In the albino rat, perikaryal diameter, volume density of the granular endoplasmic reticulum and Golgi apparatus, and lumenal diameter of cisternae of the granular endoplasmic reticulum are larger in Purkinje cells of lobule VIa (neocerebellum) than in those of lobule X (archicerebellum). In contrast, only the surface density of cisternae of the granular endoplasmic reticulum is larger in Purkinje cells of lobule X. The cisternae of granular endoplasmic reticulum are arranged into conspicuous Nissl bodies parallel to the nuclear membrane, but the content of ribosomes and polysomes is markedly less in lobule-X cells than in cells from lobule VIa. These results indicate qualitative and quantitative differences between the metabolically important organelles in Purkinje cells of the neo- and archicerebellum (cf. Larsell 1952).
Two-dimensional gel electrophoresis analysis of X0 and XY fibroblasts from a monozygotic twin pair reveals two Y-dependent polypeptides. The polypeptides have molecular weights and isoelectric points of 38,000/6.3 and 30,000/5.4 and are designated as Y-38 and Y-30.

Two-dimensional gel electrophoresis analysis of a rodent-human somatic cell hybrid containing the X as the only human chromosome reveals three polypeptides that are absent in the parental cell line. The presence of these spots in human female fibroblasts indicates their human origin. The polypeptides have molecular weights and isoelectric points of 30,000/5.8,37,000/5.4, and 57,000/4.7 and are designated as PFHG 1, PFHG 2, and PFHG 3. Comparison of their molecular characteristics with those of polypeptides assigned to the human X in two other investigations shows that some but not all polypeptides are similar. Factors are discussed that might interfere with the rapid development of a standardized polypeptide map of the human X.
Prostaglandin E2 (PGE2) seems to stimulate cAMP accumulation in ovaries of all mammals. While it acts through specific receptors in some species, our earlier observations (1) suggest absence of PGE2 receptors in the rat ovary. In order to further substantiate this assumption we digested ovarian membranes from the bovine and the rat with various enzymes and measured cAMP after stimulation with PGE2, NaF, and hCG. Pronase, trypsin, and phospholipase C abolished cAMP accumulation completely. Neuraminidase, beta-galactosidase and phospholipase D did not interfere with cAMP formation. After treatment with phospholipase A2, PGE2-mediated cAMP accumulation was abolished in the bovine but not in the rat ovary. Formation of cAMP disappeared after hCG but not after NaF in both species. Furthermore specific binding of PGE2 could not be demonstrated in phospholipase A2-treated bovine ovaries. These findings are consistent with presence of specific PGE2 receptors in the bovine and their absence in the rat ovary.
Studies published in literature have claimed that there are interconnections between seasons and mental diseases. However, such conclusions are unacceptable, since they have no methodological foundation: whatever was claimed to have been measured, was actually not measured at all. This makes any discussions on the causes of such interconnections redundant. What was actually found, were certain connections between rates of hospitalisation and certain seasons. In our study, such relationships were also measured, but partly we also found other seasonal variations in hospital admission rates. The only conclusion that can be drawn from these observations is that, while there are certain connections between seasons and hospitalisation, the reasons for such variations are rules and conditions governing the admission to hospitals which do not bear any relation to the season at a particular time. Such relations, if any, are probably so highly complex that it will be necessary for establishing any causal relationship to gradually eliminate any possible disturbing factors by examining their impact and importance in each case. This requires a stepwise and extremely cautious approach.
Two-dimensional gel electrophoresis of rat gonads in various developmental stages (embryonic days 16, 17, 21; postnatal day 5) reveals an increasing number of polypeptide spots; 0.6%-2% of the polypeptides are gonad-specific and increase from embryonic day 16 onwards. Exposure of newborn rat ovaries to testosterone for 5 days results in the appearance of eight polypeptides. These polypeptides are absent in control ovaries but present in the testis from embryonic day 16 or 17 onwards. Three do not appear in the ovary at any developmental stage. These findings indicate that testosterone plays a physiologic role in normal testicular differentiation. After long-term testosterone treatment, ovaries are depleted of germ cells. This might explain the degeneration of oocytes in the abnormal environment of a testis (e.g., in XX true hermaphrodites).
A retrospective analysis from the clinical-toxicological point of view Within a ten-year period a group of 130 patients treated with Ftorafur (FT) was selected randomly and 124 of these patients were evaluated with regard to therapeutic side effects using objective criterions. FT founded to be a therapeutic effective cytostatic drug with only a low level of therapeutic side effects. So FT seems to be combinable with more toxic cytostatics without additional serious toxicity for non-tumor tissues. The peroral FT-application was followed only by low graded side effects in comparison with a higher incidence after intravenous injection or infusion. Most observed FT side effects were: nausea and vomiting, toxic effects on the hematopoetic system and alopecia. No serious neurotoxic effects were observed.
In order to learn more about the role of sex chromosome-dependent gene products in gonadogenesis, changes in protein patterns were studied during gonadal development. Two-dimensional gel electrophoresis analysis revealed specific proteins in both sexes at all developmental stages. Evidently the gonads are not indifferent by biochemical criteria at any developmental stage and express several specific genes from the onset of differentiation. To correlate these polypeptides with the sex chromosomes, proteins were investigated in human-rodent somatic cell hybrids and in genetically identical cell clones differing in one sex chromosome only. On two-dimensional gels one Y-dependent polypeptide was found with similar characteristics (relative molecular mass and isoelectric point) as an early testicular polypeptide. Its identity, however, remains to be proven.
The validity of two-dimensional echocardiography (2 DE) in the diagnosis of evolving myocardial infarction was assessed in 123 patients with a suspected acute myocardial infarction (chest pain for less than 4 hours). 84 patients developed a myocardial infarction, 39 did not. Detection and extent of a regional wall motion disorder in 2 DE were compared to ECG, enzymes, hemodynamics, coronary angiography and clinical data. The reliability of ECG and 2 DE in the diagnosis of acute myocardial infarction was found to be similar: sensitivity 89% for 2 DE, 96% for ECG; specificity 89% vs (= versus) 73%; positive predictive value 95% vs 89%; negative predictive value 79% vs 90%. This means that in most cases the ECG is a sufficient and reliable method in detecting or excluding acute myocardial infarction. Because of the equipment needed, the limited practicability and because of specific problems (distinction between new and old infarction), the 2 DE should be regarded as a supporting method to the ECG but not as an essential one in the diagnosis of acute myocardial infarction. The possible benefit of echocardiographic examination is the identification of high risk patients by non-invasive determination of the size of infarction. There was a significant correlation between the extent of wall motion disorder and the maximum CK levels as well as the incidence of cardiac complications such as hypotension, pulmonary congestion, and ventricular arrhythmias.
The efficacy of immunotherapy (IT) of immediate type allergies is well established but its mode of action is unclear. Indication for IT is based on history confirmed by skin tests, RAST and possibly provocation tests. Nature, severity and duration of allergic symptoms must be considered. IT is especially suitable as a basic treatment of seasonal allergic rhinitis and pollen asthma as well as perennial rhinitis and asthma due to the house dust mite. The value of IT with molds and bacterial vaccines is questionable. With insect venoms IT is well established as a means of preventing anaphylactic reactions to hymenoptera stings. Improvement of IT will be brought about chiefly by better standardization and purification of allergen extracts. The significance of IT with chemically modified allergens and using different modes of application (oral, nasal) cannot be definitely evaluated at present.
458 patients with systemic allergic reactions following hymenoptera stings were investigated with regard to the incidence of atopic disease. Personal atopy (13.7%) and a family history of atopy (26.4%) were not found more frequently than in a normal population. The age of the first manifestation of hymenoptera sting allergy was however significantly lower in atopic than in non-atopic patients, sensitization occurred following a smaller number of stings and there was a preponderance of respiratory symptoms.

Intoxications by cortinarii have only been recognized in recent years. Compared to other mushroom poisoning they are characterized by the long incubation time of up to ten days or more, and predominantly hepatorenal symptoms. In this paper the first two Swiss cases of intoxication with Cortinarius phlegmacium splendens are reported. The two patients ate two meals of this mushroom one week apart. Three days after the second meal the patients became acutely ill with severe lumbar pain, polyuria, polydipsia and constipation. Laboratory examinations revealed signs of hepato-renal toxicity with increase in serum creatinine, hyposthenuria and hyperbilirubinemia. Both patients recovered fully in the course of a further two weeks.
H-Y antigen was tested in five women with sex chromosome mosaicism and gonadal streaks. Three patients had a 45,X/46,XY or 46,X,der(Y) and two a 45,X/46,X, der(X) chromosome constitution. All patients were H-Y antigen positive. Lack of testis differentiation in these women may be explained by subthreshold expression of H-Y antigen, different H-Y antigen molecules, and/or different tissue distribution of the chromosome mosaicism.
