[Determination of the esterase activity of serine proteinases using synthetic substrates].

The authors suggest a highly sensitive rapid and simple method for measuring esterase activities of bovine pancreatic chymotrypsin, human neutrophilic cathepsin D, and elastase, and of human blood serum chymotrypsin-like esterase and elastase-like esterase activities, with fluorogenic synthetic ethers, amino acid derivatives, employed as substrates: N-benzoxycarbonylphenylalanine 4-methylumbelliferyl ester (Z-Phe-OMC) and tret-butyloxycarbonyl-1-alanine 4-methylumbelliferyl ester (BOC-Ala-OMC).

[Age-related immunodeficiencies of the neutrophilic granulocyte system].

Transitory age-associated deficiency of the granulocyte cationic proteins as a cause of a phagocytosis disturbance, loss of the phagocyte cell ability to kill phagocytosed bacteria is established. Quantitative indexes of myeloperoxidase content, elastase and catepsin G in the neutrophil granulocytes of newborns and laboratory animals are determined. Hypothesis is developed on the transitory age-associated immunodeficiencies of the neutrophil granulocyte system as one of the endogenic risk factors in the development of infectious diseases during early postnatal period.

[Effect of defensin on the process of healing of aseptic skin wound and on the permeability of blood vessels].

It was shown, for the first time, that polypeptide of rabbit's neutrophils, defensin (D) has the ability to accelerate the reparation process (RP). D was infused intramuscular to rats (125 micrograms/kg) 2 days before the operation, then the skin on the back was dissected through all layers (the length of the wound was constant-10 mm). The rate of the RP was estimated by the changes of the wound length on 2, 5, 7, 13-15 days after the operation. The injection of D or physiological solution was continued during 2 weeks. It was shown, that beginning from the fifth postoperative day the RP in the experimental rats developed faster, than in control rats. In the experimental group the full reparation appeared earlier, than in the control group (on 21-22 and 24-26 days correspondingly. Qualitative (with trypan blue) and semi-quantitative (with colloid coal) methods have shown, that D augmented the permeability of blood vessels. It was proposed, that the RP acceleration was induced by D augmentation of the blood vessels permeability.

[Function of the anticoagulant system when difensin is administered into the blood stream].

I. v. administration of defensin, nonenzyme cationic protein consisting of some peptides, to the rats with either activation or depression of anticoagulating system produced a moderate decrease of anticoagulating and fibrinolytic properties of the blood and did not intensify the depression of anticoagulating system. Neither did defensin affect the response of anticoagulating system when administered prior to injection of tromboplastin. These properties of defensin make it an efficient hemostatic drug.

[Degree of structural homology of lactoferrins from milk and neutrophilic granulocytes].

Some physico-chemical properties of human and pig lactoferrins from milk and neutrophilic granulocytes were compared. It was shown that the lactoferrins from different cell and tissue sources of the same species (humans or pigs) are identical in terms of electrophoretic mobility, molecular weight, iron-binding capacity, absorbance spectra, amino acid and sugar compositions and peptide maps. Human and pig lactoferrins show a high degree of structural homology (approximately 50%), but are immunochemically different.

[Various properties of cathepsin G and elastase from swine peripheral blood neutrophils].

Using gel filtration through Sephadex G-100 and bioaffinity chromatography on contrical-Sepharose, cathepsin G and elastase were isolated from pig peripheral blood neutrophil granules and purified to homogeneity. Both enzymes hydrolyzed the total histone from calf thymus as well as synthetic substrates--tert-butoxy-L-alanine p-nitrophenyl ester (elastase) and benzoyltyrosine ethyl ester (cathepsin G). The use of natural and synthetic protease inhibitors showed that both enzymes were related to the group of serine proteases. The molecular mass of the cathepsin G subunit as determined by SDS polyacrylamide gel electrophoresis is 28-29 kD, that of elastase--30-31 kD. The pH optima for the hydrolysis of proteinaceous and synthetic substrates for cathepsin G and elastase are 8.0-8.5 and 7.0-7.5, respectively. The isoelectric points for elastase and cathepsin G are 9.7-10.0 and greater than 10, respectively; the temperature optima--30-40 degrees C and 50-60 degrees C, respectively. The amino acid composition of the two enzymes from pig granulocytes revealed a high content of arginine and was similar to that of human granulocytes.

[Isolation and physico-chemical properties of lactoferrin from swine neutrophils].

Lactoferrin, a non-heme iron-binding protein was isolated from pig neutrophils. The purification procedure included initial extraction of the protein in the presence of cetyltrimethylammonium bromide followed by chromatography on carboxymethyl-cellulose and Sephadex G-100. The thus obtained protein was found to be homogeneous on polyacrylamide gel (PAAG) electrophoresis at acidic values of pH. PAAG electrophoresis in the presence of sodium dodecyl sulfate revealed a single component with a molecular weight of 75 000-80 000. The resulting protein is capable of binding two atoms of iron molecule. The absorbance spectra for the pig neutrophil lactoferrin are identical to those for cow milk lactoferrin in the visible region and have a maximum at 465 nm. The amino acid composition of pig lactoferrin was determined. Isoelectric focusing of the protein obtained in a PAAG stabilized pH gradient revealed a component with pI of about 6.8. A single precipitin line was observed with rabbit antipig lactoferrin when examined by immunodiffusion. No immunological cross-reactions were observed between pig lactoferrin and bovine lactoferrin.

[Comparison of various physico-chemical properties of pig and cattle myeloperoxidases].

The myeloperoxidases possessing the specific activity of 500 000-700 000 of o-dianisidine units per 1 mg of protein were isolated and purified from the lysosome-like granules of pig and cattle neutrophylic leukocytes. The absorbance spectra for the enzymes in the visible region are identical; their molecular weight is 140 000-160 000. The enzymes are made up of two subunits, each containing a heavy (m. w. 68 000) and a light (m. w. 10 000) polypeptide chains. The pH optimum for myeloperoxidase oxidation of o-dianisidine lies around 5.8-6.2 for both enzymes. The dependence of the reaction rate on H2O2 concentration does not obey the Michaelis--Menten kinetics. The optimal concentrations of the reaction substrates are 0.34 mM for o-dianisidine and 0.075 mM for H2O2. The amino acid composition and the peptide maps for pig and cattle myeloperoxidases are in many ways similar, showing no coincidence of enzymes, however, in terms of antigenic determinants. The similarities of some physico-chemical properties of pig and cattle enzymes and their immunological differences are discussed.

[Morphologic features of the phagocytosis of filamentous forms of bacteria by neutrophilic granulocytes].

A model system has been developed (B. megaterium + granulocyte) imitating the mechanisms of phagocytosis of filamentous forms of bacteria by neutrophilic granulocytes. In the capture of multicellular microbial filaments by one or several granulocytes predominate the mechanisms of cell membrane invagination and formation of exocytic secretory system providing the inhibition of the activity of the phagocytized agent and release of microbicidal factors and enzymes into the extracellular environment. The importance and frequency of occurrence of the observed phenomenon in different pathological forms have not been elucidated.

[Heterogeneity of histone H1 in cells with blocked genome].

Using polyacrylamide gel electrophoresis, the composition of cationic chromosomal proteins of the cells with completely blocked genome (crayfish, triton and frog spermatozoa, bird erythrocytes, rabbit and cow neutrophils) and homologous somatic cells with matrix-active genome was studied. It was shown that the most common feature of the composition of these cationic chromosomal proteins is the presence of several subfractions of histone H1. This is probably due to the necessity of total condensation of chromosomal fibrills.

[Effect of leukocyte cation proteins in vitro on the activity of the pathogen of meningopneumonitis].

A study was made of the influence of cation proteins from the lysosomes and nuclei of rabbit polymorphonuclear leukocytes on the survival of the causative agent of meningopneumonia in vitro. As revealed, the greatest antibiotic activity was possessed by the nuclear histones and the low molecular basic proteins of lysosomes. Biological significance of the detected effects is discussed.

[Bactericidal effect of cationic proteins of cellular origin on staphylococci].

A study was made of the bactericidal action of thymus histones, histone-like substance, from the nuclei of rabbit leukocytes and lysosomal cation proteins from the same leukocyte on different staphylococcus cultures. The majority of the strains of coagulasoneagative staphylococci perished under the effect of the preparations tested; cultures forming this enzyme retained their viability, irrespective of their virulence for mice, their capacity to the lecithinase formation and to mannite fermentation. The preparations studied possessed a different bactericidal activity. It is supposed that there existed an correlation between the sensitivity of staphylococcus strains to the antimicrobial action of the leukocytic cation proteins and the index of phagocytosis completion of hese microbes.
