[The problems of the central regulation of immune system biorhythms: the role of exogenous and endogenous melatonin].

One of the key synchronizers of mammalian circadian rhythm is the hormone melatonin (MT) produced by the pineal gland. MT is characterized by a wide spectrum of biological activity including its immunopotentiating effect. At the same time, the results of studies dedicated to the effects and mechanisms of melatonin immunoactivity seem to be contradictory. Studies conducted by the authors of this article show that diurnal variations in MT content in blood and saliva of healthy humans demonstrate significant individual fluctuations connected with the seasonal factor. The character of MT relations with the immune system varies depending on the phase of the circadian cycle. Patients with immunopathological condition (bronchial asthma--BA) demonstrate a reduced number of correlations between MT level and immunological parameters. After treatment, the positive dynamics of immunological and clinical parameters in BA patients is associated with an increased number of correlations between the immunological status parameters and MT level in blood, which may indicate an increase in its immunoregulatory role. Administration of melatonin and amitriptyline, the inductor of its synthesis, in animals kept under continuous light helped to recover normal circadian rhythmicity in the cell content of lymphoid organs. Solving problems associated with the immunomodulating and synchronizing role of MT could give researchers an insight into the details of neuroendocrine and immune interrelations and help to develop new methods for the prevention and correction of immune status disturbances in diseases and desynchronoses (stresses, jet lags, shift and night work, etc.).

Effect of experimental desynchronosis on immunotoxicity of benz(a)pyrene in (CBAxC57Bl)F1 mice.

Experiments on (CBA x C57Bl)F1 mice showed that constant (day and night) illumination for 2 weeks led to the development of internal desynchronosis, which significantly modulated the reaction of the immune system to benz(a)pyrene compared to those in intact animals. Presumably, the differences were associated with changed circadian rhythms of the immune parameters in experimental animals determining different ratios and, presumably, functions of immunocompetent cells during benz(a)pyrene exposure in intact controls and animals with desynchronosis.

Effect of benzo[a]pyrene on the immune status of mice with anxious-depressive syndrome.

We studied the effect of benzo[a]pyrene on cells of lymphoid organs, energy metabolism of blood lymphocytes, and immunological reactivity in mice with anxious depressive syndrome produced by social stress. Benzo[a]pyrene exhibited a more pronounced immunotoxic activity in anxious-depressive animals, which was more than a simple sum of the effects produced by adverse ecological and psychic factors.

Circadian dynamics of cell composition of the thymus and lymph nodes in mice normally, under conditions of permanent illumination, and after melatonin injection.

The effect of melatonin on disturbed circadian variations in the lymphocyte subpopulation composition of the thymus and inguinal lymph nodes was studied in CBA mice exposed to constant illumination for 14 days. The desynchronizing effect of permanent illumination on the thymus consisted in disappearance of circadian variations in the total number of thymocytes, absolute count of thymocytes, absolute counts of CD8(+) and CD4(+) cells, and in inversion of changes in the absolute counts of CD4(+)8(+) cells from 15.00 to 20.00. In lymph nodes circadian variations in the percentage of CD4(+)+ lymphocytes disappeared, while absolute counts of CD4(+)8(+) and CD8(+) cells changed from 15.00 to 20.00. Melatonin restored circadian dynamics of some parameters mainly in the thymus.

[Functional morphology of the cells of the immune system under experimental and clinical conditions].

In this study, the peculiarities of functional morphology of the cells of the immune system are examined under experimental and clinical conditions. Lymphocyte enzymatic spectrum and DNA content are determined, the significance and information value of these parameters are demonstrated under normal conditions and in cases of pathology of immune and lymphatic system. The circadian organization of the immune system is detected, and the dependence of morpho-functional state of the immune system on the structure of its circadian organization is shown. These data have found an application for clinical purposes. They contributed to the development of effective methods of diagnosis, prognosis and correction of dysfunctions of the immune system.

[Changes in liver and in some organs of immune system of animals exposed to twenty-four-hour illumination].

The aim of this research was to study the morpho-functional organization of liver and some organs of immune system in animals exposed to continous twenty-four-hour illumination (CI) for 14 days. Using the methods of light and electron microscopy and morphometric analysis of histological preparations, liver and regional lymph nodes (hepatic in rats and iliac in mice) were studied. It was found that CI resulted in a state of dyschronosis, which was structurally manifested by the changes of diurnal variations of morphometric parameters of liver and its draining lymph nodes, disturbances of hemo- and lymphocirculation, destruction of cellular membranes. Response of central and peripheral organs of immune system to CI included the changes of diurnal variations of blast forms and mature lymphocytes, that was also indicative of dyschronosis development.

Diurnal variations in lymphocyte subpopulations in lymphoid organs of rats with genetic catalepsy and Wistar rats.

Parameters of the immune status of the thymus and spleen in rats with genetic catalepsy were lower compared to those in Wistar rats. Diurnal variations in cell subpopulations of lymphoid organs were different in animals of these strains. Behavioral characteristics and neuroendocrine state in rats with genetic catalepsy were associated with specific changes in the immune system and neuroimmune interactions.

[Effect of chronic psychoemotional stress on subpopulation spectrum of T-lymphocytes in immunocompetent organs in male mice].

Subpopulation spectrum of T lymphocytes (CD3+, CD4+, CD8+, CD25+, and CD3(+)25+) in thymus, spleen and inguinal lymphatic nodes have been studied in male mice after 20 days of psychoemotional stress produced by social defeats in daily agonistic confrontations. A reduction of total number of cells, of absolute numbers of all researched subpopulations of lymphocytes and % CD3+ cells in thymus of submissive mice was shown in comparison with intact animals. Reduction of total number of splenocytes and absolute numbers of CD4+ and CD8+ lymphocytes has been observed in a spleen of submissive mice. Besides, % CD3+, CD25+, CD4+ and CD25+ cells were increased in these animals in comparison with intact mice. The absolute number of cells with CD8 phenotype was increased in inguinal lymphatic nodes. The data obtained suggest that the chronic psychoemotional stress is accompanied by serious changes of the cellular link of immunity. The effect of chronic emotional social stress on mutual interaction of the central and peripheral links of immunity has been discussed.

Behavioral effects of novel enterosorbent Noolit on mice with mixed depression/anxiety-like state.

The aim of this work was to examine the behavioral effects of a novel lithium-based enterosorbent, Noolit (665 mg/kg), on male mice with mixed depression/anxiety-like state evoked by exposure to repeated social defeats in daily agonistic confrontations. The lithium component allows Noolit to be used as a psychotropic drug. Two experiments are described, in which the therapeutic and preventative effects of chronic Noolit treatment were examined. Response to Noolit was assessed in the plus maze, open field, partition test, and Porsolt's test. In both experiments, Noolit produced obvious anxiolytic and antidepressant effects. Treatment with Noolit fully restored some behavioral parameters in the plus maze and open field in depressed mice and prevented depression that would otherwise have developed. It has been suggested that enterosorbent Noolit can be a potent drug for the treatment of mixed anxiety/depression pathologies and for prevention of mood disorders.

Genetic differences in immune reactions to radiation exposure in humans and experimental animals.

Examination of populations living in regions of fallout radiation revealed changes in the distribution of HLA genes not only in irradiated individuals, but also in their children and grandchildren. Combined irradiation of inbred mice in comparable doses showed that H-2 genes determine immune reactions in animals and their offspring to radiation. Our results indicate that the immune system in mammals is immunogenetically regulated by low-dose radiation.

Circadian variations of melatonin concentration in saliva and blood content of immunocompetent cells in healthy individuals.

Salivary content of melatonin and parameters of the immune status were studied in young healthy individuals. Morning and evening levels of melatonin showed different correlations with some immunity parameters. It was assumed that the degree and type of correlation between the immune system and melatonin depend on the phase of circadian cycle, which should be considered during evaluation of immunoendocrine status.

Effect of enterosorbent noolith on behavior and serotonin (1A) receptors in mouse brain.

Protective properties of a new enterosorbent noolith (lithium ions immobilized on mineral matrix) were studied in C57Bl/6J mice predisposed to depression caused by intermale confrontations. The drug was administered daily for 15 days after the 5th confrontation and then the animals were tested in the forced swimming test. The number of specific(3)H-8-OH-DPAT binding sites in 3 brain regions was determined. It is shown that noolith produced an antidepressive effect manifested in decreased immobility time in the Porsolt test. Moreover, noolith reduced the number of 1A-serotonin receptors in the frontal cortex and hypothalamus. It is concluded that noolith possesses protective properties.

Effects of amitriptyline on diurnal variations of oxidation-reduction enzyme activities in rat lymphocytes during experimental desynchronosis.

We studied the effects of amitriptyline on diurnal variations of oxidative-reduction enzyme activities (succinate dehydrogenase, lactate dehydrogenase, and NADPH diaphorase) in rat peripheral blood lymphocytes during experimental desynchronosis. Desynchronosis was induced by constant light exposure for 14 days and manifested in a morning shift of maximum lactate dehydrogenase activity and elevated morning NADPH diaphorase activity. Amitriptyline normalized morning lactate dehydrogenase activity and diurnal variations in NADPH diaphorase activity.
