells, no difference was noted between animals with and without blockade. These findings suggest that heat-damaged red blood cells were destructed predominantly in the splenic RES and antibody coated cells were not broken down only by the splenic RES.
Clinical evaluation of cefotiam (panspolin), a new cephem antibiotics, was performed in the infectious disease associated with hematological disorders and in the respiratory system. In hematological dis orders, 40% of good and 25% of fair results were obtained in clinical effects. In respiratory infections, however, 92% of good results were obtained. Opportunistic infection due to Gram-negative bacilli are often experienced in patients with leukemia. Since cefotiam has sufficient bacteriocidal effects in broad spectrum, it would be a good therapeutic agent against infectious diseases associated with hematological disorders.

The quantitation of captopril in biological fluids by gas-liquid chromatography using a flame photometric detector is described. Captopril was converted into an adduct with N-ethylmaleimide and then into the hexafluoroisopropyl ester. The latter derivative was separated on a 2% OV-210 column and determined by employing the captopril-N-hexylmaleimide adduct as an internal standard. The blood level and urinary excretion of captopril administered intravenously to dogs were measured by the proposed method.
A platelet kinetic study in 2 patients with acute promyelocytic leukemia (APL) associated with disseminated intravascular coagulation (DIC) demonstrated that the platelet survival was characteristically shortened. These findings suggest increased platelet consumption during the process of DIC in APL.

Technetium-99m had been used as a radioactive label to study the migratory patterns of neutrophils. In a previous in vitro study, neutrophils were labeled with Tc-99m and infused into patients with and without various hematological disorders. Increased pulmonary localization was detected by scintillation camera within 10 min; this decreased gradually within 3 hr. Accumulation was seen in the liver and spleen at 3 hr. The same results were noted by using neutrophils labeled with Tc-99m sulfur colloid. In a patient with severe ulceration in the oral cavity, due to acute leukemia, Tc-99m-labeled transfused neutrophils that were collected by filtration leukopheresis were concentrated in the infected lesions.
