[Effect of stunning on pork muscle quality].

The following methods of insensitisation were tested for their effects on muscle pork quality: Electrocoma, 75 V, 15 sec; Electrocoma following intensive electrical stimulation and irritation of various parts of the animals's body, using the insensitising forceps; Dart gun insensitisation. Also tested was the action of muscle relaxants on pork quality. Minor deviation from proper practice of electrical insensitisation was found to cause no significant deterioration in cutlet muscle and ham muscle quality. However, intensive irritation immediately before insensitisation caused higher percentages of PSE (16.7 per cent) and DFD (16.7 per cent). Dart gun insensitisation was quite often followed by poorer quality in the form of watery and bright pork (25.0 per cent). This had detrimental consequences for boiled ham processing (higher loss rates). Sensorial assessment of boiled ham obtained from animals in various categories did not reveal any significant discrepancy between the above techniques. When muscle relaxants were used, muscle pork quality of pigs relaxed by means of succicholine dichloride was much better than that in animals to which electrical insensitisation had been applied. Characteristics of such better quality included slower decline in pH over 45 minutes after death and lower drip loss.

Peripheral arterial reactivity in patients with peptic ulcers and in healthy subjects.

With an oscillograph according to Gesenius and Keller, the authors investigated the oscillographic waves in lower limbs and their reductions after exposition to cold and exercise in 27 men and 22 women with gastric or duodenal ulcer as well as in 18 healthy men and 12 healthy women. Both groups were subdivided into age categories from 19 to 30 and from 45 to 54 years. It was found that 1) peripheral arteries in men in the lower age category with duodenal ulcer reacted to cold stimulus by a more marked vasoconstriction than the arteries in healthy men in the same age category; 2) in women with duodenal ulcer the same observation was made in not only the younger, but also in the higher age category; 3) peripheral vasoconstriction after application of cold in women with duodenal ulcer, aged up to 30 years and 45-54 years was higher than in women with gastric ulcer in the corresponding age categories; 4) peripheral arteries of the younger men as well as of the younger and older women with duodenal ulcer reacted often more strongly to exercise and to exercise following application of cold in comparison with corresponding controls, and in the case of the women, in addition, with those with gastric ulcer.

Preclinical evaluation of a novel water-soluble chlorin E6 derivative (BLC 1010) as photosensitizer for the closure of the neovessels.

In the present study, photodynamic activity of a novel photosensitizer (PS), Chlorin e(6)-2.5 N-methyl-d-glucamine (BLC 1010), was evaluated using the chorioallantoic membrane (CAM) as an in vivo model. After intravenous (i.v.) injection of BLC 1010 into the CAM vasculature, the applicability of this drug for photodynamic therapy (PDT) was assessed in terms of fluorescence pharmacokinetics, i.e. leakage from the CAM vessels, and photothrombic activity. The influence of different PDT parameters including drug and light doses on the photodynamic activity of BLC 1010 has been investigated. It was found that, irrespective of drug dose, an identical continuous decrease in fluorescence contrast between the drug inside and outside the blood vessels was observed. The optimal treatment conditions leading to desired vascular damage were obtained by varying drug and light doses. Indeed, observable damage was achieved when irradiation was performed at light doses up to 5 J/cm(2) 1 min after i.v. injection of drug doses up to 0.5 mg/kg body weight(b.w.). However, when irradiation with light doses of more than 10 J/cm(2) was performed 1 min after injection of drug doses up to 2 mg/kg body weight, this led to occlusion of large blood vessels. It has been demonstrated that it is possible to obtain the desired vascular occlusion and stasis with BLC 1010 for different combinations of drug and/or light doses.

The effect of physical activity on the incidence of circulating platelet aggregates in healthy persons. The significance for antiaggregating therapy.

In 23 healthy persons active in sports are tested circulating thrombocyte aggregates in venous blood. Their incidence is expressed as thrombocyte index (TI). In this group there is TI 0.8900 +/- 0.0193. TI compared with TI evaluated in 21 healthy persons not interested in their physical fitness (TI 0.7699 +/- 0.0162) is significantly different (p less than 0.01). The physically not active persons develop significant (p less than 0.01) higher incidence of circulating thrombocyte aggregates in comparison with the incidence in group of physically active persons. The authors use TI for monitoring antiaggregant therapy and for selection the risk group of patients, whose clinical situation requires antiaggregant treatment. The dosage of antiplatelet drugs is estimated according TI values to reach the level recorded in healthy physically active persons (TI greater than 0.7699 +/- 0.0162).

Some interrelations between blood pressure parameters and claudication distances.

Sixty males and 2 females with the mean age of 58.1 years ranging from 39 to 80 years with atherosclerotic lesions of the arteries of lower extremities were studied. The claudication distance was subdivided into two segments: the first included the distance up to the onset of pain and the second one when the patients had to stop walking because of the severity of pain. The systemic systolic pressure and the poststenotic ankle pressure over both arteries on both calves by a Doppler ultrasound apparatus were measured and the individual pressure gradients were calculated. It was demonstrated that both claudication intervals were related to the poststenotic pressure as well as to the pressure gradient. The stepwise regression analysis showed a significant relationship between both distances and the lowest value of all ankle pressure measurements over four tibial arteries. The closest correlation (R = 0.98) was recorded between the initial and final claudication distance and it persisted even after the elimination of all pressure parameters.

Utilization of [1-14C]carbon of glycine of high glycine diet fed young and old rats.

The incorporation of radioactivity from [1-14C]glycine was studied into various organ (serum, liver, muscle) fractions (acid soluble, proteins, lipids, liver glycogen) and carbon dioxide in rats fed with isonitrogenous isocaloric purified diets. The diets contained 30% casein (control), gelatin (exchange of half of the 30% casein) or glycine (corresponding level of glycine in relation to the gelatin diet). The incorporation of radio-activity into proteins was reduced by feeding high glycine diets in young (20-weeks-old) and old (18-month-old) rats in relation to the control diet. The modifications of the results for old animals may be partially explained on the base of a reduced protein turnover rate and adaptation to a high gelatin (glycine) diet.

Correlation of immune cell activities and beta-endorphin release in breast carcinoma patients treated with galactose-specific lectin standardized mistletoe extract.

A prospectively randomized clinical study was performed with breast carcinoma patients to determine the correlation of defined parameters of the cellular immunity and beta-endorphin plasma levels after mistletoe lectin (ML-1) standardized therapy. The subcutaneous administration of optimal ML-1 dosages (0.5-1.0 ng ML-1/kg body weight; twice a week) induced an increased beta-endorphin plasma level, enhanced activity of peripheral blood natural killer (NK-)cells and T-lymphocytes (expression of CD-25/interleukin-2 receptors and HLA/DR-antigens). Statistical analysis of the data (Spearman correlation) revealed a significant correlation between NK- and T-cell activity and beta-endorphin plasma level. Thus, an obvious correlation between immune system and the neuroendocrine system may be anticipated which might gain therapeutical relevance.
