Notes to the standardization of complement fixation reaction with Toxoplasma antigen.

Optimum conditions are described for the complement fixation tests, including the method of primary incubation in a refrigerator, use of serial four-fold dilutions for complement titration and serial double dilutions for haemolysin titration. An optimum haemolytic system, most sensitive to the complement and 50% unit of complement is to be found at the same time. More than 100 sera were tested simultaneously using other reactions for toxoplasmosis (microprecipitation in gel, indirect fluorescence reaction and latex test) in order to verify the appropriate dose of complement for complement fixation reaction. It was also investigated to what extent small deviations in erythrocyte concentration in the haemolytic system may influence the results of serum titration. The appropriate dose of complement was found to be five 50% units. It is recommended to correct colorimetrically the concentration of erythrocytes in the haemolytic system. The questions of an appropriate serum dilution in the titration are discussed and a rapid and exact method is chosen. A simple method of reading 50% haemolysis in complement fixation microreaction is described.

[Study of feces for parasitic stages].

A modification of faeces or soil (1 kg) examination technique for parasitic eggs and cysts is presented. The method is based on sedimentation (30 min) in a large measuring cylinder combined either with a biological test (hatching miracidia technic) or with a flotation method. For the flotation technique the solution of Kalium iodide (sp. grav. 1,3) is used. The use of kalium iodide is advantageous because the saturated solution reaches over 1,7 sp. gr. at the laboratory temperature.

[Latex reaction with toxoplasma antigen].

The author describes the latex fixation test (LFT) with Toxoplasma antigen. The main parts of this technique are as follows: the sensibilization of latex particles and incubation of the sensitized particles with sera examined in an icebox (overnight) followed by spinning at 5000 G 10 minutes.--The sera giving negative results with LRT reacted also in 78,3% with complement-fixing test (CFT) in 79.3% and with indirect fluorescent test (IFAT) in 61.8% and with microprecipitating test (MPT) in 100% negatively.--The sera reacting strongly positively with LFT reacted also with CFT in high titers (titer 160 and higher) in 60.9% and in 55% with IFAT (titers 128 and higher).

[Parasitologic collection technic and examination methods in equatorial West Africa].

The author has been a medical doctor and parasitologist over some years in different West-African countries. Based on his personal experience he underlines the special methodological conditions as well for the diagnoser as for the collector of parasitological documenting material. In this respect, the climatological factors have to be regarded as well as sociological and religious situations.

Latex reaction with toxoplasma antigen.

The technique of latex reaction with toxoplasma antigen is briefly described. The results of latex reaction are compared with those obtained by complement-fixation test, indirect fluorescence reaction and microprecipitation in gel.
