[A comparative evaluation of treatment methods for a genital papillomavirus infection in women with different viral genotypes].

Such genotypes of human papilloma virus (HPV) as "malignant" (180), "benign" (49) and indeterminate (127) were identified in 356 females by PCR and cytological procedures. The following therapeutic techniques were compared: cryodestruction of the uterine cervix (60), antiviral medication (AVM) (19), combination of AVM and cryodestruction (224), and combination of AVM and laser destruction (54). In the "malignant" HPV group, AVM in conjunction with cryo- or laser destruction was significantly more effective than cryodestruction or AVM alone. Both cryodestruction alone and in combination with AVM were more effective in treating "benign" genotypes. Repeat courses showed the same tendency.

[Human oncogene expression in the normal state and in various blood system diseases].

A study was made of the expression of the cellular oncogens c--myc, c--sis and c--abl in mononuclears from normal donors and patients with different patterns of leukemia and hemopoietic dysplasia using spot hybridization on nitrocellulose filters with virus homologs of the given oncogens. Altogether 43 persons were examined. No specific expression of the oncogens indicated was established whatever the pattern of leukemia. The relationship between the expression of these oncogens and the efficacy of the treatment of acute myeloblastic leukemia was demonstrated. However, in the given investigations, no differences were identified in the expression of oncogens in the leukemic and normal cells stimulated with PHA and phorbolic ether. Enhanced expression of cellular oncogens myc, sis and abl occurred in acute leukemia.

[Quantitative histochemical study of the reaction kinetics to inosine-5-monophosphate dehydrogenase and 5'-nucleotidase in cryostat sections of rat liver].

The data are presented on kinetics of histochemical enzymatic reactions demonstrating two enzymes taking part in the purine metabolism--inosine-5-monophosphate dehydrogenase (IMPD) and 5'-nucleotidase (5'-NT). It is shown that IMPD has a very weak affinity to substrate IMP (KM = 2.5 . 10(-2) M); this fact partially explains the low rate course in cryostat sections. Cytosol and membrane forms of 5'-NT have the maximum affinity to AMP (KM for membrane and cytosol forms being 1 . 10(-3) M and 2.1 . 10(-3) M, resp.). When IMP is used as a substrate, 5'-NT localized in cytosol has much lower KM as compared to the membrane form. Cytoplasmic 5'-NT is thermostable. It is suggested that a low rate of histochemical reaction demonstrating IMPD is caused by a degradation of substrate by the membrane form of 5'-NT, the Michaelis constant of which is 5 times less than that of IMPD.

[Changes in the myocardium in the early stages of chronic kidney failure].

The functional state of the myocardium was studied in 45 white Wistar rats in the early stages of chronic renal insufficiency. After resection of 4/6 of the renal parenchyma the initial stage of chronic renal insufficiency was created with the urea level in blood serum 7.0-10.0 mmol/l with normal content of electrolytes (duration of observation lasting 1.3 to 6 months). Towards the 3rd month myocardial hypertrophy developed in animals with statistically significant increase of the cardiac mass by 18-20%, not related to enhancement of haemodynamic load on the heart. Arterial pressure remains normal. Hypertrophy of the myocardium as shown by histochemistry was accompanied by increase of energy and protein metabolism, carbohydrates decreased simultaneously due to the decrease of myocardium tolerance to glucose.

[Quantitative evaluation of the effectiveness of using Meldola blue and sephadex in the histochemical detection of dehydrogenases in cryostatic sections].

Ther influence of meldola blue (MB), sephadex G-150 and ficoll 400 on kinetics of histochemical reactions for succinate and lactate dehydrogenases in cryostat sections of rat myocardium was estimated spectrophotometrically. MB is an effective electron carrier, although its application in quantitative dehydrogenase histochemistry is limited by its property to bind up with tissues non-enzymatically. 5 x 10(-5) M is the optimal MB concentration in the incubation medium which accelerates the enzymatic reaction in cryostat sections. The advisability of addition of sephadex G-150 to prevent from diffusion of LDH to the incubation medium is confirmed.

[Autoradiographic study of hormonal regulation of proliferating golden hamster uterus cells during postnatal ontogeny].

Study of the effects of octestrol, 17-hydroxyprogesterone caproate and tamoxifen on uterine cell proliferation in golden hamsters aged one to twenty days has shown the existence in the uterine epithelium of two cell populations with varying sensitivity to sex hormones. The cells of uterine glands being formed by day 12 of postnatal ontogenesis were found to be insensitive to the proliferation-inhibiting effect of progesterone and little sensitive to the proliferation-stimulating action of the estrogen. The epithelial cells lining the uterine cavity show a high sensitivity to sex hormones. The absence of changes in proliferative activity of the epithelial cells exposed to tamoxifen allows a suggestion that ontogenetic changes in the proliferative activity of the uterine cells do not depend on the presence of endogenous hormones.

[Use of the Elektronika-T3-16M special-purpose computer for the automatic processing of cytophotometric and cytofluorimetric data].

A system has been proposed to provide the automatic analysis of data on: a) point cytophotometry, b) two-wave cytophotometry, c) cytofluorimetry. The system provides the input of the data from a photomultiplier to a specialized computer "Electronica-T3-16M" in addition to the simultaneous statistical analysis of these. The information on the programs used is presented. The advantages of the system, compared with some commercially available cytophotometers, are indicated.

[Quantitative kinetic study of the histochemical reaction to peroxidase in salivary gland and uterine cells of golden hamsters].

The kinetics of diaminobenzidine reaction for peroxidase has been studied using cryostat sections of salivary gland and uterus of the golden hamster. It was demonstrated that the inhibition of the initial velocity of the reaction in salivary gland occurred at hydrogen peroxide concentration in the reaction medium being 5.10(-3) M or higher. On the other hand, the inhibitory action of hydrogen peroxide excess on the uterine peroxidase activity was observed at H2O2 concentrations higher than 1.10(-3) M. KM values for diaminobenzidine, determined for salivary gland and uterine peroxidases, were 2.3.10(-3) M and 3.5.10(-3) M, resp; KM values for H2O2 were 1.1.10(-4) M and 2.5.10(-5) M, resp. Kinetic difference of salivary gland and uterine peroxidases obtained in this study support the view on the organ specificity of peroxidases.

[Mucosal changes in the stomach of rats in the early stages of chronic renal failure].

In a series of chronic experiments on 30 rats subjected to nephrectomy, use was made of the present-day morphological and quantitative histochemical techniques to study the activity of oxidation-reduction enzymes, hydrolases and PAS-reaction in cellular elements of the mucous membrane of the fundal and antral parts of the stomach. It was found that in the early stages of experimental renal insufficiency, all the main cells of the glands manifested marked hyperplasia. These alterations seem likely to be adaptive in nature and are caused by the stimulatory action of the increased amounts of nitrous residue in the gastric lumen of such animals. The authors discuss the origin of gastric pathology in experimental animals and make clinical and experimental correlations.

[Electron microscopic study of the glycogen in the epithelial cells of the human oral mucosa].

In the human oral mucosa, glycogen particles of beta-configuration are mostly located in the spinous cells, being, however, scanty in the basal and superficial cells. The particles situated near the Golgi apparatus, are covered with a trilaminar unit membrane, and some of them are seen to be membrane-continuous with the Golgi cisterns. Intermingled with the typical glycogen particles, smaller particles are encountered, thus implying the earlier stages on the mature glycogen particle formation.

[Functional-morphological model of organization of the epithelium of normal, hyperplastic and neoplastic human endometrium].

A model of functional-morphological organization of epithelium of human normal, hyperplastic, and tumour endometrium is described. The model is based on a hypothesis that a population of stem cells capable of indefinite unlimited self-maintenance exists in normal glandular epithelium of the endometrium. The population of normal stem cells of endometrium epithelium is the source of emergence of differentiated types of epithelial cells among which, according to the model, three cell types can be distinguished: cells sensitive to estradiol alone, cells sensitive to both estradiol and progesteron, and cells sensitive to progesteron alone. The conditions necessary for transition of one cell type to another as well as histochemical characteristics of each of the cell types are described. The model has been used for the analysis of morphological changes developing in hyperplastic processes and cancer of the endometrium as well as for the substantiation of the current schedules for hormone therapy of these diseases.

[Pressing problems of the intravital morphological diagnosis of pathological states of the endometrium].

The important problems of morphological diagnosis of the endometrium conditions by scrapings are dealt with. The necessary information in the features of the functional organization of the normal and pathological endometrium as well as the required histochemical and histoenzymatic methods of examination are presented. The principles of diagnosis of glandular hyperplasia and atypical glandular hyperplasia of the endometrium and of differential diagnosis from highly diffentiated variants of endometrium cancer are described. The influence of hormonal therapy on endometrial carcinomas is discussed. It is emphasized that pathologists should give due consideration to the clinical data. A sample of the form for sending of the endometrial scrapings into the laboratory is presented.

Quantitative histochemical and stereological study of alkaline phosphatase in the rat thyroid gland.

The histochemical alkaline phosphatase reaction in thyroid vascular endothelium was estimated spectrophotometrically and stereologically. Thus two main characteristics of the reaction were obtained: 1. the average enzyme activity, reflecting the level of transport processes in the capillary-thyrocyte system; 2. the relative volume of functioning vessels. An index of thyroid vascularity is proposed that is equal to the product of these two characteristics. The changes of the primary characteristics as well as of the vascularity index, caused by experimental hypo- and hyperplasia of the gland, are discussed.

Cytospectrophotometric study of the kinetics of histochemical reaction of GABA-transaminase in cryostat sections of rat cerebellar cortex.

A modification of the histochemical method for demonstration of GABA-transaminase is proposed. Substrate and cofactor concentrations are chosen on the basis of kinetic investigation in cryostat sections of the rat cerebellar cortex. Enzymatic reactions were measured by quantitative microspectrophotometry. Michaelis constants for alpha-oxoglutarate in the Purkinje cell layer and granular layer (Km 1.7 x 10(-3) M) and white matter (Km 3.8 x 10(-3) M) are found. It is shown that alpha-oxoglutarate in concentrations higher than 5.2 x 10(-3) M (1 mg/ml) suppresses the reaction in sections by competitive inhibition. The advisability of addition of malonate, PMS and cyanide to the incubation medium is confirmed. It is suggested that there are some isoenzymes of GABA-transaminase with predominant localization either in neurons or glia.

On the concept of stem cell and a model of functional-morphological structure of the endometrium.

The paper discusses the fundamentals of a functional-morphological model of the structure of human endometrial ephitelium during two phases of the menstrual cycle. The model is based on the hypothesis that endometrial epithelium comprises populations of stem cells, capable of long-term self maintenance and producing differentiated epithelial cells, which can be divided according to the model, into three types: estradiol-sensitive cells, estradiol- and progesterone-sensitive cells and progesterone-sensitive cells. The significance of changes in blood-estradiol and progesterone levels during the menstrual cycle required for conversion of one type of the cells into another is considered, and histochemical characterization of different types of cells is given.

[Action of 17-hydroxyprogesterone caproate on the glandular hyperplasia of human endometrium].

The effect of 17-oxyprogesterone capronate on the proliferative activity and metabolic processes in the endometrium of patients with glandular hyperplasia of the endometrium was studied. It was shown that the proliferative activity was inhibited while the secretory processes in the glandular epithelium were activated. This effect was achieved only after a long-term (6-9 months) administration of the hormone.
