[Role of leukocyte disintegration products in increasing macrophage resistance to the causative agent of orinthosis].

The dymanics of formation of macrophagal granulomas in the liver of albino mice was studied after intravenous infection with the causative agent of ornithosis. In formation of granulomas macrophages underwent the stage of reutilization of the leukocyte disintergration and became resistant to the causative agent of ornithosis. Together with the leukocyte remants histones and cation proteins could penetrate into the macrophages cytoplasm; these displayed a high antironithosis activity in vitro. An increase of macrophage resistance to the causative agent after reutilization of the products of leukocyte disintegration apparently sreved as one of the mechanisms of resistance formation in the foci of inflammation in ornithosis.

[In vitro effect of leukocytic and thymic histones and their fractions on the activity of the causative agent of meningopneumonia].

It was found in studying the antimeningococcus activity of the leukocytic and thymus histones and their fractions that both histones were capable of neutralizing in vitro the activity of the causative agent of meningopneumonia (MP). The neutralization effect was chiefly associated with the F3 fraction rich in arginine and depended on the duration of the histone fraction contact with the MP causative agent, the weight concentration of the histone and the pH of the incubation medium.

[Myeloperoxidase in the human placenta at preterm labor].

In the present study the level of myeloperoxidase in the human placenta at premature was investigated. Mycloperoxidase content did not depend on its localization in placenta and decreased at premature labour. It is suggested that the decrease in the myeloperoxidase content in placenta results in the weakening of antimicrobial barrier in the system mother-placenta-fetus and plays an important role in pathogenesis of premature delivery at later term.

[Interaction of heparin with defensin, a nonenzymatic cationic protein from neutrophils].

Heparin was shown to form complexes with cation non-enzymatic protein defensin from neutrophils. The complex obtained in vitro did not affect the anticoagulation and fibrinolytic properties of blood, whereas defensin decreased the content of heparin, rates of non-enzymatic fibrinolysis and total fibrinolytic activity while heparin stimulated the anticoagulation activity of blood plasma. Interaction of heparin with defensin appears to be involved in protective mechanism responsible for neutralization of both the heparin anticoagulation activity and the defensin properties related to inhibition of nonenzymatic fibrinolysis.

[Cationic antimicrobial peptides as molecular immunity factors: multi-functionality].

Cationic antimicrobial peptides (AMP) of mammals (defensins, cathelicidins, protegrins and many others) are regarded as important components of congenital immunity. AMP are multifunctional molecules, capable of killing microorganisms directly by acting as endogenic, natural antibiotics ("immediate immunity"); in addition, they may take part in congenital and adaptive immune reactions (immunoregulation) and function as signal molecules, involved into tissue reparation, inflammation (including sepsis), blood coagulation and other important processes in the body. The molecular mechanisms of the direct antimicrobial action of AMP are considered. In addition to antimicrobial and immunoregulating action, AMP have influence on immunoneuroendocrine interactions, taking part in the pathogenesis of stress reactions (corticostatic action), as well as play the role of regulatory peptides of adaptogenic action. The many-sided character of the action of AMP opens prospects to the creation of new medicinal remedies on their basis. Such requirements are met by the Russian preparation "Superlymph" (a complex of natural cytokines), containing protegrin-like AMP.

[Lysosomal cationic proteins in neutrophilic granulocytes during phagocytosis and inflammation].

Literature and experimental data are reviewed on functional properties of cation proteins in neutrophil granulocytes under conditions of phagocytosis and inflammation. These proteins are not only effective antimicrobial agents but they regulate a lot of protective-adaptational reactions responsible for development of human and animal unspecific resistance.

[Physicochemical and kinetic characteristics of myeloperoxidase of cow neutrophilic leucocytes].

Myeloperoxidase (MPO) isolated from granular fraction of cow peritoneal neutrophils had the RZ value equalled to 0.79. The absorption spectrum of MPO possesses characteristic bands at 430, 500, 570, 625, 690 nm. The molecular weight and sedimentation constant of the enzyme are 135.000-140.000 Daltons and 7.9S, respectively. The data obtained demonstrate the high thermostable property of MPO. Investigations of the pH optimum and effects of varying H2O2, o-dianizidine and MPO concentrations are described.

[Cationic proteins from neutrophils as inhibitors of nonenzymatic fibrinolytic and anticoagulant activity of blood plasma].

Two cation proteins free of enzymatic activity, containing one and four peptides (CP-1 and CP-4, respectively), were studied. The cation protein consisting of four peptides proved to be the most effective inhibitor of the nonenzymatic fibrinolytic activity of heparin-containing complexes. Both these cation proteins inhibited similarly the anticoagulation activity of blood plasma. The drugs, like cation proteins, appear to be potentially important for clinical practice under conditions of excessive hemorrhage caused by elevated level in circulation of complexes containing heparin and proteins.

[Immunoradiometric analysis of rat myeloperoxidase].

The immunoradiometric assay of rat myeloperoxidase (MPO) was developed based on the usage of the CNBr-activated sepharose as a solid phase for antiMPO and of [125I] IgG (the affinity preparation of antiMPO) as a label. The analysis lasted for 8 h and standard curve limits were 0.5-500 ng per tube. Intra and inter assay coefficients of variation did not exceed 3% and 6.5%, respectively. It was shown by the above method that the intensive swimming of male rats led to an increase in the MPO concentration in blood plasma and to its decrease in neutrophils.

[Complex-formation between myeloperoxidase and fibronectin and its modulation by fibronectin ligands].

Human blood plasma fibronectin immobilised on agarose in physiological interacts with soluble myeloperoxidase (Kd = 2.43 mM). Interaction of myeloperoxidase with fibronectin adsorbed on immobilised gelatin a, natural fibronectin ligand, resulted in formation more stable complex (Kd = 0.94 mM). The presence of thermoaggregated (but not native) IgG in the liquid phase increased a stability of the complex (0.06 mM). It is suggested that myeloperoxidase could represent a component of complex super molecular structure, real immune complex.
