bcl-2-immunoglobulin transgenic mice demonstrate extended B cell survival and follicular lymphoproliferation.

Human follicular B cell lymphomas possess a t(14;18) interchromosomal translocation that juxtaposes the putative proto-oncogene bcl-2 with the immunoglobulin (Ig) heavy chain locus. We generated minigene constructs representing the bcl-2-Ig fusion gene found at this chromosomal breakpoint. These constructs were placed into the germ line of mice to assess the effects of the t(14;18) during development. The transgene demonstrates a lymphoid pattern of expression and uniformly results in an expanded follicular center cell population. Hyperplastic splenic follicles coalesce to form massive regions of splenic white pulp. Mice over 15 weeks of age demonstrate regional lymphadenopathy with abnormal cellular infiltrates. The expanded lymphoid compartment is composed predominantly of polyclonal B220-positive, IgM/IgD-positive B cells. Provocatively, the bcl-2-Ig transgene confers a survival advantage to a population of mature B cells assessed in vitro. bcl-2-Ig transgenic mice document a prospective role for the t(14;18) in B cell growth and the pathogenesis of follicular lymphoma.

Alternative promoters and exons, somatic mutation and deregulation of the Bcl-2-Ig fusion gene in lymphoma.

The most common translocation in human lymphoma, the t(14;18)(q32;q21), generates heterogeneous 4.2-7.2 kb Bcl-2-immunoglobulin (Ig) chimeric mRNAs resulting from alternative Bcl-2 5' exons and varied Ig 3' untranslated regions (UT). The normal human Bcl-2 gene has a three exon structure with an untranslated first exon, a facultative 220 bp intron I, but an enormous 370 kb intron II. S1 protection and primer extension analysis defined initiation sites in exon II associated with classic promoter elements and a decanucleotide (ATG-CAAAGCA) homologous with Ig variable region enhancers. Multiple initiation sites were also found in a GC-rich region with Sp1 binding motifs in exon I. Most t(14;18) breakpoints cluster within the 3' UT of Bcl-2 implicating that event in gene deregulation. The Bcl-2 gene introduced into the Ig constant (C gamma) locus of SU-DHL-6 displayed somatic mutation. While Bcl-2--Ig mRNAs demonstrated an unaltered 2.5 h half-life, the Bcl-2--Ig gene revealed an inappropriately high rate of transcription for a mature B-cell. This indicates the translocated Bcl-2 allele has escaped normal control mechanisms.

Accessory spleen: presentation as a large abdominal mass in an asymptomatic young woman.

Accessory spleen hypertrophy is usually related to hematologic, lymphomatous, and rheumatoid diseases. We describe here the case of an 18-year-old asymptomatic woman who presented with a very large accessory spleen occupying the lower abdomen. Its appearance on abdominal sonogram, computed tomogram, and hepatosplenic scintigraphy is correlated with the resected specimen.

Acute oral poisoning with lindane-solvent mixtures.

Six cases of oral intoxication with lindane-solvent mixtures are reviewed. The ingested doses of lindane (mean dosage 120 mg/kg +/- 86 mg/kg) and benzene (mean dosage 366 mg/kg +/- 93 mg/kg) exceeded the toxic level. Symptoms (vomiting, dizziness and hyperreflexia) occurred within 30 min and all patients had epileptiform seizures. Two patients suffered from pulmonary edema and one of them had a severe rhabdomyolysis. Diazepam was sufficient to control convulsions in five cases. Gastric lavage was performed in five patients and activated charcoal, liquid paraffin with saline cathartic, and cholestyramine were used as adsorbents. Recovery was complete in all patients.

Sampling membrane potential, membrane resistance and electrode resistance with a glass electrode impaled into a single cell.

A method is demonstrated to measure membrane resistances and membrane potentials of single cells during impalement by a single glass microelectrode. The intention was to develop a procedure which would provide data almost continuously. Therefore, a frequency-dependent voltage divider network has been chosen to represent the basic electrical properties of the electrode and cell membrane, and used to explore its voltage response to a current stimulus, consisting of two rectangular pulses of different widths. It can be shown that the resolution of the method can be improved by inverting this stimulus so that each polarization becomes a relaxation and vice versa. In order to generate, analyze and display this signal continuously, a device has been designed which has been called 'Electrophysiological Monitor, (E1M2)'. E1M2 provides a current stimulus as input into a standard bridge network and can analyze the summed response of the electrode and cell by a set of sample-hold amplifiers. It then decodes and displays the data continuously, as membrane potential (Em), input resistance of the cell (Rinp) and the electrode resistance (Re) respectively. From Rinp the membrane resistance (Rm) can be deduced. The validity of the method has been examined by measuring these parameters in frog muscle cells. Technical design considerations, the accuracy and possible pitfalls with the suggested procedure are discussed.

[Methods for differentiation of streptococci: comparison of serological, biochemical methods, triple- and camp-test (author's transl)].

Methods for group classification of beta-hemolytic streptococci were compared: biochemical versus serological techniques as well as Triple- and CAMP-Test. Attention was mainly put to the Phadebact streptococcus test, based on the co-agglutination principle which is intended to differentiate groups A, B, C and G. This test is easy to conduct and allows grouping within minutes. Classification was done with 50 strains of clinical isolates. Approximately 88% of the strains were identified as group A and B which are the most important in human streptococcal infections. The co-agglutination gave false results with 6 strains (8%), one of which was group A. The others turned out to be S. milleri, they could be identified only by the Lancefield precipitin test. In cases where several Phadebact antisera lead to an agglutination of one strain, the questionable result must be verified by conventional methods.

[100 Years of anthropological examination of school children in Jena (GDR)].

The present paper gives a review of the aims and the importance of the anthropological examinations on Jena school children, which have been carried out since 1880. It is shown that secular acceleration of stature and body weight was greater between 1964 and 1975 than the changes found in previous investigations. Special importance is attributed to a comparison of the results of the investigation made in 1880 with those of 1975.
