Kinetics of ligand binding to nucleic acids.

Ligand binding to nucleic acids (NA) is considered as a stationary Markov process. It is shown that the probabilistic description of ligand-NA binding allows one to describe not only the kinetics of the change of number of bound ligands at arbitrary fillings but also to calculate stationary values of the number of bound ligands and its dispersion. The general analysis of absorption isotherms and kinetics of ligand binding to NA make it possible to determine of rate constants of ligand-NA complex formation and dissociation.

Ligand binding with continuous modification of binding sites.

Simultaneous formulation binding and structure modification of the binding site leads to binding process that can be analyzed within the framework of the non-linear theory of dynamic systems. Such an approach allows us to obtain several properties of the binding center: plurality of stationary (stable and unstable) states at binding, recognition of bistable and hysteretic binding modes. It is also shown that adsorption centre deformation leads to a S-shaped adsorption curve.

The effect of osmotic gradients on the outward potassium current in dialyzed neurons of Helix pomatia.

1. The effect of outward and inward water flows through the membrane on outward potassium currents of dialyzed Helix pomatia neurons was studied. 2. An outward water flow increased the peak and sustained outward potassium currents and accelerated the kinetics of their activation. An inward water flow had quite opposite effects--it decreased the peak and sustained potassium currents and delayed the kinetics of their activation. 3. The analysis of the effect of water flow on the conductance of potassium channels showed that an outward water flow increased both the potassium conductance at a given potential (gk) and the maximum potassium conductance (gkmax). An inward water flow again had the opposite effect--it decreased the potassium conductance at given potential and the maximum potassium conductance. 4. Neither an outward nor an inward water flow significantly affected the fraction of open potassium channels at a given potential [n infinity(V)]. 5. These data suggest that in dialyzed neurons the changes of outward potassium current during water flow through the membrane are due mainly to the changes in single-channel conductance and the time constant of current activation.

Interaction of ganglioside-containing planar bilayers with serotonin and inorganic cations.

The binding of serotonin and inorganic cations K+, Na+, Ca2+, Mg2+ to planar bilayers formed from mixtures of phosphatidylcholine and mono-, di- and trisialogangliosides was studied by the potentiodynamic and nonactin-induced potassium conductivity method. The theoretical analysis of the results obtained was made taking into account (1) protrusion of the ganglioside charges from the membrane surface and (2) simultaneous adsorption of ions on the bilayer surface and on the ganglioside charges protruding into the solution. It was shown that there was no specific binding of K+ and Na+. The binding constants for Ca2+, Mg2+ were determined. These constants for all the gangliosides studied were equal to 500 M-1. The determined binding constants of serotonin to various gangliosides diminish in the following order: GD3 greater than GT1b greater than GD1a greater than GM1.
